Analysis of the mitochondrial structure-function relationship is required for a thorough understanding of the regulatory mechanisms of mitochondrial functionality. Fluorescence microscopy is an indispensable tool for the direct assessment of mitochondrial structure and function in live cells and for studying the mitochondrial structure-function relationship, which is primarily modulated by the molecules governing fission and fusion events between mitochondria. This paper describes and demonstrates specific methods for studying mitochondrial structure and function in live as well as in fixed tissue in the model organism Drosophila melanogaster. The tissue of choice here is the Drosophila ovary, which can be isolated and made amenable for ex vivo live confocal microscopy. Furthermore, the paper describes how to genetically manipulate the mitochondrial fission protein, Drp1, in Drosophila ovaries to study the involvement of Drp1-driven mitochondrial fission in modulating the mitochondrial structure-function relationship. The broad use of such methods is demonstrated in already-published as well as in novel data. The described methods can be further extended towards understanding the direct impact of nutrients and/or growth factors on the mitochondrial properties ex vivo. Given that mitochondrial dysregulation underlies the etiology of various diseases, the described innovative methods developed in a genetically tractable model organism, Drosophila, are anticipated to contribute significantly to the understanding of the mechanistic details of the mitochondrial structure-function relationship and to the development of mitochondria-directed therapeutic strategies.
Mitokondrier er klassisk beskrevet som den cellulære kraftcenter, da de er de vigtigste pladser i energiproduktionen i differentierede celler. Desuden mitokondrier spiller en kritisk rolle i stofskiftet, varmeproduktion, lipid modifikation, calcium og redox homeostase, orkestrering af celle- signalering processer, etc 1. Mitokondrier spiller også en aktiv rolle i induktion af celledød 2, samt i cellecyklus regel 3. Sådanne multifunktionalitet rejser følgende grundlæggende spørgsmål: a) hvordan mitokondrier udføre alle disse funktioner samtidig og b) er der specifikke mitokondrie puljer eller underområder, der er specialiseret til forskellige funktioner? I denne forbindelse er det vigtigt at bemærke, at de multifunktionelle mitokondrier er dynamiske i deres form, størrelse og struktur i individuelle celler, og at steady-state form af mitokondrier kan variere mellem celletyper. Årtiers forskning fra forskellige laboratorier tyder på, at ændring af mitokondrie form, størrelse og struktur, samlet kaldes mitokondrie dynamik, er afgørende for at opretholde forskellige mitokondrie funktioner 4,5,6. Disse resultater rejser muligheden for, at mitokondrier kan udføre deres multifunktionalitet i kraft af deres strukturelle dynamik.
Omfattende indsats i gang for at forstå den mitokondrielle struktur-funktion forhold. Dynamikken i mitokondriestruktur primært vedligeholdes af deres evne til at undergå fission og fusion arrangementer med hinanden. Spaltning af store mitokondrier omdanner dem til mindre mitokondrielle elementer, mens fusion mellem to mindre mitokondrier fletter dem i en større mitokondrie element 7. Desuden kan forbigående fusion af to mitokondrier forekomme for at tillade blanding af deres indhold. De fission og fusion begivenhederne i de indre og ydre mitochondriemembraner er omhyggeligt styret af specSp ec ifik sæt af proteiner. Kernen fission maskiner er sammensat af dynamin-relateret protein 1 (Drp1), som er rekrutteret fra cytosolen til mitokondrierne ved dets interaktion med visse bona fide mitokondrielle proteiner (f.eks Fis1 eller MSpol frem1), mens Drp1 funktion også kan reguleres ved andre proteiner på mitokondrie flade 4. Selv Drp1 opererer på den ydre membran, dets fission evner påvirke den indre membran samt. Den orkestrering af fission af ydre og indre mitochondriemembraner er ikke godt forstået. På den anden side er fusion af den indre membran er omfattet kernen af aktiviteterne i Opa1, mens mitofusins regulerer fusion af ydermembranen 5. Den resterende del af de modvirker fission og fusion begivenheder mitokondrier diktere steady-state mitokondrie form i en celle. For eksempel ville undertrykkelse af mitokondrie fission resultere i fuldstændig og enstemmigt fusion, medens overaktivitet af mitokondrierl fission ville resultere i fragmentering af mitokondrier 3.
Studiet af mitokondriestruktur-funktion forhold aktiverer primært to gratis fremgangsmåder: a) analyser af de cellulære og organismal fænotyper efter genetisk manipulation af mitokondrielle fission / fusionsproteiner og b) direkte vurdering af mitokondrisk struktur og funktion. Det er bemærkelsesværdigt, at genetiske analyser ikke altid kan afsløre direkte funktionalitet af molekylet ved hånden (i dette tilfælde, mitokondriske fission / fusionsproteiner), som kan opstå de fænotyper på grund af sekundære virkninger. Derfor er det af største vigtighed at udvikle og anvende værktøjer til at studere mitokondrielle struktur og funktion direkte. Fluorescensmikroskopi-baserede værktøjer er blevet udviklet til at studere mitokondrielle struktur og funktion i levende og faste Drosophila melanogaster væv, belyse betydningen af mitokondrie dynamik in vivo 9. Disse og beslægtede metoder er beskrevet her, med det mål at studere mitokondrisk struktur og funktion i Drosophila æggestok.
Den Drosophila æggestok består af kimcellelinje og somatiske slægter, som stammer fra deres respektive voksne stamceller, der findes i germarium 10,11.
