Jonkanaler uttrycks i tubulär epitel spelar en viktig roll i patologin av polycystisk njursjukdom. Här beskriver vi experimentella protokoll som används för att utföra patch-clamp analys och intracellulär kalcium nivå mätningar i cystisk epitel nyisolerade från gnagare njurar.
Cysta initiering och expansion under polycystisk njursjukdom är en komplex process som kännetecknas av avvikelser i tubulär celltillväxt, luminal vätskeansamling och extracellulär matrisbildning. Aktivitet av jonkanaler och intracellulärt kalcium signalering är viktiga fysiologiska parametrar som bestämmer funktioner av tubulärt epitel. Vi utvecklade en metod som lämpar sig för realtidsobservation av jonkanaler aktivitet med patch-clamp-tekniken och registrering av intracellulär Ca2 + nivå i epiteliala monoskikt nyisolerade från njurcystor. PCK råttor, en genetisk modell för autosomal recessiv polycystisk njursjukdom (ARPKD), användes här för ex vivo analys av jonkanaler och kalciumflöde. Beskrivs här är en detaljerad steg-för-steg förfarande för att isolera cystisk monolager och icke-vidgade tubuli från PCK eller normala Sprague Dawley (SD) råttor, och övervaka enda kanal aktivitet och intracellulära Ca2 + dynamik.Den här metoden kräver inte enzymatisk bearbetning och möjliggör analys i en infödd miljö av nyligen isolerade epitelial monolager. Dessutom är denna teknik mycket känslig för intracellulära kalciumförändringar och genererar bilder med hög upplösning för exakta mätningar. Slutligen kan isolerade cystisk epitel användas vidare för färgning med antikroppar eller färgämnen, beredning av primärkulturer och rening av olika biokemiska analyser.
Jonkanaler spelar en viktig roll i många fysiologiska funktioner, inklusive celltillväxt och differentiering. Autosomalt dominant och recessiva polycystiska njursjukdomar (ADPKD och ARPKD, respektive) är genetiska sjukdomar som kännetecknas av utveckling av njurvätskefyllda cystor av den rörformiga epitelceller ursprung. ADPKD orsakas av mutationer i PKD1 eller PKD2 gener som kodar polycystins 1 och 2, membranproteiner är involverade i regleringen av celltillväxt och differentiering. PKD2 i sig eller som ett komplex med PKD1 fungerar också som en Ca2 + -permeable ning kanal 1. Mutationer av PKHD1 genen som kodar fibrocystin (en cilier associerad receptorliknande protein involverat i tubulogenesis och / eller underhåll av polariteten hos epitel) är den genetiska impulser av ARPKD 2. Cysta tillväxt ett komplext fenomen tillsammans med störd spridning 3,4, angiogenes 5, dedifferentiering och förlust av poläraten av tubulära celler 6-8.
Defekt reabsorption och förstärkt sekretion i cystisk epitel bidra till vätskeansamling i lumen och cysta expansions 9,10. Nedsatt flödesberoende [Ca2 +] i signaleringen har också kopplat till cystogenesis under PKD 11-15.
Här beskriver vi ett förfarande som är lämpligt för patch-clamp mätningar av enstaka kanalaktivitet och intracellulära Ca2 + nivåer i cystisk epiteliala monoskikt isolerade från PCK råttor. Denna metod har framgångsrikt tillämpats av oss för att karakterisera aktiviteten hos epitelial Na * kanal (ENAC) 10 och [Ca2 +] i -beroende processer som induceras av Ca2 + -permeable TRPV4 och Purinergic signaleringskaskad 13.
I dessa studier använde vi PCK råttor, en modell av ARPKD orsakas av en spontan mutation i PKHD1 genen. Den PCK-stammen var originally härrörande från Sprague-Dawley (SD) råttor 16 därigenom SD-råttor används som en lämplig kontroll för jämförelse med PCK stammen. Som ett resultat kan både SD råtta nephron segment och icke-dilaterade samlingsrör isolerade från samma PCK-råttor tjänar som två olika jämförelsegrupper för experiment på cystisk epitel.
Vi beskrivs här tillämpningar av konventionell patch-clamp teknik och epifluorescence kalcium avbildning till cystisk epiteliala monolager som härrör från en mus genetisk modell av ARPKD. Protokollet består av tre steg, varav mest uppmärksamhet bör ägnas åt isoleringen av cystor (steg 1,5 avsnitt protokollen) och de elektrofysiologiska studier. Dessa viktiga förfaranden kräver omfattande utbildning och tålamod, och läsaren ska inte vara frustrerad på en gång.
