Microbiële eukaryoten zijn zowel een bron van fotosynthetisch afgeleide koolstof en top roofzuchtige soort in permanent met ijs bedekte Antarctische meren. Dit rapport beschrijft een verrijking cultuur benadering van het metabool veelzijdige microbiële eukaryoten isoleren van de Antarctische meer, Lake Bonney, en beoordeelt anorganische koolstof fixatie mogelijkheden met behulp van een radio-isotoop test voor ribulose-1 ,5-bisphophate carboxylase oxygenase (RuBisCO) activiteit.
Lake Bonney is een van de vele permanent met ijs bedekte meren gelegen in het McMurdo droge valleien op Antarctica. De eeuwige ijslaag zorgt voor een chemisch gelaagde waterkolom en in tegenstelling tot andere binnenlandse wateren, grotendeels voorkomt externe input van koolstof en voedingsstoffen uit stromen. Biota worden blootgesteld aan tal van milieu-invloeden, waaronder het hele jaar door ernstige gebrek aan voedingsstoffen, lage temperaturen, extreme schaduw, hypersalinity, en 24-uurs duisternis in de winter 1. Deze extreme omstandigheden beperken de fauna en flora in Lake Bonney bijna uitsluitend voor micro-organismen 2.
Eencellige micro-organismen eukaryoten (de zogenaamde "protisten") zijn belangrijke spelers in de wereldwijde biochemische cycli 3 en spelen een belangrijke ecologische rol in de cyclus van koolstof in de droge vallei meren, waarmee het zowel de primaire en tertiaire rollen in het aquatisch voedselweb. In de droge vallei in het water levende voedselweb, protisten dat ik dat oplossennorganic koolstof (autotrophy) zijn de belangrijkste producenten van organische koolstof voor organotrofe organismen 4, 2. Phagotrophic of heterotrofe protisten staat inname van bacteriën en kleinere protisten optreden als toppredatoren in de voedselketen 5. Last, een onbekend deel van de protisten bevolking in staat is om gecombineerde mixotrophic metabolisme 6, 7. Mixotrophy in protisten betreft het vermogen om fotosynthetische capaciteit combineren met phagotrophic inname van prooi micro-organismen. Deze vorm van mixotrophy verschilt van mixotrophic metabolisme in soorten bacteriën, die over het algemeen gaat de opname opgeloste koolstof moleculen. Er zijn op dit moment zeer weinig protisten isolaten van permanent ijs bedekte polaire meren, en studies van de protisten diversiteit en ecologie in deze extreme omgeving zijn beperkt 8, 4, 9, 10, 5. Een beter begrip van protisten metabolische veelzijdigheid in het eenvoudige droge vallei meer voedselweb zal helpen bij de ontwikkeling van modellen voor de role van protisten in de mondiale koolstofcyclus.
We gebruikten een verrijking cultuur benadering van potentieel fototrofe en mixotrophic protisten isoleren van Lake Bonney. Sampling diepten in de waterkolom werden gekozen op basis van de locatie van de primaire productie maxima en protisten fylogenetische diversiteit 4, 11, en variabiliteit in de grote abiotische factoren die van invloed protisten trofische modi: ondiepe bemonstering diepten zijn beperkt voor belangrijke voedingsstoffen, terwijl de diepere bemonstering diepten beperkt door de beschikbaarheid van licht. Bovendien zijn meer watermonsters aangevuld met verschillende soorten kweekmedia om de groei van verschillende fototrofe organismen.
RuBisCO katalyseert de snelheidsbepalende stap in de Calvin Benson Bassham (CBB) cyclus, de belangrijkste route door die autotrofe organismen vast anorganische koolstof en zorgen voor organische koolstof voor hogere trofische niveaus in aquatische en terrestrische voedselwebben 12. In deze studie we toegepast een radio-isotoop test aangepast voor gefilterde monsters 13 tot en met maximale carboxylase-activiteit te controleren als een proxy voor koolstof fixatie potentieel en metabolische veelzijdigheid in het Lake Bonney verrijking culturen.
De recente moleculaire studies hebben gerapporteerd grote diversiteit van eencellige eukaryoten een heel scala van omgevingen 3, 19, 20, maar als gevolg van een gebrek aan isolaten over de volle breedte van protisten habitats de functionele rol van deze individuele soorten in voedselwebben zijn grotendeels onbekend.
