Patch-clamp recordings and simultaneous intracellular biocytin filling of synaptically coupled neurons in acute brain slices allow a correlated analysis of their structural and functional properties. The aim of this protocol is to describe the essential technical steps of electrophysiological recording from neuronal microcircuits and their subsequent morphological analysis.
Kombinationen af patch clamp optagelser fra to (eller flere) synaptisk koblede neuroner (parrede optagelser) i akutte præparater hjernen skive med samtidig intracellulær biocytin fyldning giver en korreleret analyse af deres strukturelle og funktionelle egenskaber. Med denne metode er det muligt at identificere og karakterisere både præ- og postsynaptiske neuroner ved deres morfologi og elektrofysiologiske respons mønster. Forbundne optagelser tillader studere tilslutningsmuligheder mønstre mellem disse neuroner samt egenskaber både kemisk og elektrisk synaptisk transmission. Her giver vi et trin-for-trin beskrivelse af de procedurer, der er nødvendige for at opnå pålidelige parrede optagelser sammen med en optimal genvinding af neuron morfologi. Vi vil beskrive, hvordan par af neuroner forbundet via kemiske synapser eller gap junctions er identificeret i hjernen skive præparater. Vi vil skitsere, hvordan neuroner rekonstrueres for at få deres 3D-morfologi dendritic og aksonal domæne og hvordan synaptisk kontakter identificeres og lokaliseres. Vi vil også drøfte de forbehold og begrænsninger den parrede optagelse teknik, især dem der er forbundet med dendritiske og axonale trunkeringer under udarbejdelsen af hjerneskiver fordi disse stærkt påvirke tilslutningsmuligheder skøn. Men på grund af alsidigheden af den parrede optagelse tilgang vil det fortsat være et værdifuldt værktøj til at karakterisere forskellige aspekter af synaptisk transmission på identificerede neuronale mikrokredsløb i hjernen.
Neuronale mikrokredsløb mellem to synaptisk sammenkoblede neuroner er byggestenene i store netværk i hjernen og er de grundlæggende enheder af synaptisk informationsbehandling. En forudsætning for karakterisering af sådanne neuronale mikrokredsløb er at kende morfologi og funktionelle egenskaber af både for- og postsynaptiske partner neuroner, typen af den synaptiske forbindelse (r) og dets struktur og funktionelle mekanisme. Men i mange studier af synaptiske forbindelser mindst en af de neuroner i et mikrokredsløb er ikke godt karakteriseret. Dette skyldes de relativt uspecifikke stimulationsregimer ofte bruges i studier af synaptisk forbindelse. Derfor de strukturelle og funktionelle egenskaber af den præsynaptiske neuron enten ikke identificeres på alle eller kun en forholdsvis lille udstrækning (dvs. ekspressionen af markørproteiner etc.). Forbundne optagelser i kombination med intracellulær farvning af markører such som biocytin, neurobiotin eller fluorescerende farvestoffer er bedre egnet til at studere små neuronale mikrokredsløb. Denne teknik gør det muligt at undersøge mange strukturelle og funktionelle parametre for en morfologisk identificerede synaptisk forbindelse på samme tid.
De såkaldte "enhedsstat" monosynaptiske forbindelser mellem to neuroner er blevet undersøgt i både subkortikale hjerneområder 1-10 ved hjælp akutte præparater skive. Oprindeligt blev skarpe mikroelektroder anvendt i disse eksperimenter; senere blev patch clamp optagelse ansat for at opnå optagelser af synaptiske signaler med et lavere støjniveau og en forbedret tidsopløsning.
En væsentlig teknisk fremskridt var brugen af infrarøde differential interferens kontrast (IR-DIC) optik 11-14, en mikroskopisk teknik, væsentligt forbedret synligheden og identifikation af neuroner i hjernen skive, så det blev muligt to få optagelser fra visuelt identificeret synaptiske forbindelser 15-17. I almindelighed er parrede optagelser udført i akutte præparater slice; kun meget få publikationer er tilgængelige rapportering optagelser fra synaptisk forbundne neuroner in vivo 18-20.
Den vigtigste fordel ved parrede optagelser er, at en funktionel karakterisering kan kombineres med en morfologisk analyse på både lys- og elektronmikroskopiske niveau (se f.eks., 7,16,21). Efter histokemisk behandling, er den dendritiske og axonal morfologi synaptisk forbundet neuron par spores. Efterfølgende er det muligt at kvantificere morfologiske træk såsom længde, fysisk tæthed, orientering, forgrening mønster etc. Disse parametre kan så danne grundlag for en objektiv klassificering af en specifik synaptisk forbindelse. Derudover, i modsætning til de fleste andre teknikker, der anvendes til at studere neuronale connectivity, parret optagelser tillader også identifikation af synaptiske kontakter om ensartet synaptiske forbindelser. Dette kan gøres direkte ved hjælp af en kombination af lys og elektronmikroskopi 16,21-27 eller ved hjælp af calcium billeddannelse 28,29 af dendritiske Torner. Men med den sidstnævnte fremgangsmåde kun excitatoriske men ikke hæmmende forbindelser kan studeres, da det kræver calciumindstrømning via postsynaptiske receptor-kanaler.
Ud over en detaljeret analyse af synaptisk transmission ved en defineret neuronale mikrokredsløb parret optagelser også give studiet af synaptisk plasticitet regler 30,31 eller – i kombination med agonist / antagonist ansøgning – modulering af synaptisk transmission af neurotransmittere, såsom acetylcholin 32 og adenosin 33.
Forbundne optagelser fra synaptisk koblet excitatoriske og / eller hæmmende neuroner er en meget alsidig fremgangsmåde til undersøgelse af neuronale mikrokredsløb. Ikke alene denne fremgangsmåde, at man kan estimere synaptisk forbindelse mellem neuron typer, men også gør det muligt at bestemme de funktionelle egenskaber af forbindelsen og morfologi præ- og postsynaptiske neuroner.
