Laboratory-scale production of eukaryotic proteins with appropriate post-translational modification represents a significant barrier. Here is a robust protocol with rapid establishment and turnaround for protein expression using a mammalian expression system. This system supports selective amino acid, selective labeling of proteins and small molecule modulators of glycan composition.
Konsten att producera rekombinanta proteiner med komplexa posttranslationella modifieringar utgör en stor utmaning för studier av struktur och funktion. Den snabba etablering och hög återhämtning från transient transfekterade däggdjurscellinjer behandlar denna barriär och är ett effektivt sätt att uttrycka proteiner som naturligt kanaliseras genom ER och Golgi-medierad sekretoriska vägen. Här är ett protokoll för proteinexpression med användning av humana HEK293F och HEK293S cellinjer transfekterade med en däggdjursexpressionsvektor designad för höga proteinutbyten. Tillämpligheten av detta system demonstreras med användning av tre representativa glykoproteiner som uttryckte med utbyten mellan 95-120 mg renat protein återvinns per liter kultur. Dessa proteiner är den humana FcγRIIIa och råttan α2-6 sialyltransferas, ST6GalI, båda uttryckta med en N-terminal GFP-fusion, såväl som det omodifierade humant immunoglobulin G1-Fc. Denna robusta systemet utilizes ett serumfritt medium som är anpassningsbar för expression av isotopiskt anrikade proteiner och kolhydrater för strukturella studier med användning av masspektrometri och kärnmagnetisk resonansspektroskopi. Vidare kan sammansättningen av den N-glykan avstämmas genom tillsats av en liten molekyl för att förhindra vissa glykan modifieringar på ett sätt som inte minskar utbytet.
Producera höga utbyten av lämpligt vikta och posttranslationellt modifierade humana proteiner för detaljerad analys av struktur och funktion är fortfarande en stor utmaning. Ett stort antal uttryckssystem finns tillgängliga som producerar rekombinanta proteiner med nativ-liknande funktion och beteende. Bakteriella expressionssystem, främst Escherichia coli-stammar, representerar de mest tillgängliga och vanligen använda verktyg i forskningsarenan, på grund av enkelheten i dessa expressionssystem, även om jäst, växt-, insekts- och däggdjurssystem är också beskrivna 1-4. Majoriteten av dessa system är oförmögna att lämpliga post-translationell modifiering av målproteinerna. En grundläggande intresse Barb och Moremen laboratorier producerar eukaryota proteiner med lämplig glykosylering. Många humana proteiner kräver lämplig glykosylering för korrekt funktion (se 5).
Den eukaryotaglykosylering maskiner är omfattande och kan ge ett varierat utbud av modifieringar, både asparagin (N) – och serin / treonin (O) -länkade komplexa glykaner 6. Det uppskattas att> 50% av humana proteiner är N-glykosylerade 7. Glykaner är viktiga komponenter i många proteiner, inklusive terapeutiska monoklonala antikroppar, erytropoietin, och blod koagulationsfaktorer som faktor IX, för att nämna några. Även flera metoder finns för att förbereda sig på lämpligt sätt N-glykosylerade proteiner och sträcker sig från rent syntetisk 8-10, att chemoenzymatic 11-14 eller återhämtning från manipulerade rekombinanta system 15-20, inte helt oväntat, har humana expressionssystem som hittills visat sig vara den mest robusta metoder för att generera humana proteiner.
Många terapeutiska mänskliga glykoproteiner produceras i rekombinanta system med däggdjursceller. System att notera är den kinesiska hamsterovarie (CHO), mus myelom (NS0) Baby Hamster Kidney (BHK), humana embryonala njur (HEK-293) och mänskliga näthinnan cellinjer som används i vidhäftning eller suspensionsodling för proteinproduktion 4,21,22. Dock har däggdjursproteinexpressionssystem krävs generering av stabila cellinjer, dyra tillväxtmedier och substrat assisterad transfektion förfaranden 23.
Däggdjurscell transfektion uppnås med hjälp av ett stort antal medel inkluderande kalciumfosfater 24,25, katjoniska polymerer (DEAE-dextran, polybren, polylysin, polyetylimin (PEI)) eller positivt laddade katjoniska liposomer 26-29. PEI är en polykatjonisk, laddad, linjär eller grenad polymer (25 kDa) 26 som bildar ett stabilt komplex med DNA och är endocyteras. Vid försurning av endosomen är PEI tänkt att svälla, vilket leder till bristningar i endosomer och frisättning av DNA i cytoplasman 26,30.
Tills nyligen, transient transfektion i suspensiom kultur utfördes genom tidigare DNA / PEI komplexbildningen följt av tillsats till cellodlingen 29. Men Würm och medarbetare rapporterade en högeffektiv protokoll optimerad för rekombinant proteinproduktion i HEK293-celler som bildade en DNA / PEI-komplexet in situ 31,32. Detta undviks framställning, sterilisering av komplexet, och buffertutbyte i ett odlingsmedium. Ytterligare optimering genom att inkludera uttryck höjande plasmider ledde till betydande ökad avkastning 33. Häri är en metod som bygger på dessa framsteg och är brett tillämpbar. Uttrycks villkor kan också ändras för att påverka N-glykan komposition.
Den HEK293S cellinje med en gen deletion som stannar N-glykan bearbetning vid ett mellansteg, leder till expression av proteiner med enhetliga N-glykaner bestående av 2 N-acetylglukosaminrester plus fem mannosrester (Man 5 GlcNAc 2) 34, 35. Dessa cellersaknar den N-acetylglukosaminyltransferas I (GntI) gen som krävs för nedströms N-glykan bearbetning 36,37. Användningen av glykosyltransferasaktiviteter inhibitorer inklusive kifunensine, sialinsyra analoger och fukos analoga och 2-deoxi-2-fluor-fukos har liknande effekter och gränser N-glykan bearbetnings 38-41.
Protokollet redovisas här använder pGEn2 vektorn som visas i figur 1 42,43, PEI assisterad transient transfektion in i däggdjursceller linjer (HEK293F eller HEK293S celler), och återvinning av höga utbyten av lämpligt glykosylerat protein. Detta system är robust och kan anpassas till olika faktorer, inklusive isotopmärkning och glykan teknik för produktion av stora titrar av rekombinanta proteiner.
Detta protokoll illustrerar proteinexpression via transient transfektion av HEK293F eller S-celler.
