We describe a simple protocol using only basic lab equipment to generate and purify large quantities of a fusion protein that contains mouse Myelin Oligodendrocyte Glycoprotein. This protein can be used to induce experimental autoimmune encephalomyelitis driven by both T and B cells.
Multiple sclerosis (MS) is a chronic inflammatory disease of the central nervous system (CNS), thought to occur as a result of autoimmune responses targeting myelin. Experimental autoimmune encephalomyelitis (EAE) is the most common animal model of CNS autoimmune disease, and is typically induced via immunization with short peptides representing immunodominant CD4+ T cell epitopes of myelin proteins. However, B cells recognize unprocessed protein directly, and immunization with short peptide does not activate B cells that recognize the native protein. As recent clinical trials of B cell-depleting therapies in MS have suggested a role for B cells in driving disease in humans, there is an urgent need for animal models that incorporate B cell-recognition of autoantigen. To this end, we have generated a new fusion protein containing the extracellular domain of the mouse version of myelin oligodendrocyte glycoprotein (MOG) as well as N-terminal fusions of a His-tag for purification purposes and the thioredoxin protein to improve solubility (MOGtag). A tobacco etch virus (TEV) protease cleavage site was incorporated to allow the removal of all tag sequences, leaving only the pure MOG1-125 extracellular domain. Here, we describe a simple protocol using only standard laboratory equipment to produce large quantities of pure MOGtag or MOG1-125. This protocol consistently generates over 200 mg of MOGtag protein. Immunization with either MOGtag or MOG1-125 generates an autoimmune response that includes pathogenic B cells that recognize the native mouse MOG.
MS är en sjukdom hos människor som kännetecknas av kronisk inflammation och neurodegeneration i CNS som tros för att drivas av en autoimmun respons riktas mot myelin. Anti-myelin autoimmunitet kan experimentellt inducerad på flera sätt, men den vanligaste metoden som används idag är att immunisera möss med en kort peptid av aminosyror som imiterar den immunodominanta CD4 <supp> + T-cellepitop av en myelinprotein. Detta motsvarar minimikravet för att framkalla en patogen respons. Kanske den vanligaste av dessa är ett 21 aminosyrapeptid härledd från myelin oligodendrocyt-glykoprotein (MOG 35-55), som används för att inducera EAE hos C57Bl / 6-möss tre. Men det är för vissa försöksändamål önskvärd eller ens nödvändigt att immunisera med större proteinantigener och faktiskt det finns flera fördelar med detta över immunisering med kort peptid. Först på grund av MHC-restriktion, korta peptider är oftast bara effektiva i ett mycket begränsat antal av stammar, medan större proteinantigener som representerar antingen hela proteinet eller en specifik domän kan behandlas normalt för presentation i flera stammar inavlade mus eller ens i olika arter 4. För det andra är en större proteinantigen med förmåga att inducera en mer komplex immunsvar som innefattar flera typer av lymfocyter i antigenigenkänning, snarare än limiting antigen-igenkänning till CD4 + T-celler. Till exempel, B-celler via sin B-cellsreceptor (BCR) interagera direkt med hela snarare än bearbetat protein. Vi och andra har visat att B-celler aktiveras av MOG 35-55 immunisering inte känner igen MOG protein 5. Eftersom B-celler har nyligen visat sig spela en patogen roll i människans MS 6, EAE-modeller som innehåller B-celler vid autoimmun patologi blir allt viktigare.
Trots fördelarna med att använda större proteinantigener för att inducera EAE, kvarstår några kommersiellt tillgängliga källor för sådana proteiner. Faktiskt, medan korta peptider som MOG 35-55 kan syntetiseras mycket snabbt och till en relativt låg kostnad, de kommersiella alternativ för MOG-proteinet är begränsade och kostnad betydligt mer att köpa. Vidare är de flesta är baserade på råtta eller människa MOG 8. För vissa undersökningar av autoimmunitet hos möss, är en antigenbaserade på musen MOG autoantigen föredra. Slutligen, alla MOG-baserade proteiner som vi har identifierat, antingen kommersiella eller som expressionsvektorer, är fusionsproteiner som innehåller ytterligare aminosyror till MOG 1-125 bas. Dessa inkluderar en tagg för rening och oftast andra sekvenser också, många med en funktion som vi inte kunde identifiera.
