The branchial skeleton, including gill rakers, pharyngeal teeth, and branchial bones, serves as the primary site of food processing in most fish. Here we describe a protocol to dissect and flat-mount this internal skeleton in threespine sticklebacks. This method is also applicable to a variety of other fish species.
The posterior pharyngeal segments of the vertebrate head give rise to the branchial skeleton, the primary site of food processing in fish. The morphology of the fish branchial skeleton is matched to a species’ diet. Threespine stickleback fish (Gasterosteus aculeatus) have emerged as a model system to study the genetic and developmental basis of evolved differences in a variety of traits. Marine populations of sticklebacks have repeatedly colonized countless new freshwater lakes and creeks. Adaptation to the new diet in these freshwater environments likely underlies a series of craniofacial changes that have evolved repeatedly in independently derived freshwater populations. These include three major patterning changes to the branchial skeleton: reductions in the number and length of gill raker bones, increases in pharyngeal tooth number, and increased branchial bone lengths. Here we describe a detailed protocol to dissect and flat-mount the internal branchial skeleton in threespine stickleback fish. Dissection of the entire three-dimensional branchial skeleton and mounting it flat into a largely two-dimensional prep allows for the easy visualization and quantification of branchial skeleton morphology. This dissection method is inexpensive, fast, relatively easy, and applicable to a wide variety of fish species. In sticklebacks, this efficient method allows the quantification of skeletal morphology in genetic crosses to map genomic regions controlling craniofacial patterning.

En trofisk egenskap av økologisk betydning er fordelingen av gjelle rakers, periodiske dermal bein som linje fremre og bakre ansiktene til branchial bein og brukes til å filtrere byttedyr. Fisk som vanligvis lever av små byttedyr tendens til å ha lengre og tettere mellomrom gill rakers forhold til fisk som lever av større byttedyr 14,15. Variasjon i gjelle rakers har blitt rapportert både mTVen på og mellom arter 14-19, og aspekter ved gjelle raker mønster bidra til trofiske nisjer og trenings 16. Tiår med forskning har grundig dokumentert gjelle raker antall og lengde variasjon i threespine stingsild 17 – 21; Men disse studiene typisk fokusere på den første raden av gill rakers. Nyere arbeider har vist modularitet i den genetiske kontrollen av gill rytterens tall over branchial skjelettet 22,23 og over en enkelt rad i gjelle raker avstand 23 og lengde 24 fremhever betydningen av å studere mer enn ro en eller en enkelt gill raker å forstå utviklings genetiske grunnlaget for gill rytterens reduksjon.
En andre trofiske egenskap av både økologisk og biomedisinsk betydning er fordelingen av svelgtann. Tennene i fisk kan ligge både i den orale kjeve og i branchial skjelett, kjent som svelgtann. Oral tenner brukes primært for prey fange mens svelgtann brukes til tygging og byttedyr manipulasjon 25-27. Begge settene danne via felles utviklingsmekanismer og anses utviklings homolog 28. Interessant modularitet oppstår der noen arter, for eksempel sebrafisk, mangel muntlig og rygg svelget tenner 29, mens andre arter har flere tann ceratobranchials, pharyngobranchials, og noen ganger toothed basihyal og hypobranchials 30. I stingsild, er svelgtann funnet ventralt på den femte ceratobranchial og dorsally på fremre og bakre pharyngobranchials 31. I ciklider, lavere svelget kjeve morfologi varierer dramatisk 32,33 og har vist seg å være tilpasningsdyktige og korrelert med trofisk nisje 34. multiple ferskvann stingsild bestander har utviklet seg dramatiske økninger i ventral svelget tann nummer 23,35,36. Nyere arbeider har vist at utviklings genetiske grunnlaget for dette utviklet seg tann gevinsten er stor grad tydelig i to uavhengig avledet bestander av ferskvanns stingsild 36. I motsetning til pattedyr tenner, fisk fornye sine tenner kontinuerlig gjennom hele voksenlivet 37. Begge disse tidligere beskrevet høye tannferskvannsbestander har utviklet en akselerert tann erstatningsrate, noe som gir en sjelden virveldyr system for å studere genetiske grunnlaget for regenerering 36.
En tredje trofiske egenskap som har utviklet seg gjentatte ganger i ferskvann stingsild er lengre epibranchial og ceratobranchial bein, de branchial bue segment homologer i øvre og nedre kjeve, henholdsvis 38. Lengre branchial bein gi en større munnhulen og trolig er adaptive for å tillate større byttedyr å være consumed. Videre i annen fisk, epibranchial bein er viktig for depresjonen i rygg svelget tannplatene 25. Som gill rakers og svelgtann, de branchial bein er interne og dermed vanskelig å enkelt visualisere eller tallfeste.
Her presenterer vi en detaljert protokoll for å dissekere og flat-montere branchial skjelettet, noe som gir enkel visualisering og kvantifisering av en rekke viktige kraniofaciale egenskaper.
