Her præsenterer vi en protokol til at injicere ultralyd mikrobobler kontrastmidler i levende, isolerede sene drægtighedsstalde stadie murine embryoer. Denne metode gør det muligt at studiet af perfusion parametre og vaskulære molekylære markører i embryo ved hjælp af kontrast-forstærket højfrekvente ultralydsscanning.
Ultrasound contrast-enhanced imaging can convey essential quantitative information regarding tissue vascularity and perfusion and, in targeted applications, facilitate the detection and measure of vascular biomarkers at the molecular level. Within the mouse embryo, this noninvasive technique may be used to uncover basic mechanisms underlying vascular development in the early mouse circulatory system and in genetic models of cardiovascular disease. The mouse embryo also presents as an excellent model for studying the adhesion of microbubbles to angiogenic targets (including vascular endothelial growth factor receptor 2 (VEGFR2) or αvβ3) and for assessing the quantitative nature of molecular ultrasound. We therefore developed a method to introduce ultrasound contrast agents into the vasculature of living, isolated embryos. This allows freedom in terms of injection control and positioning, reproducibility of the imaging plane without obstruction and motion, and simplified image analysis and quantification. Late gestational stage (embryonic day (E)16.6 and E17.5) murine embryos were isolated from the uterus, gently exteriorized from the yolk sac and microbubble contrast agents were injected into veins accessible on the chorionic surface of the placental disc. Nonlinear contrast ultrasound imaging was then employed to collect a number of basic perfusion parameters (peak enhancement, wash-in rate and time to peak) and quantify targeted microbubble binding in an endoglin mouse model. We show the successful circulation of microbubbles within living embryos and the utility of this approach in characterizing embryonic vasculature and microbubble behavior.
Kontrast-forstærket ultralydsscanning gør brug af mikrobobler kontrastmidler at visualisere og karakterisere det vaskulære miljø. Disse midler gør det muligt invasiv vurdering af mikrocirkulationen, vaskularisering og kardiovaskulær funktion. Desuden kan ændring af boblen overflade resultere i målrettet mikroboble binding til endotelceller biomarkører, som vist i prækliniske anvendelser af angiogenese, aterosklerose og inflammation 1,2 gør molekylær ultralydsscanning af vaskulære hændelser mulige. Kontrastforbedret ultralyd kan derfor anvendes til at identificere de komplekse og forskelligartede miljøer, der påvirker sunde og syge vaskulære tilstande 3-5.
I de seneste mange år, har interessen for nytten af mikrobobler imaging udvides til alsidig museembryo model. Som model for udvikling pattedyr, indførelse af mikrobobler i den embryoniske vaskulatur forbedrer fysiologiskundersøgelse af udviklingen kredsløbssygdomme (f.eks blodgennemstrømning, cardiac output) og i tilfælde af transgene og målrettede mutante musemodeller for hjertesygdom 6,7, kan give indsigt i, hvordan genetiske faktorer ændrer kardiovaskulær funktion. Faktisk kvantitativ og kvalitativ 2D analyser af embryonale hjernen vaskulaturen er allerede nået 8. Endvidere musen embryo præsenterer som en fremragende model for behandlingen af bindingen af målrettede mikrobobler til vaskulære markører in vivo. Bartelle et al. 9, for eksempel, har indført avidin mikrobobler i embryo hjerte- hjertekamrene til at vurdere målrettet bindende i Biotag-Bira transgene embryoner og undersøge vaskulær anatomi. Frembringelsen af heterozygote og homozygote musemodeller kan også bruges som et surrogat for tumor modelstudier til formål at definere kvantitative karakter af molekylær ultralyd – en vigtig benchmark i at omsætte denne teknik til klinikken.
<p class = "jove_content"> mikrobobler oftest introduceret til embryonale omløb via intrakardielle injektioner i enkelt embryoner udsat gennem en laparotomi 8-10. i livmoderen injektioner står imidlertid over for en række udfordringer. Disse omfatter injektion vejledning, at det er nødvendigt at imødegå bevægelse i mor og eksterioriseret embryo, vedligeholdelse af hæmodynamisk levedygtighed i moderen og blotlagt embryoner, behandling af langsigtede virkninger af anæstesi og komplikationer på grund af blødning 11. Derfor er målet med undersøgelsen var at udvikle en teknik til at injicere mikrobobler i isolerede levende sene embryoner 12. Denne indstilling giver mere frihed i form af indsprøjtning kontrol og positionering, reproducerbarhed afbildningsplanet uden obstruktion, og forenklet billedanalyse og kvantificering.I den foreliggende undersøgelse har vi skitsere en hidtil ukendt fremgangsmåde til injektion af mikrobobler i levende murine embryoer for med henblik på at studere mikroboble kinetisk adfærd og for at studere målrettet mikroboble binding til endogene endotheloverflade markører. Ikke-lineær kontrast specifik ultralydsscanning bruges til at måle på en række grundlæggende perfusion parametre, herunder peak ekstraudstyr (PE), vask-i sats og tid til peak (TTP) i isolerede E17.5 embryoer. Vi viser også gyldigheden af embryo model til vurdering af kvantitativ natur molekylær ultralyd i en embryonisk endoglin tab af funktion transgen musemodel, hvor endoglin er et klinisk relevant mål på grund af sin høje ekspression i vaskulære endotelceller på steder med aktiv angiogenese 13 . Adhæsionen af endoglin målrettet (MB E), rotte isotype IgG 2 kontrol (MB C) og ikke-målrettede (MB U) mikrobobler evalueres i heterozygot endoglin (Eng +/-) og homozygot endoglin (Eng + / +), der udtrykker embryoner. Analyse af det målrettede binding afslører, at molekylær ultralyd er i stand til at skelne mellem endoglin genotyper og vedrører receptor tætheder til kvantificerbare molekylære ultralyd niveauer.
