Imaging the dynamic behavior of organelles and other subcellular structures in vivo can shed light on their function in physiological and disease conditions. Here, we present methods for genetically tagging two organelles, centrosomes and mitochondria, and imaging their dynamics in living zebrafish embryos using wide-field and confocal microscopy.
In vivo imaging provides unprecedented access to the dynamic behavior of cellular and subcellular structures in their natural context. Performing such imaging experiments in higher vertebrates such as mammals generally requires surgical access to the system under study. The optical accessibility of embryonic and larval zebrafish allows such invasive procedures to be circumvented and permits imaging in the intact organism. Indeed the zebrafish is now a well-established model to visualize dynamic cellular behaviors using in vivo microscopy in a wide range of developmental contexts from proliferation to migration and differentiation. A more recent development is the increasing use of zebrafish to study subcellular events including mitochondrial trafficking and centrosome dynamics. The relative ease with which these subcellular structures can be genetically labeled by fluorescent proteins and the use of light microscopy techniques to image them is transforming the zebrafish into an in vivo model of cell biology. Here we describe methods to generate genetic constructs that fluorescently label organelles, highlighting mitochondria and centrosomes as specific examples. We use the bipartite Gal4-UAS system in multiple configurations to restrict expression to specific cell-types and provide protocols to generate transiently expressing and stable transgenic fish. Finally, we provide guidelines for choosing light microscopy methods that are most suitable for imaging subcellular dynamics.
In vivo beeldvorming verschaft directe visualisatie van cellulaire gedrag in de fysiologische context. De transparantie van de zebravis embryo's, hun snelle en externe ontwikkeling en een rijk scala aan genetische tools die tl-etikettering toe hebben bijgedragen aan het toenemende gebruik van in vivo microscopie om de dynamiek van de belangrijkste ontwikkelingsstoornissen gebeurtenissen op te helderen. Imaging studies van de ontwikkeling van het zenuwstelsel in de zebravis hebben bijvoorbeeld sterk uitgebreid onze kennis van het gedrag van neurale stamcellen en het lot van hun nageslacht met inbegrip van hun latere migratie, differentiatie en integratie circuit 1-8.
Het podium is nu ingesteld om de subcellulaire dynamiek ten grondslag liggen aan deze cellulaire gedrag te onderzoeken. Sterker nog, de zebravis worden reeds benut als instrumenten voor in vivo celbiologie. Het is nu mogelijk om mitochondriën 9-11 visualiseren, centrosomes 2,8,12-14, Golgi 15, De microtubule 4 en 16 actine cytoskelet, endosomen 17 en onderdelen van de apicale membraan complex 1,18, onder andere subcellulaire structuren in de zebravis embryo's in vivo. Tot dusver veel wat bekend is over de functie van deze organellen komt van het bestuderen van hun gedrag in gekweekte cellen. Terwijl in vitro studies enorme inzicht in celbiologie hebben opgeleverd, hebben de cellen in de cultuur niet volledig vertegenwoordigen de complexiteit van de in vivo situatie en dus niet noodzakelijk overeen met de functie en de dynamiek van subcellulaire organellen in vivo. Zebravis embryo's bieden een levensvatbare in vivo alternatief voor de behandeling van subcellulaire dynamiek.
Zoals vertebraten, zebravis bezitten vele orgaansystemen (bijvoorbeeld neurale retina) die homoloog zijn met die in zoogdiersoorten zijn. Bovendien zebravisembryo's worden in toenemende mate toegepast om menselijke ziekten 19,20 modelleren </sup>, ook op het gebied centrosomal functie (bijvoorbeeld, microcefalie 21 en congenitale amaurosis 22 Leber's) en mitochondriale functie (bijvoorbeeld de ziekte van Parkinson 23, tauopathieën 10,24 en Barthsyndroom 25). In vivo beeldvorming op cellulair en subcellulair niveau in deze gevallen zal een beter begrip van de celbiologie ten grondslag liggen aan deze pathologische toestanden mogelijk te maken.
De algemene doelstelling van de hier beschreven methoden is om een uitgebreide gids te verstrekken aan organellen en andere subcellulaire structuren in de zebravis embryo's met behulp van in vivo lichtmicroscoop te onderzoeken. Het volledige work-flow die betrokken zijn bij het visualiseren en het bijhouden van subcellulaire structuren in vivo wordt beschreven – van genetische labeling benaderingen, tot het genereren van transient uitdrukken en stabiele transgene vis, en ten slotte naar de beeldvorming met behulp van wide-field en confocale microscopie. Hoewel elk van deze procedures wordt gebruikt door talrijke laboratoria zebravis, worden de beschreven protocollen geoptimaliseerd en gestroomlijnde onderzoeken van de dynamica van subcellulaire structuren. Twee bijzondere aspecten van het hier beschreven werk garandeert vermelding: Ten eerste, het gebruik van het Gal4-UAS expressiesysteem in meerdere configuraties genetisch label organellen in specifieke celtypen. Ten tweede, een directe vergelijking van wide-field en confocale microscopie om de afbeelding subcellulaire structuren in vivo.
