Recapitulation of the organ-specific microenvironment, which stimulates local angiogenesis, is indispensable for successful regeneration of damaged tissues. This report demonstrates a novel method to implant fibrin gels on the lung surface of living mouse in order to explore how the lung-specific microenvironment modulates angiogenesis and alveolar regeneration in adult mouse.
Recente significante vooruitgang in het onderzoek naar stamcellen en bio-ingenieur technieken hebben grote vooruitgang geboekt in het gebruik van biomaterialen om zich te vernieuwen en te herstellen schade in eenvoudige weefsels in de orthopedische en parodontale velden. Pogingen om de structuren en functies van meer complexe driedimensionale (3D) organen zoals de longen regenereren weinig succesvol geweest omdat de biologische processen van orgaanregeneratie zijn niet goed onderzocht. Het wordt steeds duidelijker dat de angiogenese, de vorming van nieuwe bloedvaten, speelt een belangrijke rol bij orgel regeneratie. Nieuw gevormde vasculatures leveren niet alleen zuurstof, nutriënten en verschillende celcomponenten die vereist zijn voor orgaanregeneratie maar ook leerzaam signalen aan de regenererende lokale weefsels. Daarom, om de longen bij een volwassene succes regenereren, moet de long-specifieke micro-omgevingen waarin angiogenese aandrijvingen herstel van lokaal longweefsel recapituleren. Hoewel conventional in vivo angiogenese assays, zoals subcutane implantatie van extracellulaire matrix (ECM) -rijk hydrogels (bijvoorbeeld, fibrine en collageen gels of Matrigel – ECM-eiwit mengsel afgescheiden door Engelbreth-Holm-Swarm muis sarcoom cellen), worden op grote schaal gebruikt om het verkennen algemene mechanismen van angiogenese, long-specifieke angiogenese niet goed gekarakteriseerd omdat werkwijzen voor orthotope implantatie van biomaterialen in de longen niet goed vastgesteld. Het doel van dit protocol is om een unieke methode te introduceren in fibrine gel implantaat op de long oppervlak van levende volwassen muis, waardoor de succesvolle recapitulatie van gastheer-long afgeleid angiogenese in de gel. Deze aanpak stelt onderzoekers in staat om de mechanismen die de long-specifieke micro-omgeving controleert angiogenese en alveolaire regeneratie in zowel normale en pathologische omstandigheden te onderzoeken. Sinds geïmplanteerde biomaterialen vrijkomen en leveren fysieke en chemische signalen naar aangrenzende lung weefsels, kan implantatie van deze biomaterialen op zieke long potentieel normaliseren aangrenzende zieke weefsels, waardoor onderzoekers nieuwe therapeutische benaderingen van verschillende soorten longziekten ontwikkelen.
De algemene doelstelling van dit protocol is een methode te introduceren aan fibrine gel implantaat op de long oppervlak van volwassen muizen, die het mogelijk maakt de onderzoekers om de moleculaire mechanismen van long vasculaire en alveolaire ontwikkeling karakteriseren, en om deze kennis benutten om biomimetic materialen die te ontwikkelen van recapituleren fysiologische long vasculaire en alveolaire vorming van diverse longziekten behandelen.
