Passive og aktive kræft celle fænotyper var karakteriseret ved hjælp af kvantitative fase billeddannelse. Celle spredning, migration og morfologi assays blev integreret og analyseres i en simpel metode.
Erhvervelse af angiogene fænotype er en væsentlig bestanddel af flugten fra tumor vækstdvale. Selv om flere klassiske in vitro- assays (fxspredning, migration og andre) og i vivo modeller er blevet udviklet for at undersøge og karakterisere angiogene og ikke-angiogene celle fænotyper, er disse metoder tid labor intensiv, og ofte kræver dyre reagenser samt instrumenter og betydelig ekspertise. I en nylig undersøgelse brugte vi en roman kvantitative fase imaging (QPI) teknik til at gennemføre time-lapse og mærkning-gratis beskrivelser af angiogene og ikke-angiogene menneskelige osteosarkom KHOS celler. Et panel af cellulære parametre, herunder celle morfologi, spredning og motilitet, blev kvantitativt målt og analyseret ved hjælp af QPI. Denne roman og kvantitative tilgang giver mulighed for løbende og ikke-invasivt studere relevante cellulære processer, adfærd, og Karakteristik af kræftceller og andre celletyper i en enkel og integreret måde. En af de tidligste checkpoints i udvikling og progression af en solid tumor er erhvervelse af angiogene fænotype, kendetegnende for kræft. Denne progression indebærer en række biokemiske og molekylær processer1,2,3. En teknisk udfordring i studiet af denne afgørende skridt i tumor progression er manglen på værktøjer til løbende og kvantitativt karakterisere og differentiere mellem angiogene og ikke-angiogene fænotyper af levende kræftceller i en fordomsfri måde. De traditionelle assays bliver brugt til at undersøge de cellulære adfærd af angiogene og ikke-angiogene celler normalt kræver dyre reagenser og instrumenter, for eksempel celle spredning/migration undersøgelser vedrørende4,5, 6,7,8,9,10,11,12,13,14 eller supplerende i vivo evalueringer4,5,6,8,15,16, samt at kræve betydelig ekspertise og intensiv tid og arbejdskraft forbrug.
For nylig, kvantitative fase imaging (QPI) opstået som en ny teknik, der gør det muligt for time-lapse og mærkning-gratis vurdering af en række celle morfologi og funktionsmåde parametre17,18,19, 20 , 21 , 22. i modsætning til konventionelle Optisk mikroskopi, QPI kvantificerer variationer af faseskift pixel for pixel når lys passerer gennem et optisk objekt, og rekonstruerer en holograph med konverterede optisk tykkelse og mængde, således at direkte analyse af levende celler og følgende funktioner: (1) kvantitative imaging, (2) ikke-invasive og time-lapse imaging, (3) label-gratis billedbehandling og (4) samtidige multi-parameter billeddannelse. Disse funktioner gør QPI et kraftfuldt værktøj til at vurdere og forstå patologiske processer på cellulært niveau.
I en nylig undersøgelse, vi udnyttede QPI kvantitativt karakterisere og skelne mellem angiogene KHOS-A og ikke-angiogene KHOS-N fænotyper af menneskelige osteosarkom celler på en systematisk og kvantitative måde, kombinere analyser af celle morfologi, spredning, og motilitet23. Ved hjælp af billede analyse software, et panel af celle morfologiske og opførsel var parametre kvantitativt i forhold mellem angiogene og ikke-angiogene menneskelige osteosarkom celler og fem karakteristiske forskelle blev identificeret mellem disse to fænotyper. Denne nye fremgangsmåde giver en integreret og kvantitative platform for vurdering af en række forskellige biologisk relevante cellulære karakteristika.
I denne undersøgelse beskriver vi en in vitro-, ikke-invasiv, og etiket-fri metode ved hjælp af QPI kvantitativt kendetegner den angiogene og ikke-angiogene fænotyper af menneskelige osteosarkom celler. Flere cellulære parametre er blevet analyseret samtidigt ved denne integreret, høj overførselshastighed metode, herunder celle område, celle tykkelse, cellevolumen, spredning sats, fordobling tid, migration direkthed, motilitet hastighed, migration og motilitet.
