We describe a simple protocol using only basic lab equipment to generate and purify large quantities of a fusion protein that contains mouse Myelin Oligodendrocyte Glycoprotein. This protein can be used to induce experimental autoimmune encephalomyelitis driven by both T and B cells.
Multiple sclerosis (MS) is a chronic inflammatory disease of the central nervous system (CNS), thought to occur as a result of autoimmune responses targeting myelin. Experimental autoimmune encephalomyelitis (EAE) is the most common animal model of CNS autoimmune disease, and is typically induced via immunization with short peptides representing immunodominant CD4+ T cell epitopes of myelin proteins. However, B cells recognize unprocessed protein directly, and immunization with short peptide does not activate B cells that recognize the native protein. As recent clinical trials of B cell-depleting therapies in MS have suggested a role for B cells in driving disease in humans, there is an urgent need for animal models that incorporate B cell-recognition of autoantigen. To this end, we have generated a new fusion protein containing the extracellular domain of the mouse version of myelin oligodendrocyte glycoprotein (MOG) as well as N-terminal fusions of a His-tag for purification purposes and the thioredoxin protein to improve solubility (MOGtag). A tobacco etch virus (TEV) protease cleavage site was incorporated to allow the removal of all tag sequences, leaving only the pure MOG1-125 extracellular domain. Here, we describe a simple protocol using only standard laboratory equipment to produce large quantities of pure MOGtag or MOG1-125. This protocol consistently generates over 200 mg of MOGtag protein. Immunization with either MOGtag or MOG1-125 generates an autoimmune response that includes pathogenic B cells that recognize the native mouse MOG.
MS is een menselijke ziekte gekenmerkt door chronische ontsteking en neurodegeneratie van het CZS die wordt gedacht te worden veroorzaakt door een auto-immuunreactie gericht myeline. Het verlies van myeline en axonen na verloop van tijd leiden tot een geleidelijke afname van de cognitieve en motorische functie 1. "Experimentele auto-immune Encefalomyelitis" is een overkoepelende term voor diermodellen van auto-immuunziekten gericht CNS myeline. Als menselijke MS, EAE wordt doorgaans gekenmerkt door infiltratie van immuuncellen van het centrale zenuwstelsel en in sommige gevallen, demyelinisatie 2. De mate waarin een bepaalde EAE model lijkt op menselijke MS mede afhankelijk van de soort of stam gebruikt en de complexiteit van het onderliggende anti-myeline auto-immuunreactie.
Anti-myeline autoimmuniteit kan experimenteel worden geïnduceerd op verschillende manieren, maar de meest gebruikte methode tegenwoordig gebruikt om muizen te immuniseren met een korte peptide van aminozuren nabootsen van de immunodominante CD4 <sup> + T cel epitoop van een myeline-eiwit. Dit is het minimum vereiste om een pathogene respons te induceren. Misschien is de meest voorkomende van deze is een 21 aminozuur peptide afgeleid van myeline oligodendrocyt glycoproteïne (MOG 35-55), die wordt gebruikt om EAE te induceren in C57BL / 6 muizen 3. Voor sommige experimentele doeleinden wenselijk of zelfs noodzakelijk om immuniseren met grotere eiwitantigenen en inderdaad zijn er verschillende voordelen aan deze via immunisatie met korte peptide. Ten eerste omdat MHC beperking korte peptiden gewoonlijk alleen effectief in een zeer beperkt aantal stammen, terwijl grotere eiwitantigenen die ofwel het gehele eiwit of een specifiek domein kan normaal worden voor de presentatie in verschillende soorten verwerkt meerdere inteelt muizenstammen of 4. Ten tweede, een groter eiwit antigeen kan induceren een meer complexe immuunrespons waarin meer soorten lymfocyten in antigenherkenning, plaats limiting antigeenherkenning aan CD4 + T-cellen. Bijvoorbeeld, B-cellen via hun B-cel receptor (BCR) communiceren rechtstreeks met geheel in plaats van verwerkte dierlijke eiwitten. Wij en anderen hebben aangetoond dat B-cellen geactiveerd door MOG 35-55 immunisatie niet herkent MOG eiwitten 5. Aangezien B-cellen werden onlangs aangetoond dat een pathogene rol spelen bij menselijke MS 6, EAE modellen die B-cellen bevatten bij auto pathologie steeds belangrijker.
Ondanks de voordelen van grotere eiwitantigenen EAE te induceren, blijven er enkele commercieel beschikbare bronnen voor dergelijke eiwitten. Inderdaad, terwijl korte peptiden zoals MOG 35-55 snel en tegen relatief lage kosten kunnen worden gesynthetiseerd, de commerciële mogelijkheden MOG eiwitten zijn beperkt en de kosten aanzienlijk te schaffen. Toch zijn er een aantal expressievectoren beschikbaar voor onderzoeksgroepen om MOG extracellulair domein te genereren (MOG 1-125) zelf. however, alle expressiesystemen die we hebben geïdentificeerd in de literatuur zijn gebaseerd op oudere technologie die sindsdien zijn vervangen door efficiëntere expressiesystemen 7. Daarnaast bieden de meeste zijn gebaseerd op ratten of mensen MOG 8. Voor sommige onderzoeken van auto-immuniteit bij muizen, een antigeen op basis van de muis MOG autoantigeen voorkeur. Tenslotte alle MOG-gebaseerde eiwitten die we hebben geïdentificeerd, commerciële of als expressievectoren, zijn fusie-eiwitten die additionele aminozuren aan de MOG 1-125 base. Deze omvatten een tag voor zuivering en meestal andere sequenties ook, waarvan vele een functie we konden identificeren.
Om deze beperkingen te pakken, genereerden wij een nieuwe fusie-eiwit op basis van de muis MOG extracellulaire domein gefuseerd aan een label met thioredoxine de bekende onoplosbaarheid van MOG eiwit 5 bestrijden. De tag sequentie bevat ook een 6xHis sequentie voor zuivering en een TEV protease kelAvage plaats dat zorgt voor de volledige verwijdering van tagsequenties, indien gewenst. Dit is de enige methode die we ons bewust van de zuivere MOG 1-125-eiwit te genereren. Om de productie van grote hoeveelheden eiwit te vergemakkelijken, de MOG 1-125 sequentie is codon-geoptimaliseerd voor bacteriële expressie en MOG tag fusie-eiwit werd in de pET-32 expressiesysteem ingebracht. Hier beschrijven we in detail het protocol te produceren en te zuiveren MOG tag eiwit en pure MOG 1-125, met behulp van niet-gespecialiseerde apparatuur beschikbaar voor de meeste immunologie laboratoria.
