Hier presenteren we een protocol voor de productie van gramnegatieve Escherichia coli (E. coli) spheroplasts en gram-positieve Bacillus megaterium (B. megaterium) protoplasten duidelijk visualiseren en snel karakteriseren Peptide-bacteriën interacties. Dit biedt een systematische methode om te definiëren membraan lokaliseren en translocating peptiden.
Het gebruik van de confocal microscopie als een methode om te beoordelen van peptide lokalisatie patronen binnen bacteriën wordt gewoonlijk geremd door de grenzen van de resolutie van conventionele lichte microscopen. Zoals de resolutie voor een bepaalde Microscoop kan niet gemakkelijk worden verbeterd, presenteren we protocollen te transformeren de kleine staafvormige gramnegatieve Escherichia coli (E. coli) en gram-positieve Bacillus megaterium (B. megaterium) in grotere, gemakkelijk verbeelde bolvormige vormen spheroplasts of protoplasten genoemd. Deze transformatie kan waarnemers te snel en duidelijk bepalen of peptiden zelf in het bacteriële membraan (dat wil zeggen, membraan lokaliseren indienen) of kruis het membraan voor het invoeren van de cel (bijvoorbeeld translocating). Met deze aanpak presenteren we ook een systematische methode om te karakteriseren van peptiden als membraan lokaliseren of translocating. Hoewel deze methode kan worden gebruikt voor een verscheidenheid van membraan-actieve peptiden en bacteriestammen, we tonen het nut van dit protocol door het observeren van de interactie van Buforin II P11A (BF2 P11A), een antimicrobiële peptide (AMP), met E. coli spheroplasts en B. megaterium protoplasten.
Antimicrobiële peptiden (AMPs) hebben opgedaan aandacht vanwege hun potentiële gebruik als alternatieven voor conventionele antibiotica1,2,3,4,5. Versterkers doden bacteriën door ofwel translocating tussen de celmembranen en de interactie met intracellulaire componenten zoals nucleïnezuren of door het membraan veroorzaakt door lekkage van cel inhoud6permeabilizing. Naast hun gebruik als antibiotica, kan translocating versterkers worden aangepast voor drug delivery toepassingen omdat ze kunnen niet-onverwacht is het oversteken van de ondoordringbare celmembraan7,8. Daarom willen wij, begrijpen fundamentele AMP mechanismen van actie te leggen de basis voor hun gebruik in drug design.
Confocale microscopie biedt een manier om te beoordelen van lokalisatie patronen van fluorescently geëtiketteerde versterkers in bacteriecellen biedt inzichten in hun mechanisme van actie9,10,11,12, 13 , 14. door het labelen van de membraan van de bacteriën, kan men bepalen als een fluorescently geëtiketteerde peptide het membraan of de intracellulaire ruimte van een bacteriële cel lokaliseert. Deze techniek wordt echter beperkt door het kleine formaat en rod vorm van bacteriën, waardoor imaging uitdagend als gevolg van de grenzen van de resolutie van conventionele lichte microscopen en de variabele oriëntatie van de bacteriën op de dia15.
Het doel van de onderhavige methode is om verbeterde visualisatie van de fluorescently geëtiketteerde peptide lokalisatie patronen met behulp van de confocal microscopie. Visualisatie wordt versterkt door het draaien van de kleine, dunne, staafvormige gramnegatieve Escherichia coli (E. coli) en gram-positieve Bacillus megaterium (B. megaterium) bacteriën in uitgebreide, bolvormige vormen genoemd spheroplasts (voor gramnegatieve stammen) en protoplasten (voor gram-positieve stammen)16,17,18,19,20,21. Spheroplasts en protoplasten zijn gemakkelijker te afbeelding vanwege hun toegenomen omvang zowel hun symmetrische vorm, waardoor de richting van een bacterie op een dia niet relevant voor de beeldvorming. Daarnaast presenteren wij een systematische aanpak kwantitatief confocale microscopie om gegevens te analyseren om de versterkers te karakteriseren als beide membraan lokaliseren of translocating. Toepassing van deze methoden maakt het gemakkelijker om te onderscheiden fluorescently label peptide lokalisatie patronen. De protocollen die hier gepresenteerd kunnen worden gebruikt ter beoordeling van de lokalisatie van een verscheidenheid van membraan-actieve actoren dan versterkers, met inbegrip van cel-penetrating peptiden.
Een duidelijk voordeel van deze techniek is dat het biedt inzicht in het werkingsmechanisme van versterkers op eencellige niveau, die kan onthullen cel-naar-cel heterogeniteit15, in tegenstelling tot andere fluorescentie testen vaak gebruikt om te identificeren de mechanismen van werking van versterkers, waarmee alleen bulk schattingen9,22,23,24,25. Het gebruik van spheroplasts en protoplasten teneinde AMP cel invoeren is bijzonder nuttig26 , omdat ze meer fysiologisch relevante15 dan andere modellen die gebruikt worden voor de beoordeling van de cel invoeren, zoals lipide blaasjes24.
De protocollen die hier gepresenteerd maken het mogelijk voor onderzoekers sneller het verkrijgen van grotere omvang van de steekproeven van bacteriële beelden omdat de uitgebreide, bolvormige bacteriën veel gemakkelijker zijn te vinden, oriënteren en beeld. Deze verbeterde mogelijkheid om gegevens te verzamelen is waardevol in verschillende opzichten. Eerst, is hierdoor een meer systematische kwantitatieve analyse van peptide lokalisatie patronen.
