En pålitelig og lett reproduserbar metode for fremstilling av funksjonelt, nær-infrarød emitterende fotoluminescerende gull nanoklynger og deres direkte deteksjon inne hela celler ved flyt cytometri og konfokal laser skanning mikroskopi er beskrevet.
I løpet av det siste tiåret har fluorescerende gullnanoklynger (AuNCs) vært vitne til økende popularitet i biologiske applikasjoner og enorm innsats har vært viet til deres utvikling. I denne protokollen har en nylig utviklet, facile metode for fremstilling av vannløselig, biokompatibel og kolloidalt stabil nær-infrarød emitterende AuNCs blitt beskrevet i detalj. Denne kjemiske syntesen for romtemperatur, nederen opp gir lett funksjonelt AuNCer avkortet med thioctic acid og tiolmodifisert polyetylenglykol i vandig løsning. Den syntetiske tilnærmingen krever verken organiske løsemidler eller ekstra ligandutveksling eller omfattende kunnskap om syntetisk kjemi for å reprodusere. De resulterende AuNCs tilbyr gratis overflate karboksylsyrer, som kan funksjonaliseres med ulike biologiske molekyler bærer en fri amingruppe uten å påvirke de fotoluminescerende egenskapene til AuNCs negativt. En rask, pålitelig prosedyre for flyt cytometrisk kvantifisering og konfokal mikroskopisk avbildning av AuNC opptak av HeLa celler også blitt beskrevet. På grunn av det store Stokes-skiftet er riktig innstilling av filtre i flow cytometri og konfokal mikroskopi nødvendig for effektiv påvisning av nær-infrarød fotoluminescens av AuNCs.
I det siste tiåret har ultrasmå (≤ 2 nm) fotoluminescerende gull nanoklynger (PL AuNCs) dukket opp som lovende sonder for både grunnleggende forskning og praktiske applikasjoner1,2,33,4,5,6,7,8,9,10. Deres mange ønskelige egenskaper inkluderer høy fotostabilitet, tunable utslipp maxima, lang utslippslevetid, store Stokes skift, lav toksisitet, god biokompatibilitet, renal clearance og facile biokonjugering. PL AuNCs kan gi fotoluminescens fra blått til nær-infrarød (NIR) spektral regionen, avhengig av antall atomer i klyngen11 og arten av overflaten ligand12. NIR (650-900 nm) som sender ut AuNCs er spesielt lovende for langsiktig in vitro og in vivo avbildning av celler og vev, da de tilbyr høyt signal-til-støy-forhold på grunn av minimum overlapping med egenautofluorescens, svakere spredning og absorpsjon, og høy vevspenetrasjon av NIR-lys13,14.
I de senere årene har ulike tilnærminger som drar nytte av Au-S kovalente interaksjoner blitt utviklet for å forberede NIR-PL AuNCs avkortet med en rekke thiol-holdige ligander13,,15,16,17. For biomedisinske applikasjoner må AuNCs funksjonaliseres med en biologisk komponent for å lette bindingsinteraksjoner. Dermed er AuNCs med høy kolloidal stabilitet som lett kan funksjoneliseres i vandig løsningsmiddel svært ønskelig. Det overordnede målet med den nåværende protokollen er å beskrive en tidligere rapportert18 forberedelse av AuNCs med en funksjonelt karboksylsyregruppe på overflaten ved å bruke tioctic acid og polyetylenglykol (PEG) i et vandig miljø i detalj og deres bøyning med molekyler som bærer en primær amin etter syre-aminkoblingsmetoden. På grunn av den enkle syntesen og høy reproduserbarhet, kan denne protokollen brukes og tilpasses av forskere fra ikke-kjemi bakgrunn.
En av de viktigste forutsetningene for anvendelser av AuNCs i biomedisinsk forskning er evnen til å observere og måle AuNCs inne i celler. Blant metodene som er tilgjengelige for å overvåke nanopartikkelopptak av celler, flow cytometri (FCM) og konfokal laserskanning mikroskopi (CLSM) tilbyr robuste, høy gjennomstrømning metoder som tillater rask måling av internalisering av fluorescerende nanomaterialer i stort antall celler19. Her har FCM og CLSM-metoden for direkte måling og analyse av PL AuNCs inne i celler, uten behov for ekstra fargestoffer, også blitt presentert.
NIR-emitterende AuNCer ble syntetisert ved hjelp av en nedenfra-opp-tilnærming der gullforløperløsningen (HAuCl4) ble behandlet med egnede tiolligander, etterfulgt av reduksjon av Au3+. Reduksjon av metallioner i vandig løsning har en tendens til å aggregere og resulterer i store nanopartikler i stedet for ultrasmå NCer21.
