Patch-clamp recordings and simultaneous intracellular biocytin filling of synaptically coupled neurons in acute brain slices allow a correlated analysis of their structural and functional properties. The aim of this protocol is to describe the essential technical steps of electrophysiological recording from neuronal microcircuits and their subsequent morphological analysis.
Kombinasjonen av patch clamp opptak fra to (eller flere) postsynaptisk kombinert nevroner (sammenkoblede innspillinger) i akutte hjerne slice preparater med samtidig intracellulær biocytin fylling gir en korrelert analyse av deres strukturelle og funksjonelle egenskaper. Med denne metoden er det mulig å identifisere og karakterisere både pre- og postsynaptiske nevroner av deres morfologi og elektrofysiologisk reaksjonsmønster. Sammenkoblede innspillinger tillate studere tilkoblingsmønsteret mellom disse nevronene samt egenskapene til både kjemisk og elektrisk synaptisk overføring. Her gir vi en steg-for-steg beskrivelse av prosedyrene som kreves for å få pålitelige sammenkoblede innspillinger sammen med en optimal utvinning av nervecellen morfologi. Vi vil beskrive hvordan par av nevroner som er tilkoblet via kjemiske synapser eller gap veikryss er identifisert i hjernen slice forberedelser. Vi vil skissere hvordan nerveceller blir rekonstruert for å få deres 3D morfologi dendritic og aksonal domene og hvordan synaptiske kontakter er identifisert og lokalisert. Vi vil også diskutere de begrensningene og begrensninger av den sammenkoblede opptaksteknikk, spesielt de som er forbundet med dendrittiske og aksonale trunkeringer under utarbeidelsen av hjerneskiver fordi disse sterkt påvirke tilkoblings estimater. Men på grunn av allsidigheten til den sammenkoblede opptak tilnærming vil det fortsatt være et verdifullt verktøy for å karakterisere ulike sider ved synaptisk overføring på identifiserte nevrale kretser i hjernen.
Nevrale kretser mellom to postsynaptisk kombinert nevroner er byggesteinene i store nettverk i hjernen og er de grunnleggende enheter av synaptisk informasjonsbehandling. En forutsetning for karakterisering av slike neuronal mikrokretser er kjent at morfologien og funksjonelle egenskaper av både pre- og postsynaptiske nevroner partner, typen av synaptisk forbindelse (r), og dens struktur og funksjonelle mekanisme. Men i mange studier av synaptiske forbindelser i det minste en av nevroner i en mikrokrets er ikke godt karakterisert. Dette skyldes den relativt uspesifikke stimuleringsregimer som ofte brukes i studier av synaptisk tilkobling. Derfor er de strukturelle og funksjonelle egenskaper av den presynaptiske neuron heller ikke identifisert i det hele tatt eller bare i ganske liten grad (dvs. ekspresjon av markørproteiner etc.). Sammenkoblede opptak i kombinasjon med intracellulær farging av markører such som biocytin, neurobiotin eller fluorescerende fargestoffer er bedre egnet for å studere små nevrale kretser. Denne teknikken gjør det mulig å undersøke mange strukturelle og funksjonelle parametre for en morfologisk identifisert synaptisk forbindelse på samme tid.
Såkalte 'enhetlige' monosynaptisk forbindelser mellom to nerveceller har blitt undersøkt i begge kortikale og subkortikale hjerneregioner 1-10 bruker akutte slice forberedelser. Initialt ble laminert mikroelektroder som brukes i disse forsøkene; senere ble patch clamp monitorerings anvendes for å oppnå opptak av synaptiske signaler med et lavere støynivå og et forbedret tidsmessig oppløsning.
Et betydelig teknisk fremskritt var bruken av infrarød differensialinterferenskontrast (IR-DIC) optikk 11 til 14, en mikroskopisk metode som i betydelig grad forbedret synlighet og identifisering av nevronene i hjernen skive slik at det ble mulig to innhente opptak fra visuelt identifisert synaptiske forbindelser 15-17. Generelt er sammenkoblede innspillinger gjort i akutte slice forberedelser; bare svært få publikasjoner er tilgjengelige rapporterings opptak fra postsynaptisk koblet nevroner in vivo 18-20.
Den viktigste fordelen med sammenkoblede opptak er det faktum at en funksjonell karakterisering kan kombineres med en morfologisk analyse ved både lys og elektronmikroskopnivå (se f.eks., 7,16,21). Etter histokjemisk behandling, er dendrittiske og aksonal morfologi av postsynaptisk koblet neuron par spores. Deretter er det mulig å kvantifisere morfologiske egenskaper som lengde, romlig tetthet, orientering, forgrening mønster etc. Disse parametrene kan deretter gi grunnlag for en objektiv klassifisering av en spesifikk synaptisk tilkobling. Videre, i motsetning til de fleste andre teknikker som brukes for å studere neuronal connectiteten, sammen innspillinger også tillate identifisering av synaptiske kontakter for enhetlige synaptiske forbindelser. Dette kan gjøres direkte med en kombinasjon av lys og elektronmikroskopi 16,21-27 eller bruke kalsium bildebehandling 28,29 av dendrittutløperne. Men med den sistnevnte tilnærmingen bare eksitatoriske men ikke hemmende tilkoblinger kan studeres som det krever kalsium tilstrømningen via de postsynaptiske reseptor kanaler.
I tillegg til en detaljert analyse av synaptisk transmisjon i en definert neuronal mikrokretssammenkoblet opptak også tillater studiet av synaptisk plastisitet regler 30,31 eller – i kombinasjon med agonist / antagonist søknad – modulering av synaptisk transmisjon av neurotransmittere slik som acetylkolin 32 og adenosin 33.
Sammenkoblede opptak fra postsynaptisk kombinert eksitatoriske og / eller hemmende nerveceller er en svært allsidig tilnærming til studiet av nevrale kretser. Ikke bare gjør denne tilnærmingen tillater en å anslå synaptic tilkobling mellom nevroner typer, men også tillater å bestemme de funksjonelle egenskapene til tilkoblingen og morfologi av pre- og postsynaptiske nevroner.