Huidige strategieën om genetisch label organellen en andere subcellulaire structuren in de zebravis ofwel gebruik maken van de afgetopte mRNA 1,4,8 of op basis van DNA-constructen, waar promotor elementen die rechtstreeks de expressie van fusie-eiwitten 9,14,15. In vitro getranscribeerd afgetopte RNA resultaten in te rijden snelle en brede expressie, die niet weefselspecifiek echter. Daarnaast expressie niveaus afnemen in de tijd als de afgetopte RNA wordt verdund of afgebroken. Waardoor het gebruik van RNA gebaseerdeconstrueert naar organel dynamiek in latere stadia van ontwikkeling is beperkt onderzocht (meestal tot 3 dagen na de bevruchting).
Deze beperkingen kunnen worden overwonnen met behulp van DNA-constructen, waarin ruimtelijke en temporele controle van expressie wordt bepaald door promotor elementen. Wanneer DNA gebaseerde constructen worden gebruikt in het kader van het Gal4-UAS systeem aanzienlijk verbetert transgenexpressie gerespecteerd worden 26,27. In dit bipartiete expressiesysteem, celtype-specifieke promoter elementen drijven de expressie van een transcriptionele activator Gal4, terwijl reporter genen stroomafwaarts van het Gal4-bindende stroomopwaartse activerende sequentie (UAS) gekloneerd. Door het combineren van UAS verslaggevers met de juiste Gal4 drivers, kan expressie worden beperkt tot specifieke celtypen, het omzeilen van de noodzaak om de reporter genen te klonen achter verschillende promotors elke keer dat een specifieke uitdrukking patroon gewenst is. Bovendien kan de expressie van meerdere genen UAS reporteraangedreven door een enkele Gal4 activator. Het Gal4-UAS systeem verschaft aldus een veelzijdig en flexibel genetische benadering voor subcellulaire etikettering.
Wide-field en confocale microscopen zijn de werkpaarden van de meeste laboratoria. Breedveldbeeld systemen gebruiken typisch een booglamp als lichtbron en het uitgezonden licht te detecteren met een gevoelige camera die is geplaatst aan het einde van de lichtweg. Dit beeldvormende modaliteit wordt meestal beperkt tot dunne monsters als out-of focus licht verduistert in-focus informatie in dikkere monsters. Confocale microscopen verschillen van wide-field systemen in dat ze zijn gebouwd om signalen die afkomstig zijn van de focal plane over degenen die afkomstig zijn uit focus (dat wil zeggen, "optische sectie") 28 te bevorderen. Om optische sectie bereiken van een pinhole wordt in de emissie pad in een conjugaat staat de puntlichtbron. Lasers worden gebruikt als lichtbronnen en signalen worden gedetecteerd met fotomultiplicatorbuizen (PMT). Praktisch, een laserbundel wordt gehaald via sample point-by-point en de fluorescentie-emissie Bij iedere (pixel) wordt gedetecteerd door de PMT.
Hier beeld dat we precies dezelfde subcellulaire structuren in levende zebravis embryo's met behulp van zowel wide-field en confocale microscopie om een directe vergelijking van beide microscopie modaliteiten te bieden. Het achterliggende doel van het verstrekken van dergelijke vergelijkingen is om richtlijnen voor het kiezen van de meest geschikte microscopie techniek voor de specifieke vraag bij de hand te bieden.
Met de hier beschreven tonen we aan Gal4 UAS-gebaseerde genetische kenmerken van mitochondria en centrosomes benaderingen. Deze organellen worden afgebeeld in verschillende celtypes van het zenuwstelsel en in spiercellen via wide-field en confocale microscopie om de geschiktheid van elke beeldvormende modaliteit tonen. De hier beschreven werkwijzen kunnen gemakkelijk worden aangepast voor het onderzoeken van andere organellen en subcellulaire structuren in levende zebravis embryo.
Hier tonen we de veelzijdigheid van het Gal4-UAS expressiesysteem om fluorescent label mitochondria, centrosomes en de cellulaire membranen van specifieke celtypen in vivo in zebravisembryo's.
