Caspases are the key mediators of apoptotic cell death via their proteolytic activity. When caspases are activated in cells to levels detectable by available technologies, apoptosis is generally assumed to occur shortly thereafter. Caspases can cleave many functional and structural components to cause rapid and complete cell destruction within a few minutes. However, accumulating evidence indicates that in normal healthy cells the same caspases have other functions, presumably at lower enzymatic levels. Studies of non-apoptotic caspase activity have been hampered by difficulties with detecting low levels of caspase activity and with tracking ultimate cell fate in vivo. Here, we illustrate the use of an ultrasensitive caspase reporter, CaspaseTracker, which permanently labels cells that have experienced caspase activity in whole animals. This in vivo dual color CaspaseTracker biosensor for Drosophila melanogaster transiently expresses red fluorescent protein (RFP) to indicate recent or on-going caspase activity, and permanently expresses green fluorescent protein (GFP) in cells that have experienced caspase activity at any time in the past yet did not die. Importantly, this caspase-dependent in vivo biosensor readily reveals the presence of non-apoptotic caspase activity in the tissues of organ systems throughout the adult fly. This is demonstrated using whole mount dissections of individual flies to detect biosensor activity in healthy cells throughout the brain, gut, malpighian tubules, cardia, ovary ducts and other tissues. CaspaseTracker detects non-apoptotic caspase activity in long-lived cells, as biosensor activity is detected in adult neurons and in other tissues at least 10 days after caspase activation. This biosensor serves as an important tool to uncover the roles and molecular mechanisms of non-apoptotic caspase activity in live animals.
Kaspaser är cysteinproteaser som förmedlar apoptotisk celldöd genom att klyva många intracellulära proteiner efter viktiga aspartat rester. Till exempel, initiator kaspaser aktivera effektor kaspaser, derepress DNA nukleaser, klyver cytoskelettala komponenter och förändra lipidkompositionen av cellmembran för att snabbt avveckla celler och stimulera deras erkännande och omvälvning av angränsande celler som omhändertar cell lik. 1-4 Det uppskattas att miljarder celler dör per dag i den mänskliga kroppen, och apoptos är en viktig mekanism för kemoterapiinducerad tumörcelldöd. 5 en annan uppsättning av caspaser kan orsaka celldöd av distinkta icke-apoptotiska processer för att stimulera medfödd immunitet. 6 Därför den mesta forskningen på kaspaser har fokuserat på sina pro-döden funktioner.
Intressant tidiga tecken på området visade att samma kaspaser ansvarar för att främja celldöd har också icke-död fSpecialfunktioner. Banbrytande studier har visat att kaspaser är involverade i olika cellulära funktioner i friska celler, inklusive reglering av celltillväxt och migration under fosterutvecklingen. 7-9 Kaspaser krävs för spermatid individualisering i Drosophila 10,11, för att blockera ett alternativ necroptotic celldöd vägen i möss 12,13, och mikroRNA bearbetning i C. elegans. 14,15 I kanske längsta bodde celler, neuroner, kaspaser och andra apoptotiska maskiner är inblandade i regleringen av nervaktivitet genom beskärning synaptiska ändelser, en process tros vara nödvändigt att stärka andra synapser för inlärning och minne. 16- 18 Det är möjligt att kaspaser underlättar synaptiska beskärning av en typ av mini-apoptos av små neuronala projektioner utan hela celldöd. 19 emellertid kaspaser kan ha alternativa funktioner inte har samband med apoptos-liknande händelser. 20,21 dubbla rollsi liv och död är inte unika för kaspaser, BCL-2 familjeproteiner och cytokrom c har roller i cell energier i friska celler, men är också en del av kärnan apoptotiska vägen som aktiveras av många typer av cellstress. 22-25 Även om det inte bevisat, verkar det logiskt att evolutionen har kopplat dag -jobb till döds jobb inom samma molekyler för att snabbt avskaffa olämpliga eller oönskade celler.
För närvarande är de molekylära mekanismerna för icke-apoptotiska kaspas-aktivitet inte förstått, och omfattningen av icke-apoptotiska kaspas aktivitet under embryonal utveckling och i vuxna vävnader är inte heller känt. En stor utmaning är att det är svårt att skilja dagsjobb från döds jobb kaspaser. I motsats till apoptos och pyroptosis, när kaspasaktivitet förstärks av en proteolytisk kaskad, är dags jobb kaspaser förväntas ske vid mycket lägre nivåer av enzymatisk aktivitet, sannolikt under upptäckt av många tillgängliga teknologies.
Före det arbete som presenteras här, andra utvecklat en rad olika caspase biosensorer för olika ändamål. Scat biosensorer (t.ex. ECFP-DEVD-Venus) snabbt upptäcka realtid kaspas-aktivitet i odlade celler och djurvävnader med hjälp av FRET. 26,27 Vid caspase klyvning, kärn riktade GFP delen av Apoliner (mCD8-RFP-DQVD- nucGFP) genomgår subcellulära relocalization inom några minuter när dess plasmamembran-tjuder klyvs av kaspaser. 28 Likaså ApoAlert-pCaspase3-sensor (NES-DEVD-YFP-NLS) relocalizes från cytosolen till kärnan på caspase klyvning. 29,30 Mer nyligen kromoforen i iCasper skickligt konstruerad för att fluorescera när klyvs av kaspaser, medger detektering av biosensoraktivitet i realtid i nervceller i Drosophila embryon, men framför allt i samband med utvecklings celldöd. 31 kaspas-beroende död lukt neuroner under åldrandet var demonstrankad av immuno-detektion av kaspas-klyvd form av CPV biosensorer (t.ex. mCD8-PARP-Venus). 32,33 Viktigt var den aktiverade formen av kaspas-3 detekterades i frånvaro av celldöd genom känsliga immunostain i taggar i odlade nervceller, och i soma med hjälp av kaspas-beroende fluorescens av kärn CellEvent reporter färgämne, men svårigheter påträffades på grund av fototoxicitet, men celldöd försenades tills efter eliminering ryggraden. 19 Således nya kaspas biosensorer behövs för att upptäcka och spåra celler med basal kaspas-aktivitet in vivo.
För att övervinna dessa svårigheter, genererade vi en ny dubbel färg kaspas biosensor, betecknad CaspaseTracker. Denna strategi kombinerar en modifierad version av Drosophila kaspas känsliga Apoliner biosensor 28 med Drosophila G-TRACE FRT rekombinassystem 34 för att permanent märka och spåra celler in vivo. <sup> 35 Den Gal4-aktiverade G-TRACE systemet tillåter mycket låga nivåer av kaspaser att aktivera CaspaseTracker, vilket resulterar i RFP uttryck i cytoplasman och permanent kärninriktade GFP-uttryck i någon cell som någonsin har upplevt kaspas-aktivitet. 35 Detta system kan märka celler under hela livet i hela djur med användning av Drosophila melanogaster, en lätthanterlig och mycket använt modellsystem för studium av kaspaser och celldöd. 36-38
Här visar vi bygg- och inre arbetet i CaspaseTracker som underlättar upptäckt av omfattande basal kaspas-aktivitet i friska vävnader.
