Här presenterar vi ett protokoll för att injicera ultraljudsmikrobubblor kontrastmedel i levande, isolerade sena dräktighetsstadiet murina embryon. Denna metod gör det möjligt att studera perfusion parametrar och kärl molekylära markörer inom embryot använder kontrastförstärkt högfrekvent ultraljud.
Ultrasound contrast-enhanced imaging can convey essential quantitative information regarding tissue vascularity and perfusion and, in targeted applications, facilitate the detection and measure of vascular biomarkers at the molecular level. Within the mouse embryo, this noninvasive technique may be used to uncover basic mechanisms underlying vascular development in the early mouse circulatory system and in genetic models of cardiovascular disease. The mouse embryo also presents as an excellent model for studying the adhesion of microbubbles to angiogenic targets (including vascular endothelial growth factor receptor 2 (VEGFR2) or αvβ3) and for assessing the quantitative nature of molecular ultrasound. We therefore developed a method to introduce ultrasound contrast agents into the vasculature of living, isolated embryos. This allows freedom in terms of injection control and positioning, reproducibility of the imaging plane without obstruction and motion, and simplified image analysis and quantification. Late gestational stage (embryonic day (E)16.6 and E17.5) murine embryos were isolated from the uterus, gently exteriorized from the yolk sac and microbubble contrast agents were injected into veins accessible on the chorionic surface of the placental disc. Nonlinear contrast ultrasound imaging was then employed to collect a number of basic perfusion parameters (peak enhancement, wash-in rate and time to peak) and quantify targeted microbubble binding in an endoglin mouse model. We show the successful circulation of microbubbles within living embryos and the utility of this approach in characterizing embryonic vasculature and microbubble behavior.
Kontrastförstärkt ultraljud använder mikrobubblor kontrastmedel för att visualisera och karakterisera vaskulära miljön. Dessa medel möjliggör icke-invasiv bedömning av mikrocirkulationen, kärlteckning och kardiovaskulär funktion. Dessutom kan modifiering av bubblan ytan resultera i målinriktad mikrobubblor bindning till endotelceller biomarkörer, som visats i prekliniska applikationer av angiogenes, åderförkalkning och inflammation 1,2 gör molekylär ultraljud av vaskulära händelser möjliga. Kontrastförstärkt ultraljud kan därför användas för att identifiera de komplexa och skiftande miljöer som påverkar friska och sjuka kärltillstånd 3-5.
Under det senaste antal år har intresset för nyttan av mikrobubblor avbildning utvidgas till den mångsidiga musembryo modell. Som en modell för utveckling däggdjur, förbättrar fysiologisk införandet av mikrobubblor i embryonala kärlstudie av utvecklingscirkulationssystemet (t.ex. blodflöde, hjärtminutvolym) och i fall av transgena och riktade muterade musmodeller av hjärtsjukdom 6,7, kan ge insikter i hur genetiska faktorer förändrar kardiovaskulär funktion. I själva verket, analyserar kvantitativ och kvalitativ 2D av embryonala hjärnkärl har redan uppnåtts 8. Dessutom presenterar musen embryot som en utmärkt modell för att undersöka bindningen av riktade mikrobubblor till vaskulära markörer in vivo. Et al. Bartelle 9, till exempel, har infört avidin mikrobubblor i embryohjärt ventriklarna att bedöma genom riktad bindning i Biotag-BirA transgena embryon och undersöka kärlanatomi. Den generation av heterozygota och homozygota musmodeller kan också användas som ett surrogat för tumörmodellstudier som syftar till att definiera kvantitativa natur molekylär ultraljud – ett viktigt riktmärke i att översätta denna teknik till kliniken.
<p class = "jove_content"> Mikrobubblor oftast introduceras till den embryonala cirkulationen via Intrakardiella injektioner i enstaka embryon exponeras genom en laparotomi 8-10. I livmodern injektioner, dock står inför ett antal utmaningar. Dessa inkluderar injektion vägledning, behovet av att motverka rörelse i modern och exterioriserad embryo, underhåll av hemodynamisk livskraft hos modern och exterioriserades embryon, adresse långsiktiga effekter av narkos och komplikationer på grund av blödning 11. Därför var målet med undersökningen att utveckla en teknik för att injicera mikrobubblor i isolerade levande slutskedet embryon 12. Detta alternativ ger större frihet i fråga om insprutningsstyrning och positionering, reproducerbarhet av avbildningsplanet utan hinder, och förenklad bildanalys och kvantifiering.I den aktuella studien, vi beskriva ett nytt förfarande för injektion av mikrobubblor i levande murina embryon for att studera mikrobubblor kinetiska beteende och studera riktade mikrobubblor bindning till endogena endotelceller ytmarkörer. Icke-linjär kontrastspecifika ultraljud används för att mäta om ett antal grundläggande perfusion parametrar inklusive toppförstärkning (PE), tvätta-in hastighet och tid till maximal (TTP) i isolerade E17.5 embryon. Vi visar också giltigheten av embryot modell för bedömning av kvantitativ karaktär molekylär ultraljud i en embryonal endoglin funktionsförlust transgen musmodell där endoglin är en kliniskt relevant mål på grund av dess höga uttryck i vaskulära endotelceller på ställen där aktiv angiogenes 13 . Vidhäftningen av endoglin inriktade (MB E), råtta isotypkontroll IgG 2 kontroll (MB C) och oriktade (MB U) mikrobubblor utvärderas i heterozygot endoglin (Eng +/-) och homozygot endoglin (Eng + / +) uttrycker embryon. Analys av den målinriktade binding avslöjar att molekylär ultraljud är kapabel att skilja mellan endoglin genotyper och avser receptortäthet till kvantifierbara molekylära ultraljudsnivåer.
Ultraljudskontrastmedel injicerades i slutskedet dräktighet musembryon och olinjära kontrastbilder förvärvades för att mäta perfusion parametrar och riktade mikrobubblor bindande.
