Kaposi-Sarkom (KS) ist ein Tumor induziert durch eine Infektion mit onkogenen Viren menschliche Herpesvirus-8/KS Herpesvirus (HHV-8/KSHV). Die Endothelzellen Kultur Modell hier beschriebenen eignet sich eindeutig für die Untersuchung der Mechanismen, mit denen KSHV Wirtszellen verwandelt.
Kaposi-Sarkom (KS) ist eine ungewöhnliche Tumor bestehend aus wuchernden Spindel Zellen, die durch Infektion der Endothelzellen (EG) mit KSHV initiiert und entwickelt sich in den meisten Fällen im Rahmen der Immunsuppression. Trotz jahrzehntelanger Forschung optimale Behandlung von KS bleibt schlecht definiert und klinischen Ergebnisse sind besonders ungünstig in beschränkten Ressourcen. KS-Läsionen sind getrieben von pathologischen Angiogenese, chronische Entzündungen und Onkogenese und verschiedene in-vitro- Kultur Zellmodelle wurden entwickelt, um diese Prozesse zu studieren. KS ergibt sich aus KSHV-infizierten Zellen endothelialen Ursprungs, also EC-Linie Zellen am besten geeignete in-vitro- Surrogate von der Spindel Zelle Vorläufer zur Verfügung stellen. Da EG haben eine begrenzte in-vitro- Lebensdauer, und die onkogenen Mechanismen KSHV angestellt sind weniger effizient als die der anderen tumorigenic Viren, es war jedoch schwierig zu beurteilen, die Prozesse der Transformation in der PV oder Telomerase-verewigt EC Daher wurde ein neuartiger EG basierende Kultur-Modell entwickelt, die Transformation nach Infektion mit KSHV bereitwillig unterstützt. Ektopische Expression der Gene des menschlichen Papillomavirus Typ 16 E6 und E7 erweiterte Kultur von Alter und Durchgang abgestimmt Mock und KSHV-infizierten EG ermöglicht und unterstützt die Entwicklung einer wahrhaft transformierten (d. h. tumorigenic) Phänotyp in infizierten Zellkulturen . Diese gefügig und hoch reproduzierbare Modell KS hat die Entdeckung von mehreren wesentlichen Signalwege mit hohem Potenzial für die Übersetzung in klinischen Umgebungen erleichtert.
Kaposi-Sarkom (KS) ist ein Multi-focal Angioproliferative Tumor Auswirkungen auf Haut, Schleimhaut und viszeralen Websites, der am häufigsten in der Umgebung von fortgeschrittenen Immununterdrückung1entwickelt. Vier epidemiologische Formen sind beschrieben worden: Classic, eine träge Form, das in der Regel ältere Menschen des Mittelmeerraums und des Nahen Ostens Erbes; iatrogene, durch Behandlung mit immunsuppressiven Medikamenten nach Organtransplantationen; Epidemie, eine AIDS-definierenden Krebs; und eine HIV-unabhängige Form häufig bei Kindern in endemischen Gebieten in Afrika endemisch. Mit dem Aufkommen der effektive Kombination Anti-retroviral Drogen-Therapien für die Behandlung von HIV ist Epidemie KS in Entwicklungsländern viel seltener diagnostiziert. Jedoch bleiben die klinisch aggressiven endemischen und epidemischen Formen unter die am häufigsten diagnostizierten Krebsarten in vielen afrikanischen Ländern2,3,4. Daher ist die Identifizierung von wirksame Pathogenese gezielt Medikamente zur Behandlung von KS Forschungsschwerpunkt.
Histologisch, zeichnen sich KS-Läsionen durch umfangreiche aber abnorme Neovaskularisation wobei Spindel Zellen EG Ursprungs diskontinuierliche vaskulären Netzwerken5bilden. Diese abnormen Gefäße (“vaskuläre Schlitze”) erlauben Extravasation der Erythrozyten, die Läsionen ihre charakteristische Farbe geben. Darüber hinaus enthalten Läsionen zahlreiche Leukozyten, die chronischen Entzündung (z.B. Lymphozyten, Makrophagen und Plasmazellen) charakterisieren. Rückbildung der KS-Läsionen nach Immunrekonstitution wurde beschrieben, was darauf hindeutet, dass KS Funktionen einer hyper-proliferative Läsion und wahre Tumor6,7,8,9.
KS Herpesvirus (KSHV), dem Erreger der KS, wurde im Jahr 199410identifiziert. Seit dann viele in Vitro Zellkultur Modelle entwickelt worden um Pathogenese Studien ermöglichen, einschließlich der Zellen aus Biopsie Tumormaterial und primäre explantiert oder Telomerase exprimierenden EG infiziert mit KSHV in-vitro-11 , 12 , 13 , 14 , 15 , 16 , 17 , 18. keines der derzeit verfügbaren Modelle voll rekapituliert die KS Tumor Mikroumgebung, aber alle haben wertvolles Wissen zu unserem Verständnis der Pathobiologie KSHV Infektion beigetragen. Im Gegensatz zu den anderen bekannten tumorigenic menschliche Herpesvirus Epstein – Barr-Virus (EBV), KSHV nicht leicht transformiert Zellen in Kultur nach de Novo Infektion19,20,21, 22. aber diese Einschränkung überwunden durch transducing primären menschlichen EG entweder gemischt mikrovaskuläre oder lymphatischen Ursprungs mit der E6 und E7 Gene aus humanem Papillomavirus Typ 16 vor der Infektion mit KSHV23,24 . Ausdruck dieser exogene Onkogene erhöht drastisch das transformierende Potenzial der KSHV in-vitro- teilweise durch weitere Hemmung der Retinoblastoma Protein und p5323,24. Diese EG-Transduktion Methode hat mehrere Labore, wichtige Veränderungen im Host zu identifizieren erlaubt Zelle Genexpression, die durch KSHV Infektion hervorgerufen und, die Erleichterung der KS Zelle überleben und Verbreitung25,26 , 27 , 28 , 29 , 30 , 31 , 32. der hierin beschriebenen Protokolle sind unkompliziert und hoch reproduzierbare und führt zu der Generation von Alter und Durchgang abgestimmt KSHV-infizierten EG und Mock-infizierten Steuerelemente, können viel länger als primäre Zellen gezüchtet werden und werden, ermöglichen Sie die Untersuchung von onkogenen Mechanismen KSHV angestellt.
