The molecular mechanisms of the decondensation of highly compacted mitotic chromatin are ill-defined. We present a cell-free assay based on mitotic chromatin clusters isolated from HeLa cells and Xenopus laevis egg extract that faithfully reconstitutes the decondensation process in vitro.
During the vertebrate cell cycle chromatin undergoes extensive structural and functional changes. Upon mitotic entry, it massively condenses into rod shaped chromosomes which are moved individually by the mitotic spindle apparatus. Mitotic chromatin condensation yields chromosomes compacted fifty-fold denser as in interphase. During exit from mitosis, chromosomes have to re-establish their functional interphase state, which is enclosed by a nuclear envelope and is competent for replication and transcription. The decondensation process is morphologically well described, but in molecular terms poorly understood: We lack knowledge about the underlying molecular events and to a large extent the factors involved as well as their regulation. We describe here a cell-free system that faithfully recapitulates chromatin decondensation in vitro, based on mitotic chromatin clusters purified from synchronized HeLa cells and X. laevis egg extract. Our cell-free system provides an important tool for further molecular characterization of chromatin decondensation and its co-ordination with processes simultaneously occurring during mitotic exit such as nuclear envelope and pore complex re-assembly.
Xenopus laevis ei extract is een krachtige en algemeen toegepaste gereedschap studeren complexe cellulaire gebeurtenissen in de eenvoud van een celvrije assay. Sinds hun eerste beschrijving van Lohka & Masui 1 zij zijn uitgebreid gebruikt om mitotische processen zoals chromatinecondensatie 2, assemblage van de spoel 3, afbraak nucleaire envelop 4, maar ook nucleocytoplasmatisch vervoer 5 of DNA-replicatie 6 bestuderen. De gebeurtenissen die plaatsvinden aan het eind van mitose, noodzakelijk voor hervorming van de interfase nucleus zoals envelope reformatie nucleaire en nucleaire porie complex montage veel minder begrepen vergelijking met de vroege mitotische gebeurtenissen, maar kan eveneens worden bestudeerd door Xenopus ei extract 7. We hebben onlangs een assay gebaseerd op Xenopus ei extract chromatine decondensatie studie eind mitose 8 vastgesteld, een onder onderzochte proces i wachtts gedetailleerde karakterisering.
In meercelligen wordt zeer gecondenseerde chromatine op mitotische vermelding om getrouw scheiding van het genetisch materiaal uit te voeren. Om de chromatine toegankelijk voor genexpressie en DNA-replicatie tijdens interfase, moet worden verdicht de-eind mitose. In vertebraten, chromatine tot vijftig maal meer samengeperst tijdens mitose opzichte interfase 9, in tegenstelling tot gisten wanneer de mitotische verdichting meestal lager, bijvoorbeeld, twee-voudig S. 10 cerevisiae. Gewervelde chromatine decondensatie is voornamelijk bestudeerd in de context van sperma DNA reorganisatie na ei bevruchting. Een moleculair mechanisme, waarbij nucleoplasmin, een overvloedige eicel eiwit, ruilt sperma-specifieke protaminen naar histonen H2A en H2B opgeslagen in het ei. Deze werkwijze werd opgehelderd middels Xenopus ei extract 11, 12. De uitdrukkingvan nucleoplasmin is beperkt tot eicellen 13 en mitotische chromatine niet van deze sperma-specifieke protaminen bevatten. Daarom chromatine decondensatie eind mitose is nucleoplasmin onafhankelijk 8.
Voor de in vitro decondensatie reactie gebruiken we extract gegenereerd uit geactiveerde X. laevis eieren en chromatine clusters geïsoleerd uit gesynchroniseerde HeLa-cellen. Behandeling van eieren met een calciumionofoor bootst de calcium introductie in de eicel die door sperma ingang tijdens de bevruchting. De calcium golf activeert de celcyclus hervatting en het ei, gearresteerd in de tweede metafase van de meiose, ontwikkelt tot de eerste interfase 14. Daarom ei extracten bereid vorm geactiveerd eieren vertegenwoordigen de mitotische afslag / interphase staat en bevoegd zijn om evenementen die specifiek zijn voor mitotische exit zoals chromatine decondensatie, nucleaire envelop induceren en porie complex reformatie. Voor de isolatie van mitotische chromatin clusters we gebruikten een enigszins aangepaste versie van het protocol gepubliceerd door Gasser & Laemmli 15, waarbij chromosoom clusters worden vrijgemaakt door lysis van HeLa-cellen in mitose gesynchroniseerd en geïsoleerd in polyamine bevattende buffers met gradiënt centrifugeren.
Xenopus laevis ei extracten zijn een zeer nuttig instrument om getrouw weer cellulaire processen in vitro en dit systeem werd met succes gebruikt in de karakterisering van de celcyclus en celdeling gebeurtenissen 2, 3,5,6,17. Door grote winkels nucleaire componenten afgezonderd in het ei tijdens oögenese, ei-extracten zijn een uitstekende bron van cellulaire componenten.
