We hebben gesynthetiseerd een nieuwe analoog van pancratistatin met vergelijkbare anti-kanker activiteit als native pancratistatin, interessant, combinatorische behandeling met tamoxifen leverde een drastische verbetering in de apoptotische en autofagocytische inductie door mitochondriale richten met minimale effecten op goedaardige fibroblasten. Zo kon JCTH-4 in combinatie met tamoxifen zorgen voor een veilige anti-kanker therapie.
Borstkanker is een van de meest voorkomende vormen van kanker onder vrouwen in Noord-Amerika. Vele huidige antikankerbehandelingen, zoals ioniserende straling, apoptose via DNA-schade. Helaas is een dergelijke behandeling niet-selectief voor kankercellen en produceren soortgelijke toxiciteit in normale cellen. Wij hebben gemeld selectieve inductie van apoptose in kankercellen door de natuurlijke verbinding pancratistatin (PST). Onlangs is een nieuwe PST analoog een C-1 acetoxymethyl derivaat van 7 deoxypancratistatin (JCTH-4), die door de novo synthese en vertoont vergelijkbare selectieve apoptose inducerende activiteit in verscheidene cellijnen. Onlangs heeft autofagie betrokken bij maligniteiten als zowel pro-overleving en pro-dood mechanismen in reactie op chemotherapie. Tamoxifen (TAM) heeft altijd aangetoond inductie van pro-survival autofagie in tal van vormen van kanker. In deze studie werd de effectiviteit van JCTH-4 alleen en in combinatie met TAM om celdood te induceren in de moedermelk Cancer (MCF7) en neuroblastoom (SH-SY5Y) cellen werd geëvalueerd. TAM alleen autofagie veroorzaakt, maar onbeduidend celdood terwijl JCTH-4 alleen al veroorzaakt significante inductie van apoptose met een aantal inductie van autofagie. Interessant is dat de combinatorische behandeling leverde een drastische toename van apoptotische en autofagocytische inductie. We tijdsafhankelijke morfologische veranderingen in MCF7 cellen die door TAM-geïnduceerde autofagie, JCTH-4-geïnduceerde apoptose en autofagie, en versnelde celdood bewaakt met behulp van combinatorische behandeling met behulp van time-lapse microscopie. We hebben aangetoond dat deze verbindingen apoptose / autofagie induceren door mitochondriale targeting in deze kankercellen. Belangrijker is dat deze behandelingen geen invloed op de overleving van goedaardige menselijke fibroblasten. Zo geven deze resultaten aan dat JCTH-4 in combinatie met TAM kunnen worden gebruikt als een veilig en zeer krachtige anti kanker therapie tegen borstkanker en neuroblastoomcellen.
PST en soortgelijke verbindingen is aangetoond om anti-kanker eigenschappen 11-15,21. We hebben eerder gemeld natuurlijke PST om selectief te destabiliseren de mitochondriën in kankercellen, die veroorzaakt daarbij apoptose door het vrijkomen van apoptotische factoren 12,14. Het is zeer waarschijnlijk dat JCTH-4 werkt via hetzelfde werkingsmechanisme heeft; JCTH-4 veroorzaakt MMP ineenstorting van de MCF7 cellen gezien met TMRM kleuring (afb. 5a), en een verhoogde productie van RO…
