Hier presenteren we een stapsgewijze protocol om te onderzoeken de mitochondriale ademhaling en de glycolytic functie in Candida Albicans met behulp van een extra flux-analyzer.
Mitochondriën zijn essentiële organellen voor het cellulaire metabolisme en het voortbestaan. Een aantal belangrijke gebeurtenissen plaatsvinden in de mitochondriën, zoals cellulaire ademhaling, oxidatieve metabolisme, signaaltransductie en apoptosis. Bijgevolg, mitochondriale dysfunctie wordt gemeld dat een belangrijke rol spelen in de antischimmel drug tolerantie en de virulentie van pathogene schimmels. Recente gegevens hebben ook geleid tot de erkenning van het belang van het mitochondrium als een belangrijke bijdrage geleverd aan schimmel pathogenese. Ondanks het belang van de mitochondriën in de schimmel biologie, zijn gestandaardiseerde methoden om haar functie te begrijpen slecht ontwikkeld. Hier presenteren we een procedure om te bestuderen van de basale zuurstof verbruik tarief (OCR), een maatregel van de mitochondriale ademhaling en extracellulaire verzuring tarieven (ECAR), een maatregel van glycolytic functie bij C. albicans stammen. De hier beschreven methode kan worden toegepast op alle stammen Candidaspp. zonder de noodzaak voor het zuiveren van de mitochondriën van de intact schimmel cellen. Bovendien, dit protocol kan ook worden aangepast aan het scherm voor remmers van mitochondriale functie in C. albicans stammen.
Invasieve schimmelinfecties doden meer dan 1,5 miljoen mensen per jaar wereldwijd. Dit nummer is op de stijging te wijten aan een toename in het aantal mensen met gecompromitteerde immuniteit, met inbegrip van de bejaarden, premature zuigelingen, transplantatie ontvangers en kanker patiënten1. C. albicans is een opportunistische menselijke schimmel pathogeen die een deel uitmaakt van de menselijke darmflora. Hij bewoont ook mucosal oppervlakken en het maag-darmkanaal als een commensale organisme. C. albicans produceert ernstige systemische ziekte bij mensen met immuun tekortkomingen, die een operatie hebben ondergaan, of die zijn behandeld met lange cursussen van antibiotica. De Candida soorten behoren tot de top drie tot vier oorzaken van nosocomiale infectieziekten (NID) in mens2,3,4,5,6,7. Het jaarlijkse globale aantal Candida bloedbaan infecties wordt geschat op 400.000 ~ gevallen, met bijbehorende sterfte van 46-75%1. De jaarlijkse sterfte vanwege candidiasis is ongeveer 10.000 in de Verenigde Staten alleen. De omvang van NID veroorzaakt door schimmels wordt ook weerspiegeld in astronomische geduldige kosten5. In de Verenigde Staten overtreft de jaarlijkse kosten voor de behandeling van invasieve schimmelinfecties 2 miljard dollar, een enorme spanning toe te voegen aan de toch al overbelaste systeem van de gezondheidszorg. Momenteel zijn beschikbare standaard antischimmel therapieën beperkt vanwege toxiciteit, steeds meer voorkomende resistentie en interactie van de drug-drug. Daarom is er dringend behoefte aan nieuwe antischimmel drug targets die in betere behandelingsopties voor hoog-risico patiënten resulteren zal identificeren. Echter is de ontdekking van nieuwe drugs op schimmel doelstellingen ingewikkeld omdat schimmels, eukaryoten zijn. Dit beperkt aanzienlijk het aantal schimmel-specifieke drug targets.
Recente studies hebben aangegeven dat de mitochondriën zijn een essentiële bijdrage aan de schimmel virulentie en tolerantie tot antischimmel geneesmiddelen Aangezien mitochondriën belangrijk voor cellulaire ademhaling, oxidatieve metabolisme, signaaltransductie en apoptosis8 zijn ,9,10,11. Zowel de glycolytic als de niet-glycolytic metabolisme zijn essentieel voor het voortbestaan van C. albicans in de zoogdieren host12,13,14,15,16. Bovendien is verschillende C. albicans mutanten ontbreekt mitochondriale eiwitten, zoals Goa1, Srr1, Gem1, Sam37 etc. gebleken dat defect in filamentation, een belangrijke virulentiefactor van C. albicans17, 18 , 19 , 20 , 21 , 22. Bovendien deze mutanten waren ook aangetoond worden verzacht voor virulentie in een muismodel van verspreid candidiasis17,18,19,20,21 ,22. Dus, schimmel mitochondriën vertegenwoordigen een aantrekkelijk doelwit voor drugontdekking. Echter, de studie van mitochondriale functie in C. albicans is uitdagend omdat C. albicans petite negatieve23, wat betekent is dat het kan niet zonder het mitochondriaal genoom overleven.
Hier beschrijven we een protocol dat kan worden gebruikt voor het onderzoeken van mitochondriale en glycolytic functie in C. albicans zonder de noodzaak voor het zuiveren van de mitochondriën. Deze methode kan ook worden geoptimaliseerd voor het onderzoek naar het effect van de genetische manipulatie of chemische modulatoren op mitochondriale en glycolytic trajecten in C. albicans.
De Bioenergetica extra flux assay dient als een uitstekend middel om het voorlezen van de mitochondriale functie door het meten van de oxidatieve fosforylatie (OXPHOS)-afhankelijke zuurstofverbruik in real-time. Daarnaast kan ook een glycolytic functie die wordt gemeten als een percentage van de extracellulaire verzuring (estafetteovergave in de extracellulaire pH) tegelijkertijd in real-time analyse te worden onderzocht.
