Adipose-derived stromal cells may be useful for engineering new tissue from a patient’s own cells. We present a protocol for the isolation of a subpopulation of human adipose-derived stromal cells (ASCs) with increased osteogenic potential, followed by application of the cells in an in vivo calvarial healing assay.
Invasive cancers, major injuries, and infection can cause bone defects that are too large to be reconstructed with preexisting bone from the patient’s own body. The ability to grow bone de novo using a patient’s own cells would allow bony defects to be filled with adequate tissue without the morbidity of harvesting native bone. There is interest in the use of adipose-derived stromal cells (ASCs) as a source for tissue engineering because these are obtained from an abundant source: the patient’s own adipose tissue. However, ASCs are a heterogeneous population and some subpopulations may be more effective in this application than others. Isolation of the most osteogenic population of ASCs could improve the efficiency and effectiveness of a bone engineering process. In this protocol, ASCs are obtained from subcutaneous fat tissue from a human donor. The subpopulation of ASCs expressing the marker BMPR-IB is isolated using FACS. These cells are then applied to an in vivo calvarial defect healing assay and are found to have improved osteogenic regenerative potential compared with unsorted cells.
Grote botdefecten als gevolg van letsel, infectie of invasieve kanker hebben een aanzienlijke invloed op het herstel en de kwaliteit van het leven van een patiënt. Technieken bestaan om deze gebreken gezond bot vulling van elders in lichaam van de patiënt, maar deze overdracht draagt zijn eigen morbiditeit en risico van complicaties 1, 2, 3. Bovendien zijn sommige gebreken zo omvangrijk of complex die voldoende donorbot beschikbaar is om het defect te vullen. Protheses zijn een mogelijke optie voor het vullen van botdefecten, maar deze zijn geassocieerd met een aantal nadelen waaronder infectierisico werkende hardware en vreemd lichaam reactie 4.
Om deze redenen is er grote belangstelling voor de mogelijkheid van engineering van biologische botvervangers behulp van eigen cellen van de patiënt 5. -Vet-afgeleide stromale cellen (ASC)hebben potentieel voor deze toepassing, omdat zij overvloedig aanwezig in de eigen vetweefsel van de patiënt en ze hebben het vermogen om botdefecten te helen door het genereren van nieuw botweefsel 6, 7 getoond. ASC's zijn een diverse populatie van cellen en verscheidene studies hebben aangetoond dat het selecteren van specifieke celoppervlak markers celpopulaties met verhoogde osteogene activiteit 8, 9 kan produceren. ASC selecteren met het hoogste osteogeen potentieel zou de waarschijnlijkheid toenemen dat een scaffold bezaaid met deze cellen een grote botdefecten kan regenereren.
Bone morfogenetisch eiwit (BMP) signalering is essentieel voor het reguleren van differentiatie en bot- vorming 10 en het BMP receptor type IB (BMPR-IB) is bekend belangrijk voor osteogenese in ASCs 11 zijn. Onlangs hebben we aangetoond dat expressie van BMPR-IB kan be gebruikt om te selecteren voor het ASC met verhoogde osteogene activiteit 12. Hier laten we een protocol voor de isolatie van BMPR-IB tot expressie ASC's uit menselijk vet, gevolgd door een analyse van hun osteogene activiteit met een in vivo model calvarial defect.
Kritische stappen in het protocol
Tijdens de oogst van ASC, de kritische stap is voldoende vertering van vet met collagenase. Onvoldoende vertering leidt tot een lage opbrengst van ASC.