Meer dan 35 miljoen Amerikanen lijden aan chronische longziekten waaronder chronische obstructieve longziekte en pulmonaire fibrose. Deze patiënten hebben een langdurige chronische respiratoire symptomen zoals kortademigheid, pijn op de borst, zeurende hoest, en vermoeidheid, die hun dagelijks leven 1-3 aanzienlijk beperken. Ondanks een grote hoeveelheid inspanning om effectieve therapieën voor deze longziekten te ontwikkelen, momenteel is er geen genezing; Daarom, de kwaliteit van leven voor deze patiënten is slecht en de economische en menselijke kosten zijn high 4-7. Momenteel longtransplantatie is de enige manier om patiënten met eindstadium van chronische longziekten op te slaan. Vanwege het gebrek aan transplantatie donoren, hoge kosten, ernstige complicaties en lage overlevingspercentage 8-11, transplantatie geen optimale benadering. Recente snelle vooruitgang in de tissue engineering technieken heeft onderzoekers nodig om implanteerbare long bio-ingenieur door herbevolken gedecellulariseerde hele long met diverse soorten stamcellen of geïnduceerde pluripotente stamcellen (iPS) cellen 12,13. Echter, deze bioengineered longen functioneel in gastheerdieren slechts enkele uren na implantatie 12,14,15. Gebruik makend biomaterialen de complexe structuren en functies van de longen regenereren is ook tamelijk succesvol. Dit kan zijn omdat de belangrijkste biologische processen die volwassen longen regeneratie regeren zijn niet goed onderzocht. In de long, vorming van het vasculaire systeem is een van de vroegste en belangrijkste gebeurtenissen during ontwikkeling en regeneratie 16-21. Nieuw gevormde vasculatures in de long leveren niet alleen zuurstof, voedingsstoffen en diverse mobiele componenten die nodig zijn voor orgel vorming, maar ook leerzaam regulerende signalen naar omliggende cellen 22-25. Zo angiogenese speelt een belangrijke rol in de regeneratieve alveolarization bij volwassen longen 24,26,27. Bovendien gedereguleerde angiogenese bijdraagt aan chronische longziekten zoals chronische obstructieve longziekte (COPD) 28, bronchopulmonaire dysplasie (BPD) 21-23 en longfibrose 29. Dus om efficiënter uit manipuleren longen of behandelen van chronische longziekten ontwikkelen is het noodzakelijk om het werkingsmechanisme van long-specifieke angiogenese begrijpen.
Elk orgaan geeft unieke mechanische en chemische eigenschappen, die kan verschillen tussen fysiologische en pathologische omstandigheden 30-33. Deze orgaanspecifieke microenvironmenten reguleren endotheelcellen gedrag en orkestreren vasculaire netwerkvorming in een orgaan-specifieke manier 24,34-36. Dus om efficiënter uit long regeneratie ontwikkelen, het mechanisme dat long-specifieke angiogenese te worden begrepen. Terwijl conventionele in vivo angiogenese assays zoals subcutane implantatie hydrogel zijn uitgebreid gebruikt voor angiogenese onderzoek 37-39, hebben deze methoden niet herhalen orgaanspecifieke angiogenese. Onlangs is een nieuwe werkwijze te Matrigel implanteren in een elastische schimmel op de muizenlong ontwikkeld en getoond bloedvaten en long epitheelcellen in de gels 22 succesvol werven. Deze unieke benadering kunnen de onderzoekers het mechanisme van long-specifieke angiogenese alsmede interactie tussen bloedvaten en niet-vasculaire longcellen verkennen in fysiologische en pathologische omstandigheden. Sinds 1) Matrigel is niet geschikt voor klinische toepassing; 2) elastic mal gebruikt om de gel kunnen beïnvloeden interacties tussen hydrogels en gastheer longweefsel en 3) de elastische schimmel op de longen veroorzaakt potentieel verminderde longfunctie en pijn tijdens de ademhaling gegoten als een klinisch relevante benadering een 3D fibrine matrix die angiogene factoren (vasculaire endotheliale groeifactor (VEGF) / basische fibroblast groeifactor (bFGF)) is geïmplanteerd op de muizenlong zonder gieten de elastische matrijs, en met succes geïncorporeerd gastheer-long afgeleide angiogenese. Fibrinegel, polymeer fibrillen gegenereerd uit trombinegekliefde fibrinogeen, bekend te vangen diverse angiogene factoren zoals bFGF en VEGF angiogenese versnellen vivo 40,41. Door zijn regeneratieve vermogen en biologisch afbreekbaar 42 wordt fibrine gel veel gebruikt op het gebied van weefselmanipulatie.
Dit artikel introduceert een nieuwe en unieke benadering van fibrine gel implantaat op de long oppervlak van levende adult muis en toont aan dat gastheer-long afgeleide angiogenese wordt geïncorporeerd in de gel in vivo.
