Denne protokollen beskriver en effektiv og praktisk analytiske prosessen eksempel utvinning og samtidige fastsettelse av flere rusmidler, doksorubicin (DOX), mitomycin C (MMC) og en cardio-giftig DOX metabolitten, doxorubicinol (DOXol), biologisk prøver fra prekliniske brystet tumor modell behandlet med hydrogenion formuleringer av synergistisk medikament kombinasjon.
Kombinasjon kjemoterapi er ofte brukt i klinikken for behandling; tilhørende bivirkninger på normalt vev kan imidlertid begrense dens terapeutiske nytte. Hydrogenion-baserte narkotika kombinasjon har vist seg å redusere problemene gratis narkotika Kombinasjonsbehandling. Våre tidligere studier har vist at kombinasjonen av to anticancer legemidler, doksorubicin (DOX) og mitomycin C (MMC), produsert en synergistisk effekt mot både murint og menneskelige brystkreft celler i vitro. DOX og MMC co lastet polymer-lipid hybrid nanopartikler (DMPLN) forbigått ulike middelklasseinnbyggere transporter pumper som overdrar multidrug motstand og viste forbedret effekt i brystet tumor modeller. Sammenlignet med konvensjonelle Løsningsskjemaer, ble slik overlegen effekten av DMPLN tilskrevet synkronisert farmakokinetikken av DOX og MMC og økt intracellulær narkotika biotilgjengelighet innen kreftceller aktiveres av nanocarrier PLN.
For å evaluere farmakokinetikken og bio-distribusjon av administrert co DOX og MMC både gratis løsning og hydrogenion former, en enkel og effektiv multi-Drug analysemetode bruker omvendt-fase høy ytelse flytende kromatografi (HPLC) var utviklet. I motsetning til tidligere rapportert metoder som analysert DOX eller MMC individuelt i plasma, er denne nye HPLC metoden kjøpedyktig samtidig quantitate DOX, MMC og en cardio-giftig DOX hovedmetabolitten, doxorubicinol (DOXol), i ulike biologiske matriser ( f.eks fullblod, brystet svulsten og hjertet). En dobbel fluorescerende og ultrafiolett absorberende sonde 4-methylumbelliferone (4-MU) ble brukt som en intern standard (er) for ettrinns påvisning av flere narkotika analyse med annerledes oppdagelsen bølgelengder. Denne metoden ble brukt for å bestemme konsentrasjonen av DOX og MMC levert av både hydrogenion og løsning tilnærminger i fullblod og ulike vev i en orthotopic brystet tumor murine modell. Analysemetode presentert er et nyttig verktøy for pre-klinisk analyse hydrogenion-baserte levering av rusmiddelkombinasjoner.
Kjemoterapi er en primær behandling modalitet for mange kreft men det er ofte forbundet med alvorlige bivirkninger og begrenset effekt på grunn av resistens og andre faktorer1,2,3. For å forbedre resultatet av kjemoterapi er narkotika kombinasjon regimer brukt i klinikken basert på hensyn som ikke-overlappende toksisitet, ulike mekanismer av stoffet handling, og ikke-cross narkotika motstand4,5 , 6. i kliniske forsøk, en bedre svulst svarprosent ble ofte observert bruke samtidig administrert rusmiddelkombinasjoner sammenlignet med en diett av sekvensiell stoffet levering7,8. Men på grunn av sub-optimale bio-fordelingen av gratis narkotika, kan samtidig injeksjon av flere stoffer forårsake fremtredende normalt vev toksisitet som oppveier den terapeutiske effekt9,10,11. Nanocarrier-baserte narkotika-leveranser har vist seg å endre farmakokinetikken og bio-fordelingen av innkapslede narkotika, styrke svulst målrettede akkumulering12,13,14. Som omtalt i våre siste artikler, har nanopartikler co lastet med synergistisk rusmiddelkombinasjoner vist evne til å redusere problemene gratis rusmiddelkombinasjoner, på grunn av deres kontrollert timelige og romlig co levering av flere legemidler tumor vev, aktivere synergistisk narkotika effekter mot kreft celler4,15,16. Resultatet har overlegen terapeutiske effekten og lav toksisitet blitt vist både prekliniske og kliniske studier4,17,18.
Våre tidligere i vitro studier funnet at kombinasjonen av to anticancer legemidler, doksorubicin (DOX) og mitomycin C (MMC), produsert en synergistisk effekt mot flere bryst kreft celler linjer, og videre co lasting DOX og MMC i polymer-lipid hybrid nanopartikler (DMPLN) overvant ulike multi-resistente tilknyttede middelklasseinnbyggere pumper (f.eks, P-glykoprotein og bryst kreft motstandsdyktig protein)19,20,21. I vivo, DMPLN aktivert romlig-temporalt co levering av DOX og MMC svulst områder og økt bioavailability av narkotika kreftceller, som indikert av moderasjon av dannelse av DOX metabolitten doxorubicinol (DOXol)22. Resultatet forbedret av DMPLN svulst celle apoptose, tumor vekst hemming og langvarig vert overlevelse sammenlignet med gratis DOX og MMC eller en liposomal DOX formulering22,23,24, 25.
Analysere faktiske mengden av narkotika co levert av en nanocarrier er avgjørende for å utforme effektive hydrogenion formuleringer. Mange metoder har blitt utviklet for å analysere plasma nivået av enkelt DOX eller MMC doser bruker Væskekromatografi (HPLC) alene eller i kombinasjon med massespektrometri (MS)26,27,28 , 29 , 30 , 31 , 32 , 33 , 34. disse metodene er imidlertid ofte tidkrevende og upraktisk for Kombinasjonsbehandling som et stort antall biologiske prøver må være forberedt separat for analyse av flere stoffer (noen ganger inkludert narkotika metabolitter). I tillegg til sterk plasma protein binding av DOX og MMC har røde blod celler også en stor evne til å binde og konsentrere mange anticancer narkotika35,36. Plasma analyse for DOX eller MMC kan dermed Beskytt faktiske blod narkotika konsentrasjoner. Den nåværende arbeidet (figur 1) beskriver en enkel og robust flere narkotika analysemetode bruker omvendt fase HPLC samtidig og quantitate DOX, MMC og DOX metabolitten doxorubicinol (DOXol) fra fullblod og ulike vev ( f.eks svulster). Det har vært anvendt for å bestemme farmakokinetikken og bio-distribusjon av DOX og MMC og dannelsen av DOXol etter stoffet levering via gratis løsninger eller hydrogenion skjemaer (dvs.DMPLN og liposomal DOX) i en orthotopically implantert murine bryst-svulst musemodell etter intravenøs (IV) injeksjon22.
Sammenlignet med andre brukt kromatografiske metoder som aktiverer deteksjon av en enkelt stoff Art samtidig, er dagens HPLC protokollen kjøpedyktig samtidig quantitate tre narkotika forbindelser (DOX, MMC og DOXol) i den samme biologisk matrisen uten behov for å endre mobile fasen.
