Pathology of the aorta can lead to severe morbidity and mortality, therefore research of disease progression and potential therapies is warranted. Here, we present a protocol to isolate and excise the murine aorta to aid researchers in their investigation of cardiovascular disease.
Cardiovascular disease is a broad term describing disease of the heart and/or blood vessels. The main blood vessel supplying the body with oxygenated blood is the aorta. The aorta may become affected in diseases such as atherosclerosis and aneurysm. Researchers investigating these diseases would benefit from direct observation of the aorta to characterize disease progression as well as to evaluate efficacy of potential therapeutics. The goal of this protocol is to describe proper isolation and excision of the aorta to aid investigators researching cardiovascular disease. Isolation and excision of the aorta allows investigators to look at gross morphometric changes as wells as allowing them to preserve and stain the tissue to look at histologic changes if desired. The aorta may be used for molecular studies to evaluate protein and gene expression to discover targets of interest and mechanisms of action. This technique is superior to imaging modalities as they have inherent limitations in technology and cost. Additionally, primary isolated cells from a freshly isolated and excised aorta can allowing researchers to perform further in situ and in vitro assays. The isolation and excision of the aorta has the limitation of having to sacrifice the animal however, in this case the benefits outweigh the harm as it is the most versatile technique in the study of aortic disease.
De aorta speelt een centrale rol in cardiovasculaire functie leveren het lichaam met zuurstofrijk bloed en vergelijkbaar met andere delen van het lichaam, is vatbaar voor ziekten. Voorkomende aorta ziekten omvatten atherosclerose en aneurysma, waarin de resultaten van genetische en omgevingsfactoren zoals voeding, roken en sedentaire levensstijl 1 zijn. Atherosclerose is de afzetting van calcium en op lipide gebaseerde plaque de aortawand typisch in patiënten met chronische hyperlipidemie 2. Aneurysma's worden gekenmerkt door afbraak van de structurele componenten van de aorta en verdunning van de vaatwand, gevolgd door een vergrote diameter die uiteindelijk kunnen leiden tot scheuren 3.
Diermodellen zijn belangrijke instrumenten gebruikt om het mechanisme van ziekte en werkzaamheid van mogelijke behandelingen bestuderen. Vaak gebruikte diermodellen in cardiovasculair onderzoek, specifiek onderzoek zijn vasculaire en metabole stoornissenonder andere genetisch gemodificeerde muizen aan lipide niveaus zoals apolipoproteïne E-gen knock-out en low density lipoprotein receptor knockout muizen 4 veranderen. De meeste methoden mechanismen van ziekte en werkzaamheid van de therapie te evalueren omvatten de isolatie en excisie van de aorta.
Als de aorta is gelokaliseerd en geïsoleerd, kunnen morfometrische analyse bepaald, zoals aanwezigheid van aneurysma, typisch gedefinieerd als een stijging van meer dan 50% in diameter 5. Na noodzakelijk situ analyse is voltooid, kan de aorta worden uitgesneden voor verdere analyse. Een uitgesneden aorta kan worden diepgevroren moleculaire studies zoals eiwitten en / of genexpressie testen of die bevestigd met 4% paraformaldehyde en later ingebed, coupes en gekleurd voor histologische analyse. Histologische analyse van de aorta kan aantonen gemeenschappelijke kenmerken van de aorta zoals structurele afbraak, plaquevorming en infiltratie van leukocyten <sup> 6,7. Bovendien kan een uitgesneden aorta worden gebruikt om primaire cellijnen waaronder endotheel- en gladde spiercellen die vervolgens kan worden gebruikt voor een verscheidenheid van in vitro studies 7 isoleren.
Momenteel zijn er geen andere methoden om deze grondige karakterisering van de aorta verstrekken en voorzien bedrijven met hulpmiddelen om verdere studie hart- en vaatziekten. Beeldvormende modaliteiten zoals magnetische resonantie beeldvorming, computertomografie en echografie zijn de dichtstbijzijnde methoden om de aorta morfologisch, maar dit is moeilijk in kleine dieren en het verkrijgen van een apparaat met voldoende techniek is duur 8. Onsterfelijk gemaakte cellijnen zijn verkrijgbaar mogelijke mechanismen van ziekte en werkzaamheid van therapieën, maar de artificiële aard van deze cellen beperkend bestuderen van de effecten op de cel levenscyclus en apoptose 9 onderzoeken.
De algemene doelstelling vandit manuscript is de steriele isolatie en excisie van het muizen aorta bij het onderzoek van hart- en vaatziekten tonen.
Aortic disease can lead to significant systemic pathology and possibly death in severe cases. Treatment options are currently limited therefore continued research in the field is necessary10. Murine models are ideal to further study disease progression and potentially therapeutic options4. Therefore, the successful completion of the method described in this manuscript will provide investigators with the tools to measure disease progression and drug efficacy.
