Hier beschrijven we een protocol voor een reproduceerbare laser vangt microdissection (LCM) voor het isoleren van trabecular gevlochten (TM) voor downstream RNA analyse. De mogelijkheid om het analyseren van wijzigingen in genexpressie in het Vertaalgeheugen zal helpen bij het begrijpen van de moleculaire mechanismen van TM-gerelateerde oogbeschadigingen en/of ziekten.
Laser vangt microdissection (LCM) heeft toegestaan gen expressie analyse van afzonderlijke cellen en cel populaties in weefselsecties verrijkt. LCM is een geweldig hulpmiddel voor de studie van de moleculaire mechanismen die ten grondslag liggen aan de celdifferentiatie, de ontwikkeling en de progressie van verschillende ziekten, met inbegrip van glaucoom. Glaucoom, die bestaat uit een familie van progressieve optic neuropathieën, is de meest voorkomende oorzaak van onomkeerbare blindheid wereldwijd. Structurele veranderingen en schade binnen het trabecular gevlochten (TM) kunnen leiden tot verhoogde intraoculaire druk (IOP), dat een belangrijke risicofactor is voor het ontwikkelen van glaucoom. De exacte moleculaire mechanismen die betrokken zijn echter nog steeds slecht begrepen. De mogelijkheid om het gen expressie analyses uit te voeren zullen van cruciaal belang bij het verkrijgen van meer inzicht in de functie van deze cellen en haar rol in de regulatie van de ontwikkeling van het IOP en glaucoom. Om dit te bereiken, verrijkt een reproduceerbare methode voor het isoleren van sterk TM uit bevroren secties van muis ogen en een methode voor de analyse van de expressie van de stroomafwaartse gen, zoals RT-qPCR en RNA-Seq nodig is. De hier beschreven methode is ontwikkeld om zeer zuivere TM van muis ogen isoleren voor downstream digitale PCR en microarray analyse. Deze techniek kunnen bovendien gemakkelijk aan te passen voor de isolatie van andere hoogverrijkt oogbeschadigingen en/of cellen en de cel compartimenten die moeilijk geweest te isoleren van de ogen van de muis. De combinatie van LCM en RNA analyse kan bijdragen tot een vollediger begrip van de cellulaire gebeurtenissen ten grondslag liggen aan de glaucoom.
Glaucoom is een groep van ziekten gekenmerkt door optische neuropathie en retinopathie die uiteindelijk tot onomkeerbare blindheid1,2 leidt. Geschat wordt dat door 2020 meer dan 70 miljoen mensen wereldwijd moeten met een vorm van de ziekte3,4,5,6,7 leven zal. Primaire open hoek glaucoom (POAG), het meest voorkomende type glaucoom, wordt gekenmerkt door een daling in aqueous humor (AH) uitstroom leidt tot verhoogde intraoculaire druk (IOP)8,9,10, 11,12,13,143,15,16,17,18. Linker onbehandeld, chronisch verhoogde IOP leidt tot progressieve en onomkeerbare schade aan de retina en oogzenuw hoofd veroorzaakt radiale blindheid1,2,19. Alle huidige methoden voor het vertragen van de progressie van glaucoom focus op vermindering van de IOP, hetzij door het verminderen van het percentage van de productie van AH door het straalvormig lichaam of verbetering van het uitstroom1,8,9, 10 , 11 , 12 , 13 , 14. het trabecular gevlochten (TM) speelt een cruciale rol bij het reguleren van de primaire AH uitstroom pathway actief en zijn onjuiste functie is een oorzakelijke factor voor hypertensieve glaucoom,1,,2,19. Echter de moleculaire mechanismen gekoppeld TM dysfunctie en hoe regelt het AH drainage worden nog niet volledig begrepen en is momenteel een belangrijk aandachtspunt van glaucoom onderzoek1,2,19, 20. terwijl verschillende genoom-brede vereniging studies (GWAS) een aantal genen aan glaucoom en een verhoogde weerstand tegen AH uitstroom faciliteit in de TM hebt gekoppeld, de exacte moleculaire mechanismen die leiden tot de ziekte niet nog volledig begrepen21 , 22 , 23 , 24 , 25.
Dierlijke modellen hebben aanzienlijk verbeterd onze huidige kennis van de progressie van de ziekte in glaucoom (uitgebreid herzien in3,15,16,26,27,28, 29,30,31,32,33). Verschillende baanbrekende methoden hebben ontwikkeld om te bestuderen van de TM34,35,,36 en deze methoden hebben wijd verbeid gebruikt om ons huidige begrip van normale en zieke weefsel. Een gebied dat is niet uitgebreid onderzocht is het gebruik van genetisch gemodificeerde Muismodellen te bestuderen van de moleculaire mechanismen van TM mislukking. Transgene knock-in en knock-out muis studies van TM verbonden genen, zoals Myocilin (Myoc)37,38 en39van het Cyp1b1, zijn de belangrijkste instrumenten voor het bestuderen van de moleculaire mechanismen van TM functie. Begrijpelijk, de geringe omvang van de TM in muizen vertegenwoordigt een ernstige hindernis die moet worden overwonnen om te beginnen met het bestuderen van dit weefsel. Muismodellen vertegenwoordigen een krachtig hulpmiddel voor het bestuderen van de genetica en de moleculaire mechanismen van ziekte, hoewel de vooruitgang in de LCM technologieën bieden de noodzakelijke hulpmiddelen om de studie van de kleinste en meest delicate weefsels, met inbegrip van de TM machtigen.
In dit verslag wordt een robuust en reproduceerbare methode beschreven voor de LCM van hoogverrijkt TM van muis ogen samen met de latere RNA isolatie en versterking voor downstream expressie analyse. Soortgelijke methoden hebben met succes gebruikt bij muizen te isoleren van andere soorten oog weefsels40,41,42,43,44, de methodologie die hierin gemeld kan worden toegepast op andere discrete weefsels van het oog om te studeren van RNA, microRNA, DNA en proteïnen. Belangrijk is dat maakt deze techniek het gebruik van genetisch gemodificeerde muizen voor een beter begrip van de moleculaire pathogenese van TM waardevermindering in glaucoom en oculaire ziekte3,15,16,17 ,18,26,31,45,46. De mogelijkheid om te isoleren van de TM van muis ogen door LCM zullen een handige techniek bij het verkrijgen van meer inzicht in de moleculaire mechanismen van verschillende oogbeschadigingen en/of ziekten.
De TM speelt een vitale rol in het actief onderhouden homeostatische IOP en zijn dysfunctie wordt algemeen erkend als de belangrijkste oorzakelijke factor voor hypertensieve glaucoom,1,,2,19.
