En pålidelig og let reproducerbar metode til fremstilling af funktionaliserbare, nær-infrarøde udsender fotoluminescerende guld nanoclusters og deres direkte detektion inde HeLa celler ved flow cytometri og konfokal laser scanning mikroskopi er beskrevet.
I løbet af det seneste årti har fluorescerende guld nanoclusters (AuNCS) været vidne til stigende popularitet i biologiske applikationer og enorme indsats har været afsat til deres udvikling. I denne protokol er en nyligt udviklet, letkøbt metode til fremstilling af vandopløselige, biokompatible og i daglige stabile nær-infrarøde udleder AuNC’er blevet beskrevet i detaljer. Denne rum-temperatur, bottom-up kemisk syntese giver let funktionsdygtige AuNC’er udjævnet med thioctisk syre og thiol-modificeret polyethylenglycol i vandig opløsning. Den syntetiske tilgang kræver hverken organiske opløsningsmidler eller yderligere ligandudveksling eller omfattende viden om syntetisk kemi for at reproducere. De resulterende AuNC’er tilbyder gratis overfladecarboxylsyrer, som kan funktionaliseres med forskellige biologiske molekyler, der bærer en gratis amingruppe uden at påvirke AuNC’ernes fotoluminescerende egenskaber negativt. En hurtig, pålidelig procedure for flow cytometrisk kvantificering og konfokal mikroskopisk billeddannelse af AuNC optagelse af HeLa celler også blevet beskrevet. På grund af den store Stokes skift, korrekt indstilling af filtre i flow cytometri og konfokal mikroskopi er nødvendig for effektiv påvisning af nær-infrarød fotoluminescens af AuNC’er.
I det seneste årti har ultrasmå (≤ 2 nm) fotoluminescerende guldnanoklynger (PL AuNC’er) vist sig at være lovende sonder til både grundforskning og praktiske anvendelser1,2,3,4,5,6,7,8,9,10. Deres mange ønskelige egenskaber omfatter høj fotostabilitet, afstemmelige emissionsmaksima, lange emissionslevetider, store Stokes skift, lav toksicitet, god biokompatibilitet, nyreclearance og letkøbt biokonjugering. PL AuNC’er kan levere fotoluminescens fra det blå til det nær-infrarøde (NIR) spektrale område, afhængigt af antallet af atomer i klyngen11 og overfladens art ligand12. NIR (650-900 nm) udsender AuNC’er er særligt lovende for langsigtede in vitro og in vivo billeddannelse af celler og væv, da de tilbyder høj signal-støj-forhold på grund af minimal overlapning med iboende autofluorescens, svagere spredning og absorption, og høj væv penetration af NIR lys13,14.
I de senere år er der udviklet forskellige tilgange , der udnytter Au-S’s kovalente interaktioner til at forberede NIR-PL AuNCs udjævnet med en række thiolholdige ligander13,15,16,17. Til biomedicinske anvendelser skal AuNC’er funktionaliseres med en biologisk komponent for at lette bindende interaktioner. Således aunccs med høj kolloid stabilitet, der er let funktionsdygtige i vandigopløsningsmiddel er meget ønskeligt. Det overordnede mål med den nuværende protokol er at beskrive et tidligere rapporteret18 præparat af AuNC’er med en funktionaliserbar carboxylsyregruppe på overfladen ved at anvende thioctisk syre og polyethylenglycol (PEG) i et vandigt miljø og deres bøjning med molekyler, der bærer en primær amin efter syreaminkoblingsmetoden. På grund af den lette syntese og høje reproducerbarhed, kan denne protokol bruges og tilpasses af forskere fra ikke-kemiske baggrunde.
En af de vigtigste forudsætninger for anvendelse af AuNC’er i biomedicinsk forskning er evnen til at observere og måle AuNC’er inde i celler. Blandt de metoder til rådighed til at overvåge nanopartikel optagelse af celler, flow cytometri (FCM) og konfokal laser scanning mikroskopi (CLSM) tilbyder robuste, høj-throughput metoder, som giver mulighed for hurtige målinger af internalisering af fluorescerende nanomaterialer i stort antal celler19. Her er FCM- og CLSM-metoden til direkte måling og analyse af PL AuNC’er i celler, uden behov for yderligere farvestoffer, også blevet præsenteret.
NIR-udsender AuNC’er blev syntetiseret ved hjælp af en bottom-up-metode, hvor guldprækursoropløsningen (HAuCl4) blev behandlet med egnede thiolligands efterfulgt af en reduktion af Au3+. Reduktion af metalioner i vandig opløsning har tendens til at samle og resulterer i store nanopartikler i stedet for ultrasmå NCs21. For at forberede ultrasmå (≤2 nm) PL AuNC’er blev de syntetiske forhold justeret for at forhindre dannelse af store partikler og fremme dannelsen af ult…
