Dit protocol richt zich op gebruik te maken van de inherente vermogen van stamcellen om cue te nemen van hun omringende extracellulaire matrix en worden aangezet om te differentieren naar verschillende fenotypes. Deze methode manuscript strekt de beschrijving en karakterisering van een model met behulp van een dubbellagige hydrogel, bestaande uit PEG-fibrine en collageen, gelijktijdig gelijktijdig onderscheid vet-stamcellen<sup> 1</sup>.
Natuurlijke polymeren door de jaren heen hebben meer belang, omdat van hun gastheer biocompatibiliteit en het vermogen om samen te werken met cellen in vitro en in vivo. Een gebied van onderzoek die belofte houdt in de regeneratieve geneeskunde is de combinatorische gebruik van nieuwe biomaterialen en stamcellen. Een fundamentele strategie op het gebied van tissue engineering is het gebruik van drie-dimensionale steiger (bijvoorbeeld gedecellulariseerde extracellulaire matrix, hydrogels, micro / nano deeltjes) voor de aansturing van de celfunctie. Deze technologie heeft zich ontwikkeld uit de ontdekking dat cellen een substraat waarop ze kunnen zich nodig hebben, vermenigvuldigen, en uiten hun gedifferentieerde cellulaire fenotype en functie 2-3. Meer recent heeft ook vastgesteld dat de cellen niet alleen deze substraten te gebruiken voor de naleving, maar ook interactie en neem signalen uit de matrix substraat (bijvoorbeeld, extracellulaire matrix, ECM) 4. Daarom werden de cellen en steigers een wederzijdse verbinding diedient om te controleren weefsel ontwikkeling, organisatie, en de uiteindelijke functie. Vet-afgeleide stamcellen (ASC) mesenchymale zijn, niet-hematopoëtische stamcellen aanwezig in vetweefsel dat multi-lijn differentiatie kan vertonen en dienen als een gemakkelijk toegankelijke bron van cellen (dat wil zeggen pre-vasculaire endotheel en pericyten). Onze hypothese is dat het vet-afgeleide stamcellen kunnen worden gericht op verschillende fenotypes tegelijk door gewoon co-kweken ze in dubbellagige matrices 1. Ons laboratorium is gericht op dermale wondgenezing. Daartoe hebben we een samengestelde matrix van natuurlijke biomaterialen fibrine, collageen en chitosan kunnen nabootsen die de eigenschappen en functies van de dermale specifieke wondgenezing ECM omgeving.
ASC's zijn bekend om hun gemak van isolatie en het vermogen om onderscheid te maken in de richting van verschillende celtypes. Met de technieken beschreven in dit manuscript zijn wij in staat om de plasticiteit van ASC te benutten door het blootstellen van deze cellen om meerdere biomatrices tegelijk.
