The term anastasis refers to the phenomenon in which dying cells reverse a cell suicide process at a late stage, repair themselves, and ultimately survive. Here we demonstrate protocols for detecting and tracking cells that undergo anastasis.
Anastasis (Grieks voor "oplopend tot leven") verwijst naar het herstel van stervende cellen. Voordat deze cellen terug te krijgen, hebben ze gepasseerd belangrijke checkpoints van apoptose, waaronder mitochondriale fragmentatie, release van mitochondriale cytochroom c in het cytosol, de activering van caspases, chromatinecondensatie, DNA-schade, kernfragmentatie, plasmamembraan blebbing, celinkrimping, blootstelling celoppervlak fosfatidylserine en vorming van apoptotische lichamen. Anastasis kan optreden wanneer apoptotische stimuli voor het overlijden worden verwijderd, waardoor stervende cellen apoptose en mogelijk andere dood mechanismen te keren. Daarom lijkt Anastasis om fysiologische genezingsprocessen dat ook zou kunnen ondersteunen beschadigde cellen op ongepaste wijze te betrekken. De functies en mechanismen van Anastasis zijn nog onduidelijk, gehinderd gedeeltelijk door de beperkte hulpmiddelen voor het opsporen van gebeurtenissen uit het verleden na het herstel van de ogenschijnlijk gezonde cellen. Strategieën om Anasta detecterensis zal onderzoeken van de fysiologische mechanismen, de gevaren van ondoden cellen bij de ziekte van pathologie, en mogelijke therapieën mogelijk te Anastasis moduleren. We beschrijven hier effectieve strategieën met levende cel microscopie en zoogdieren caspase biosensor voor het identificeren en volgen anastasis in zoogdiercellen.
Apoptose (Grieks voor "vallen tot de dood") wordt over het algemeen aangenomen dat het een one-way proces eindigt in cel zelfmoord 1-7 zijn. Genetische verstoring van pro-dood-genen resulteert in de overleving van extra cellen die anders zouden sterven in gehele dieren, waaronder cellen die reeds de apoptose pathway 8,9 hebben geopend. Ook genetische manipulaties laat gezonde cellen van zoogdieren die op kunstmatige wijze weer te geven "eet me" signalen of die hechting verliezen hun extracellulaire matrix tot de dood door hele cellen fagocytose of entosis respectievelijk 10,11 ontsnappen. Echter, wij en anderen hebben aangetoond dat zonder genetische manipulatie normale gezonde zoogdiercellen en cellijnen kunnen herstellen van de vroege stadia van apoptose 12-15. Gebruik van tools om individuele cellen te volgen, hebben we verder aangetoond herstel van late stadia van apoptose 12,13, na cellen belangrijke checkpoints die typisch zijn verstrekenly markeren de "point of no return" 2-6. Deze checkpoints van laat stadium apoptose omvatten mitochondriale release van cytochroom c, activering van caspases, nucleaire fragmentatie, en de vorming van apoptotische lichamen. Wij keurden een Grieks samengesteld woord "Anastasis", wat betekent "oplopend tot leven", om deze omkering van apoptose beschrijven aan de rand van celdood 2-6.
Tenzij de gehele stervenden-herstelproces wordt waargenomen door live cell imaging, is het een uitdaging om cellen die Anastasis uit cellen die nooit meegemaakt apoptotische gebeurtenissen hebben ondergaan onderscheiden. Decennia van werk is gebleken dat de morfologische kenmerken van de cel zelfmoord door apoptose worden gedreven door evolutionair geconserveerde biochemische en moleculaire gebeurtenissen 16-19. Deze gebeurtenissen bevorderen zelfvernietiging van cellen om ontwikkelings- en homoeostatic processen in eencellige en meercellige organismen te reguleren door het elimineren van beschadigde of dangerous cellen 16-19. Terwijl apoptotische cellen gemakkelijk kunnen worden onderscheiden door standaard morfologische, biochemische en moleculaire manifestaties van apoptose 1,5,6,16,20, momenteel is er geen bekende marker specifiek Anastasis 12,13. Belangrijk cellen die anastasis hebben ondergaan lijken normale gezonde cellen, en cellen die net beginnen omkeren apoptose verschijnen apoptotische stervende cellen 12,13 zijn. Zo worden nieuwe instrumenten nodig zijn om met zekerheid dat een bepaalde overlevende cel eerder had meegemaakt actieve apoptotische processen.
Apoptose wordt algemeen aangenomen als een onomkeerbare cascade omdat het een snelle en massale vernietigingsproces. Hoewel het enkele minuten zou kunnen nemen om voor sommige cellen om apoptose te starten, zodra mitochondriën apoptotische factoren zoals cytochroom c in het cytosol 21,22 hebben vrijgegeven, kan caspasen worden geactiveerd binnen 5 minuten 23,24, gevolgd door cytoplasmatische ennucleaire condensatie binnen 10 min 25-27, en celdood kort daarna 25-27. Geactiveerde caspasen orkestreren apoptose door splitsing en inactiveren belangrijke structurele en functionele componenten ten behoeve van cellulaire afbraak 2,28, zoals endonuclease inhibitor DFF45 / ICAD 29,30. Caspasen activeert ook pro-apoptotische factoren, zoals Bcl-2 familielid BID, die translokeert mitochondriën mitochondriale afgifte van cytochroom c 31,32 bevorderen. Caspaseactiviteit resulteert ook in celoppervlak blootstelling van fosfatidylserine als een "eet me" signaal voor het bevorderen van afblazen van stervende cellen door macrofagen of buur cellen via fagocytose 33. Bovendien apoptotische gebeurtenissen maken mitochondriën disfunctioneel, verstoren cellulaire bio-energetica en metabolisme 34,35,36. Zo, het herstel van de vernietiging lijkt intuïtief onwaarschijnlijk.
Anders originele verwachtenties, kunnen cellen de apoptotische celdood proces om te keren, zelfs in een laat stadium. Door het continu bewaken van het lot van de stervende cellen in cultuur, zagen we de omkeerbaarheid van laat stadium apoptose in een reeks van primaire cellen en cellijnen 12,13. Verwijdering van de dood stimulans toegestaan herstel van de openlijke kenmerken van apoptose, zoals mitochondriale fragmentatie, chromatinecondensatie, DNA-schade, plasmamembraan blebbing, celoppervlak blootstelling van fosfatidylserine, vrijkomen van mitochondriale cytochroom c, caspaseactivering, kernfragmentatie, cel krimp, en de vorming van apoptotische lichamen. Deze waarnemingen verhogen onbeantwoorde vragen over de functies, gevolgen en mechanismen van Anastasis. Om deze vragen te beantwoorden, een voorwaarde is om betrouwbaar cellen die Anastasis hebben ondergaan identificeren. Hier beschrijven we live microscopie methoden en een caspase biosensor voor de detectie van cellen die eerder omgekeerd laat stadium apoptose en then overleefde.
Anastasis verwijst naar het verschijnsel waarbij cellen die celdood route geactiveerd vervolgens omgekeerde stervensproces en overleven. Hier hebben we aangetoond dat live cell imaging kan worden gebruikt om te bevestigen dat het dezelfde individuele cellen in feite apoptotische celdood proces om te keren in een laat stadium, en dan verder overleven en reproduceren.
