Laboratory-scale production of eukaryotic proteins with appropriate post-translational modification represents a significant barrier. Here is a robust protocol with rapid establishment and turnaround for protein expression using a mammalian expression system. This system supports selective amino acid, selective labeling of proteins and small molecule modulators of glycan composition.
Die Kunst der Herstellung von rekombinanten Proteinen mit komplexen post-translationale Modifikationen stellt eine große Herausforderung für die Untersuchung der Struktur und Funktion. Die rasche Einrichtung und hohe Rückgewinnung aus transient transfizierten Säugerzelllinien adressiert diese Barriere und ist ein wirksames Mittel zur Expression von Proteinen, die natürlicherweise durch das ER und Golgi-vermittelte sekretorischen Weg geleitet werden. Hier ist ein Protokoll für die Proteinexpression mit Hilfe der menschlichen HEK293F und HEK293s Zelllinien mit einem Säugetier-Expressionsvektor für hohe Proteinausbeuten entwickelt transfiziert. Die Anwendbarkeit dieses Systems wird gezeigt, unter Verwendung von drei repräsentativen Glycoproteine, die mit Ausbeuten zwischen 95 bis 120 mg gereinigtes Protein gewonnen pro Liter Kultur ausgedrückt. Diese Proteine sind die menschlichen FcγRIIIa und die Ratte α2-6 Sialyltransferase, ST6GalI, beide mit einem N-terminalen GFP-Fusions zum Ausdruck, ebenso wie das unmodifizierte humanen Immunglobulins G1 Fc. Das robuste System utilizes ein serumfreies Medium, das für die Expression von isotopisch angereicherten Proteine und Kohlenhydrate für Strukturuntersuchungen unter Verwendung von Massenspektrometrie und Kernspinresonanzspektroskopie anpassungsfähig ist. Weiterhin kann die Zusammensetzung des N-Glycan durch Zugabe einer kleinen Moleküls an bestimmten Glycans Modifikationen in einer Weise, die Ausbeute nicht verringert verhindern abgestimmt werden.
Hohe Ausbeuten an in geeigneter Weise gefaltet und posttranslational modifiziert Humanproteinen für die detaillierte Analyse von Struktur und Funktion wie vor eine große Herausforderung. Eine große Anzahl von Expressionssystemen zur Verfügung, die von rekombinanten Proteinen mit nativ-ähnlichen Funktion und Verhalten zu erzeugen. Bakterielle Expressionssysteme, überwiegend Escherichia coli-Stämme, repräsentieren die am leichtesten zugänglichen und am häufigsten verwendeten Werkzeuge in der Forschung Arena, wegen der Einfachheit dieser Expressionssysteme, obwohl Hefe-, Pflanzen-, Insekten- und Säugersystemen werden ebenfalls beschrieben 1-4. Der eukaryotischeGlykosylierungsmaschinerie ist umfangreich und der Lage, eine breite Palette von Modifikationen, einschließlich sowohl Asparagin (N) – und Serin / Threonin (O) -verknüpften komplexer Glykane 6. Es wird geschätzt, dass> 50% der menschlichen Proteine sind N-glycosyliert. 7 Glykane sind wesentliche Bestandteile von vielen Proteinen, einschließlich therapeutischer monoklonaler Antikörper, Erythropoietin und Blutgerinnungsfaktoren, wie Faktor IX, um einige zu nennen. Viele therapeutische Human Glykoproteine in rekombinanten Systemen, die Säugetierzellen produziert. Systeme Bemerkenswert sind die Chinese Hamster Ovary (CHO), Maus-Myelom (NS0), Baby-Hamster Niereny (BHK), menschliche embryonale Nieren (HEK-293) und die menschliche Netzhaut-Zelllinien, die in der Haftung oder Suspensionskultur für die Proteinproduktion 4,21,22 eingesetzt werden. Allerdings haben Säugerproteinexpressionssysteme die Erzeugung von stabilen Zelllinien, teure Nährmedien und Substrat unterstützte Transfektion Verfahren 23 erforderlich.
Säugerzelle Transfektion wird mit Hilfe einer Vielzahl von Mitteln, einschließlich Kalziumphosphate 24,25, kationische Polymere (DEAE-Dextran, Polybren, Polylysin, Polyethylimin (PEI)) oder positiv geladene kationische Liposomen 26-29 erreicht. PEI ist ein polykationisches, geladene, lineares oder verzweigtes Polymer (25 kDa) 26, die einen stabilen Komplex bildet mit DNA und wird durch Endozytose. Beim Ansäuern des Endosoms wird PEI gedacht zu quellen, was zu dem Bruch der Endosomen und Freisetzung der DNA ins Zytoplasma 26,30.
Bis vor kurzem war die transiente Transfektion in Sperre wiederauf Kultur wurde durch vorherigen DNA / PEI-Komplexbildung, gefolgt von der Zugabe zu der Zellkultur 29 durchgeführt. Allerdings Würm und Mitarbeiter berichteten über eine hocheffiziente Protokoll für die Produktion rekombinanter Proteine in HEK293-Zellen, die eine DNA / PEI-Komplex in situ gebildet 31,32 optimiert. Expressionsbedingungen können ebenfalls verändert werden, um das N-Glycan-Zusammensetzung auswirken.
Die HEK293s Zelllinie mit einem Gen-Deletion, das N-Glycan Verarbeitung stoppt in einer Zwischenstufe, zur Expression von Proteinen mit einheitlichen N-Glycanen, die aus 2-N-Acetylglucosamin-Reste plus fünf Mannosereste (Man 5 GlcNAc 2) 34, 35. Diese Zellenfehlt das N-Acetylglucosaminyl Transferase I (GntI) Gen, das für nachfolgende N-Glycan Verarbeitungs 36,37 erforderlich. Die Verwendung von Glycosyltransferase-Inhibitoren, einschließlich kifunensine Sialinsäure-Analogen und dem Fucose analogen und 2-Deoxy-2-fluor-Fucose hat ähnliche Wirkungen und Grenzen N-Glycan Verarbeitungs 38-41.
Das Protokoll verwendet die hier berichteten pGEn2 Vektors wie in Figur 1 gezeigt, 42,43, PEI stützten transiente Transfektion in Säugetierzelllinien (HEK293F oder HEK293s Zellen) und die Rückgewinnung von hohen Ausbeuten von geeignet glykosylierte Protein. Das System ist robust und kann von verschiedenen Faktoren, einschließlich Isotopenmarkierung und Glycan Technik für die Herstellung von großen Titern an rekombinante Proteine unterzubringen.
