Accurate assessment of anti-inflammatory effects is of utmost importance for the evaluation of potential new drugs for the treatment of inflammatory bowel disease. Digital holographic microscopy provides assessment of inflammation in murine and human colonic tissue samples as well as automated multimodal evaluation of epithelial wound healing in vitro.
Forekomsten av inflammatorisk tarmsykdom, det vil si, Crohns sykdom og ulcerøs kolitt, har økt betydelig det siste tiåret. Etiologien av IBD er fortsatt ukjent og nåværende terapeutiske strategier er basert på den uspesifikke undertrykkelse av immunsystemet. Utviklingen av behandlinger som spesifikt retter tarmbetennelse og epitel sårtilheling kunne gi en betydelig bedre styring av IBD, men dette krever nøyaktig påvisning av betennelsesforandringer. Foreløpig er potensielle legemiddelkandidater vanligvis evaluert med dyremodeller in vivo eller med cellekulturbaserte teknikker i vitro. Histologisk undersøkelse krever vanligvis cellene eller vev av interesse å bli farget, noe som kan endre prøven egenskaper og dessuten kan tolkningen av resultatene varierer fra utprøver ekspertise. Digital holografisk mikroskopi (DHM), basert på påvisning av optiske veilengden forsinkelse, tillaterflekkfrie kvantitativ fase kontrast bildebehandling. Dette gjør at resultatene skal være direkte korrelert med absolutte biofysiske parametre. Vi viser hvordan måling av endringer i vev tetthet med DHM, basert på brytningsindeks måling, kan kvantifisere inflammatoriske forandringer, uten flekker, i forskjellige lag av colonic vevsprøver fra mus og mennesker med kolitt. I tillegg viser vi kontinuerlig multimodale label-fri overvåking av epitel sårtilheling in vitro, mulig å bruke DHM gjennom den enkle automatisert bestemmelse av de sårede området og samtidig bestemmelse av morfologiske parametere som tørr masse og tykkelse av trekkende celler. I konklusjonen, representerer DHM en verdifull, roman og kvantitative verktøy for vurdering av tarmbetennelse med absolutte verdier for parametrene mulige, forenklet kvantifisering av epitel sårtilheling in vitro, og derfor har stort potensial for translasjonsforskning diagnostisk use.
Inflammatorisk tarmsykdom (IBD), det vil si, ulcerøs kolitt (UC) og Crohns sykdom (CD) er idiopatiske inflammatoriske sykdommer i mage-tarmkanalen 1. Forskning på den underliggende patofysiologien av IBD og vurdering av nye medikamenter eller nye diagnostiske metoder er særlig av betydning. I både grunnforskning og den kliniske behandlingen av IBD pasienter, har tarmslimhinnen blitt et fokus for oppmerksomhet 2,3. Mukosa representerer en anatomisk grense, ved hvilken interaksjonen mellom kommensale bakterier, epitelceller og forskjellige cellulære komponenter av tarmimmunsystemet organisere gut homeostase 4,5. Men i IBD pasienter, fører ukontrollert og vedvarende tarmbetennelse på slimhinneskader, påviselig som sår eller stenose, som til slutt kan ende i sammenbrudd av epitelial barrierefunksjon, noe som i seg selv forverrer lokal betennelse 6.
<pclass = "jove_content"> Epitelial sårheling er derfor avgjørende for epitelial regenerering følgende betennelse, men er også en sentral forutsetning for helbredelse av gastrointestinale sår eller anastomotic lekkasje etter gastrointestinal kirurgi 7. Epitelial sårheling kan simuleres i in vitro assays sårheling og i murine modeller av tarmbetennelse 8,9. Både in vitro og in vivo fremgangsmåter har ulemper, som begrenser nøyaktigheten av eksperimentell bedømmelse. In vitro-undersøkelser, som klassisk skrape analyser krever langvarige flekker prosedyrer eller transfeksjon med fluorescerende chromophores. De er ofte begrenset av deres usammenhengende overvåking av celleproliferasjon og migrasjon som ikke kan automatiseres 10. In vivo modeller, for eksempel dekstran natriumsulfat (DSS) -indusert kolitt, mangler ofte robuste read-outs, delvis på grunn av den betydelige variasjonen sett i laboratoriemarkører, making slike markører upassende å evaluere kolitt alvorlighetsgrad 11,12. DHM tillater vurdering av vev tetthet ved å måle optisk veilengde forsinkelse (OPD) som utsiktene roman kreftdiagnose 16-18 og kvantifisering av betennelser relatert vev endringer 19. I tillegg gir DHM overvåking av cellemorfologi dynamikk ved å bestemme celletykkelse, celle dekket flateareal og intracellulært (protein) innhold mengde <sup> 15,20. I in vitro-analyser, DHM muliggjør også analyse av fysiologiske prosesser, f.eks, cellulær vannpermeabiliteten ved å evaluere forandringer i cellevolumet og tykkelse 21,22. Videre kan DHM målinger automatiseres som hindrer etterforsker-forbundet sample bias.
Her viser vi bruken av DHM i en murin modell av intestinal inflammasjon, og gjelder også DHM for analyse av humane vev prøver for kvantitativ overvåkning av sårheling som en etikett fritt in vitro-analysen. Først vurderer vi inflammatoriske forandringer av ulike colonic vegg-lag i colitic mus og seksjoner vev fra mennesker med IBD. Etter beskriver DHM kvantitativ fase bildebehandling prosedyren, gir vi detaljerte instruksjoner for bruk av mikroskop komponenter, utarbeidelse av vevssnitt og også beskrive evalueringen av de oppkjøpte kvantitative fase bilder.
Deretter viser vi at DHM kan være UVIlized for kontinuerlig multimodal overvåking av epitel sårtilheling in vitro, og beskrive analyse av mobilnettet egenskaper som cellelag tykkelse, tørr masse og cellevolum gi innsikt i legemiddelinduserte og fysiologisk celle forandringer.
Vi viser at DHM gir nøyaktig vurdering av histologiske skader i murine kolitt modeller og prøver ex vivo menneskelige colonic vev. Videre har vi vist DHM kan kontinuerlig overvåke epitel sårheling samtidig gi multimodal informasjon om cellulære endringer.
