Een betrouwbare en gemakkelijk reproduceerbare methode voor de voorbereiding van functionalizable, nabij-infrarood uitzendende fotoluminescente goudnanoclusters en hun directe detectie in HeLa-cellen door stromingscytometrie en confocale laserscanningmicroscopie wordt beschreven.
In de afgelopen tien jaar, fluorescerende goud nanoclusters (AuNCs) zijn getuige van een groeiende populariteit in biologische toepassingen en enorme inspanningen zijn gewijd aan hun ontwikkeling. In dit protocol is een onlangs ontwikkelde, facile methode voor de bereiding van in water oplosbaar, biocompatibel en colloïdaally stabiel nabij-infrarood uitzendende AuNC’s in detail beschreven. Deze kamertemperatuur, bottom-up chemische synthese biedt gemakkelijk functionalizable AuNOCs bedekt met thioctisch zuur en thiol-gemodificeerde polyethyleenglycol in waterige oplossing. De synthetische aanpak vereist noch organische oplosmiddelen of extra ligand uitwisseling, noch uitgebreide kennis van synthetische chemie te reproduceren. De resulterende AuNOCs bieden vrije oppervlakte carboxylic zuren, die kunnen worden gefunctionaliseerd met verschillende biologische moleculen met een vrije amine groep zonder nadelige gevolgen voor de fotoluminescente eigenschappen van de AuNOCs. Een snelle, betrouwbare procedure voor flow cytometrische kwantificering en confocale microscopische beeldvorming van AuNC opname door HeLa cellen ook beschreven. Door de grote Stokes-verschuiving is een goede instelling van filters in stromingscytometrie en confocale microscopie noodzakelijk voor een efficiënte detectie van nabij-infrarood fotoluminescentie van AuNC’s.
In de afgelopen tien jaar zijn ultrakleine (≤ 2 nm) fotoluminescente goudnanoclusters (PL AuNC’s) naar voren gekomen als veelbelovende sondes voor zowel fundamenteel onderzoek als praktische toepassingen1,2,33,4,5,,67,8,8,9,10. Hun vele wenselijke kenmerken zijn hoge fotostabiliteit, tunable emissie maxima, lange emissie levensduur, grote Stokes verschuivingen, lage toxiciteit, goede biocompatibiliteit, nierklaring en facile bioconjugatie. PL AuNC’s kunnen fotoluminescentie van het blauwe naar het spectrale gebied (NEAR-infrared (NIR) leveren, afhankelijk van het aantal atomen binnen cluster11 en de aard van het oppervlak ligand12. NIR (650-900 nm) die AuNC’s uitzenden zijn bijzonder veelbelovend voor langdurige in vitro en in vivo beeldvorming van cellen en weefsels, omdat ze een hoge signaal-ruisverhouding bieden als gevolg van minimale overlap met intrinsieke autofluorescentie, zwakkere verstrooiing en absorptie, en hoge weefselpenetratie van NIR-licht13,14.
In de afgelopen jaren zijn verschillende benaderingen ontwikkeld die gebruik maken van covalente interacties van Au-S om NIR-PL AuNC’s voor te bereiden, afgetopt met een verscheidenheid aan thiolbevattende liganden13,15,16,17. Voor biomedische toepassingen moeten AuNC’s worden gefunctionaliseerd met een biologische component om bindende interacties te vergemakkelijken. AuNC’s met een hoge colloïdale stabiliteit die gemakkelijk te functionaliseren zijn in waterig oplosmiddel, zijn dus zeer wenselijk. Het algemene doel van het huidige protocol is om een eerder gerapporteerde18 preparaat van AuNOCs met een functionalizable carboxylic zuurgroep op het oppervlak te beschrijven door thioctisch zuur en polyethyleenglycol (PEG) in een waterige omgeving in detail te gebruiken en hun vervoegingsmoleculen met een primaire amine volgens de zuur-aminekoppelingsmethode. Vanwege het gemak van synthese en hoge reproduceerbaarheid, kan dit protocol worden gebruikt en aangepast door onderzoekers met een niet-chemische achtergrond.
Een van de belangrijkste vereisten voor toepassingen van AuNC’s in biomedisch onderzoek is de mogelijkheid om AuNC’s in cellen te observeren en te meten. Onder de beschikbare methoden om de opname van nanodeeltjes door cellen te monitoren, bieden flow cytometrie (FCM) en confocale laserscanmicroscopie (CLSM) robuuste, hoge doorvoermethoden die snelle metingen van internalisatie van fluorescerende nanomaterialen in een groot aantal cellen mogelijk maken19. Hier zijn ook FCM- en CLSM-methode voor directe meting en analyse van PL AuNC’s in cellen gepresenteerd, zonder dat extra kleurstoffen nodig zijn.
NIR-emitterende AuNO’s werden gesynthetiseerd met behulp van een bottom-up benadering waarbij de goudprecursoroplossing (HAuCl4)werd behandeld met geschikte thiolliganden, gevolgd door reductie van Au3+. Vermindering van metaalionen in waterige oplossing hebben de neiging om te aggregeren en resulteert in grote nanodeeltjes in plaats van ultrakleine NC’s21.
