En protokoll for utviklingen av en elektrokjemisk DNA biosensor bestående av en polylactic syre-stabilisert, gull nanopartikler endret, trykt karbon elektrode for å oppdage Vibrio parahaemolyticus presenteres.
Vibrio parahaemolyticus (V. parahaemolyticus) er en felles infeksjon patogen som bidrar til en stor andel av folkehelseproblemer globalt, betydelig påvirker frekvensen av menneskelige dødelighet og sykelighet. Konvensjonelle metoder for påvisning av V. parahaemolyticus som kultur-baserte metoder, immunologiske analyser og molekylær-baserte metoder krever komplisert eksempel håndtering og er tidkrevende, kjedelig og dyrt. Nylig har biosensors vist seg for å være en lovende og omfattende gjenkjennings-metoden med fordelene ved rask oppdagelsen, kostnadseffektivitet og praktiske. Denne forskningen fokuserer på å utvikle en rask metode for å oppdage V. parahaemolyticus med høy selektivitet og følsomhet bruker prinsippene om DNA hybridisering. I arbeidet, ble karakterisering av syntetisk polylactic syre-stabilisert gull nanopartikler (PLA-AuNPs) oppnådd med X-ray Diffraksjon (XRD), UV-synlig spektroskopi (UV-Vis), overføring elektronmikroskop (TEM), felt-utslipp Skanning elektronmikroskop (FESEM), og syklisk Voltammetry (CV). Vi har også gjennomført ytterligere testing stabilitet, følsomhet og reproduserbarhet for PLA-AuNPs. Vi fant at PLA-AuNPs dannet en lyd struktur av stabilisert nanopartikler i vandig løsning. Vi har også observert at følsomheten forbedret mindre kostnad overføring motstand (Rct) verdien og en økning av aktive areal (0.41 cm2). Utviklingen av vårt DNA biosensor var basert på endring av en trykt karbon elektrode (SPCE) med PLA-AuNPs og bruke methylene Blåfarge (MB) som redoks indikator. Vi vurdert hendelsene immobilisering og blanding av differensial puls voltammetry (DPV). Vi fant ut at komplementære, ikke-utfyllende, og oligonucleotides var spesielt preget av den ferdige biosensor. Det viste også pålitelig følsom oppdagelsen kryssreaktivitet studier mot ulike mat-borne patogener og identifikasjon av V. parahaemolyticus i ferske.
Et sentralt tema offentlige og vitenskapelig debatt de siste årene, matforgiftning er hovedsakelig forbundet med 3 agenter: mikroorganismer1, kjemikalier2og parasitter3. Forurenset mat kan føre til alvorlige helseproblemer hos mennesker, spesielt i risikogruppen for høyere i de med svak uimottakelig systemer, eldre, gravide kvinner, spedbarn og små barn4. Med mer enn en million tilfeller av akutt diaré forekommer årlig i barn under 5 år i Afrika, Asia og Latin-Amerika, matforgiftning er en stor global sykdom5,6 og Verdens helseorganisasjon har etablert mikroorganismer som det viktigste bidragsyter7. Vibrio parahaemolyticus skiller seg ut blant de mest anerkjente virulente stammene. Vanligvis finner i kyst, marinsystemer og hav8, er det en Gram-negative bakterie, som blir aktivt i høy salt miljøer, og forårsaker alvorlige menneskelige gastroenteritt når spist i mangelfullt kokte, feilsendt eller rå marine produkter9. I tillegg gjør eksisterende medisinske tilstander i noen mennesker dem utsatt for sår infeksjon, septicemia eller øre infeksjon oppstår fra V. parahaemolyticus10. Virulens faktorene av V. parahaemolyticus hemolysins er delt inn i to typer som bidrar til sykdom patogenesen: thermostable direkte hemolysin (TDH) kodet av tdh gener og TDH-relaterte hemolysin kodet av trh gener11. Virulens markører (tdh og trh gener) av V. parahaemolyticus finnes hovedsakelig i klinisk prøver ikke i miljømessige prøver.
V. parahaemolyticus har muligheten til å overleve under en rekke forhold, reagerer raskt miljøendringer12. Dens spredning mekanikk eskalerer dens farepotensial som sin toksisitet øker parallelt med cellen masse13. Enda verre, klimaendringer gir disse bakteriene med rikelig forhold å akselerere deres celle befolkningen vekst14. På grunn av dens høy frekvens må V. parahaemolyticus overvåkes langs verdikjeden mat, særlig i handel og produksjon av sjømat siden disse produktene er hvor de finnes i enorme mengder15,16 over hele verden. Foreløpig bakterier er identifisert og isolert bruker en rekke metoder inkludert biokjemiske tester, berikelse og selektiv media17, enzym knyttet immunomagnetic absorberende analysen (ELISA)18, puls-feltet gel geleelektroforese (PFGE) 19, latex agglutinerende tester og polymerase kjedereaksjon (PCR)-tester20. Disse metodene krever vanligvis kvalifisert personell, komplekse instrumenter og arbeidskrevende teknikker som ikke gir informasjon om forurensning umiddelbart. Dette begrenser alvorlig sannsynligheten for raskt oppdage skadelig forurensning og stedets programmer. Rask søkeverktøy forblir en enestående utfordring.
Biosensing fremstår som en lovende alternativ for påvisning av matbårne patogener fordi det tilbyr en tidsbesparende, kostnadseffektive, praktiske og sanntids analyse metoden21,22,23,24 . Men selv om det har vært mange positive resultater av analytt piggete prøver og standard løsning ved hjelp av biosensors, er det fortsatt en mangel på forskning på virkelige eksempler i vandig blandinger eller organisk ekstrakter25. Nylig har elektrokjemisk biosensors bruker direkte og/eller indirekte deoksyribonukleinsyre (DNA) oppdagelsen fått økt oppmerksomhet blant forskere, på grunn av deres spesifikke oppdaget komplementære målet via en hybridisering hendelsen26 , 27 , 28 , 29. disse unike tilnærminger er mer stabile forhold enzym-baserte biosensors, dermed tilby en lovende teknologi for miniatyrisering og kommersialisering. Målet for studien rapporterte her er å bygge en rask verktøy som kan oppdage V. parahaemolyticus med høy selektivitet, følsomhet og praktiske, basert på DNA sekvensen spesifisitet under hybridisering. Identifikasjon strategier innebærer kombinasjonen av polylactic syre-stabilisert gull nanopartikler (PLA-AuNPs)30 og trykt karbon elektroder (SPCEs) i nærvær av hybridisering indikatoren, methylene Blåfarge (MB). Potensialet i den utviklede oppdagelse konstruere er utforsket bakterier DNA lysate og frisk hjerteskjell utvalg.
Kritisk trinnene i et rammeverk for vellykkede utviklingen av denne typen elektrokjemisk biosensor er valg av riktige biologiske anerkjennelse elementer for svingeren (nukleinsyre eller DNA her); kjemisk tilnærming for å konstruere sensing laget svingeren; signaltransduksjon materiale; optimalisering av DNA immobilisering og hybridisering; og validering av den utviklede biosensor bruker virkelige eksempler.
