A quick protocol for proteolytic digestion with an in-house built flow-through tryptic microreactor coupled to an electrospray ionization (ESI) mass spectrometer is presented. The fabrication of the microreactor, the experimental setup and the data acquisition process are described.

In dit werk beschrijven we een eenvoudige en rendabele benadering die in elk laboratorium kan worden toegepast voor het bereiken van snelle enzymatische vertering van een proteïne. Het eiwit (of eiwit mengsel) wordt geadsorbeerd op C18-gebonden omgekeerde fase performance vloeistofchromatografie (HPLC) silicadeeltjes voorgeladen in een capillaire kolom en trypsine in waterige buffer wordt toegediend gedurende de deeltjes gedurende korte tijd. Om on-line MS detectie mogelijk te maken, worden de tryptische peptiden geëlueerd met een oplosmiddel systeem met een verhoogd gehalte aan organisch materiaal direct in het MS ionenbron. Deze aanpak vermijdt het gebruik van dure geïmmobiliseerde enzymdeeltjes en vereist niet geen steun voor voltooiing van het proces. Eiwitvertering en volledige monsteranalyse kan worden uitgevoerd in minder dan 3 min ~ en ~ 30 min, respectievelijk.
De identificatie en karakterisering van gezuiverde eiwitten wordt vaak bereikt door MS technieken. Het eiwit wordt geknipt met een enzym en de peptiden worden verder geanalyseerd door MS via een eenvoudige infusie experimentele opstelling. Proteolytische digestie noodzakelijk voor het genereren van kleine peptidefragmenten die in de nuttige massabereik meeste MS analyzers vallen, en die gemakkelijk worden gefragmenteerd door lage botsenergie fragmentatie te aminozuursequentie informatie te genereren. Voor geïsoleerde eiwitten of eiwitmengsels eenvoudig, er geen verdere behoefte voor chromatografische scheiding van peptiden voorafgaand aan MS detectie. Een mengsel van 25-50 peptiden kunnen gemakkelijk worden geanalyseerd door het inbrengen van het monster met een spuitpomp direct in het MS ionenbron.
De massaspectrometer kan de analyse uit te voeren en bevestig de sequentie van een eiwit binnen een kort tijdsbestek. Met moderne data-acquisitie methoden, kan dit proces worden bereikt wedurende enkele minuten of zelfs seconden. De beperkende factor bij het voltooien van het hele proces op een korte tijdschaal is de proteolytische vertering stap. Dit wordt typisch uitgevoerd gedurende enkele uren (of O / N) in oplossing bij 37 ° C met behulp van substraat: enzym verhoudingen van (50-100): 1. Ondanks de vele voordelen van geïmmobiliseerde enzym reactoren, de kosten voor commerciële cartridges is hoog, de beschikbaarheid van microfluïdische inrichtingen voor routinegebruik beperkt en het gebruik van microgolf of PCT technieken leidt tot behoefte aan extra apparatuur. Het doel van dit werk was om een methode te ontwikkelen die circumvents deze nadelen en die gemakkelijk kunnen worden geïmplementeerd in elk laboratorium onderzoekers machtigen met een eenvoudige en doeltreffende benadering voor het uitvoeren van enzymatische splitsing van eiwitten ter voorbereiding MS analyse binnen minuten. De in dit werk beschreven microreactor biedt een eenvoudig te implementeren experimentele opstelling voor het uitvoeren van enzymatische vertering van eiwitten MS analyse en identificatie in minder dan 30 minuten mogelijk.
