Laboratory-scale production of eukaryotic proteins with appropriate post-translational modification represents a significant barrier. Here is a robust protocol with rapid establishment and turnaround for protein expression using a mammalian expression system. This system supports selective amino acid, selective labeling of proteins and small molecule modulators of glycan composition.
Kunsten å produsere rekombinante proteiner med komplekse post-translasjonelle modifikasjoner er en stor utfordring for studier av struktur og funksjon. Den raske etableringen og høy utvinning fra forbigående-transfekterte pattedyrcellelinjer løser denne barrieren, og er et effektivt middel for å uttrykke proteiner som er naturlig kanaliseres gjennom ER og Golgi-mediert sekresjonsveien. Her er en protokoll for protein uttrykk ved hjelp av menneskelig HEK293F og HEK293S cellelinjer transfektert med en pattedyrekspresjonsvektor designet for høye proteinutbytter. Anvendeligheten av dette systemet er vist ved bruk av tre representative glykoproteiner som uttrykt med utbytter mellom 95 til 120 mg av renset protein utvunnet pr liter kultur. Disse proteinene er den humane FcγRIIIa og rotte α2-6 sialyltransferase, ST6GalI, begge uttrykt med et N-terminalt fusjons GFP, samt umodifiserte humant immunglobulin Fc G1. Denne robuste system utilizes et serumfritt medium som kan tilpasses for ekspresjon av isotopisk anrikede proteiner og karbohydrater for strukturelle studier ved hjelp av massespektrometri og kjernemagnetisk resonansspektroskopi. Videre kan sammensetningen av N-glykan være innstilt ved tilsetning av et lite molekyl for å hindre at visse sukker modifikasjoner på en måte som ikke reduserer utbyttet.
Produsere høy avkastning av riktig brettet og post-translasjonelt modifiserte humane proteiner for detaljert analyse av struktur og funksjon er fortsatt en betydelig utfordring. Et stort antall ekspresjonssystemer er tilgjengelige som produserer rekombinante proteiner med naturlig-lignende funksjon og virkemåte. Bakterielle ekspresjonssystemer, hovedsakelig Escherichia coli-stammer, representerer de mest tilgjengelige og vanlige verktøy i forskning arenaen, på grunn av enkelheten i slike ekspresjonssystemer, skjønt gjær, planter, insekter og pattedyrsystemer er også beskrevet 1-4. Men de fleste av disse systemene er i stand til hensiktsmessige post-translasjonell modifikasjon av målproteinene. En grunnleggende interesse av Barb og Moremen laboratorier produserer eukaryote proteiner med passende glykosylering. Mange menneskelige proteiner kreve egnet glykosylering for riktig funksjon (se 5).
Den eukaryoteglykosylering maskiner er omfattende og i stand til å lage et variert utvalg av modifikasjoner, inkludert både asparagin (N) – og serin / treonin (O) -bundne komplekse glykaner 6. Det er anslått at> 50% av humane proteiner er N-glykosylert 7. Glykaner er essensielle komponenter av mange proteiner inkludert terapeutiske monoklonale antistoffer, erytropoietin og blodkoaguleringsfaktorer som faktor IX, for å nevne noen. Selv om flere metoder finnes for å forberede hensiktsmessig N-glykosylert proteiner og spenner fra rent syntetisk 8-10, til chemoenzymatic 11-14 eller utvinning fra konstruerte rekombinante systemer 15-20, ikke overraskende, har menneske ekspresjonssystemer så langt vist seg å være den mest robuste metoder for å generere humane proteiner.
Mange terapeutiske humane glykoproteiner produsert i rekombinante systemer ved bruk av pattedyrceller. Systemer av notatet er den kinesiske hamsterovarium (CHO), mus myelom (NS0), Baby Hamster Kidney (BHK), Human Embryonic Nyre (HEK-293) og menneske retinal cellelinjer som er ansatt i adhesjon eller suspensjon kultur for proteinproduksjon 4,21,22. Imidlertid har pattedyrprotein ekspresjonssystemer nødvendig å generere stabile cellelinjer, dyre vekstmedier og underlaget assistert transfeksjonsmidler prosedyrer 23.
Transfeksjon i pattedyrceller blir oppnådd ved hjelp av en rekke midler som omfatter kalsiumfosfater 24,25, kationiske polymerer (DEAE-dekstran, polybren, polylysin, polyethylimine (PEI)) eller positivt ladede kationiske liposomer 26-29. PEI er en polykationisk, ladet, lineær eller forgrenet polymer (25 kDa) 26 som danner et stabilt kompleks med DNA og endocytose. Ved surgjøring av endosomet, PEI er antatt å svelle, som fører til brudd av endosomer og frigjøring av DNA inn i cytoplasma 26,30.
Inntil nylig, transient transfeksjon i opphengningsi kultur ble utført ved forhånds DNA / PEI-kompleksdannelse, etterfulgt av tilsetning til cellekulturen 29. Men Würm og medarbeidere rapporterte en svært effektiv protokoll optimalisert for rekombinant proteinproduksjon i HEK293 celler som dannes en DNA / PEI kompleks in situ 31,32. Dette unngås fremstillingen, sterilisering av komplekset, og bufferbytte i et kulturmedium. Ytterligere optimalisering av blant annet uttrykk fremmende plasmider førte til betydelig avkastning øker 33. Heri er en metode som bygger på disse fremskrittene, og er bredt anvendelig. Expression forhold kan også endres for å påvirke N-glykan sammensetning.
Den HEK293S cellelinjen med et gen delesjon som stopper N-glykan prosessering på et mellomliggende trinn, fører til ekspresjon av proteiner med uniform N-glykaner som består av 2-N-acetylglucosaminrester pluss fem mannoserester (Man 5 GlcNAc 2) 34, 35. Disse cellenemangler den N-acetylglucosaminyl transferase I (GntI) genet som er nødvendig for nedstrøms N-glykan prosessering 36,37. Bruken av glykosyltransferase-inhibitorer, inkludert kifunensine, sialinsyre-analoger og fucose analog og 2-deoksy-2-fluor-fukose har lignende virkninger og begrensninger N-glykan prosesserings 38-41.
Av protokollen angitt her benytter pGEn2 vektoren som vist i figur 1 42,43, PEI assistert forbigående transfeksjon inn i pattedyrcellelinjer (HEK293F eller HEK293S celler), og gjenvinning av høye utbytter av korrekt glykosylert protein. Dette systemet er robust og kan romme ulike faktorer, inkludert isotop merking og glykan teknikk for fremstilling av store titere av rekombinante proteiner.
Denne protokollen illustrerer protein uttrykk via transient transfeksjon av HEK293F eller S-celler. De optimale transfeksjonsmidler vilkårene fastsatt i Barb og Moremen labs ansette en kritisk kombinasjon av celletetthet og reagenskonsentrasjoner å oppnå høy effektivitet transfeksjon.
