Anastasis is technisch uitdagende te detecteren in vivo omdat de cellen die de cel dood proces hebben omgekeerd morfologisch niet te onderscheiden van normale gezonde cellen kunnen. Hier beschrijven we protocollen voor het opsporen en volgen van cellen die anastasis in levende dieren ondergaan met behulp van ons nieuw ontwikkelde in vivo CaspaseTracker biosensor systeem.
Anastasis (Grieks voor “stijgen tot leven”) is een recent ontdekte cel herstel fenomeen waarbij cellen sterven kunt omkeren alsnog cel dood processen die in het algemeen wordt verondersteld dat zijn intrinsiek onomkeerbaar. Bevordering van anastasis kan in principe reddings- of behouden gewond cellen die moeilijk te vervangen zoals cardiomyocytes of neuronen, teneinde het herstel van het weefsel. Omgekeerd, onderdrukken van anastasis in kankercellen, apoptosis na anti-kanker therapieën, ondergaan kan zorgen voor de dood van de cel van de kanker en verminderen de kans op herhaling. Echter, deze studies hebben belemmerd door het ontbreken van hulpmiddelen voor het bijhouden van het lot van de cellen die u ondergaan anastasis in levende dieren. De uitdaging is het identificeren van de cellen die de cel dood proces ondanks hun morfologisch normale uiterlijk na herstel hebben omgekeerd. Om deze moeilijkheid te overwinnen, hebben wij ontwikkeld Drosophila en zoogdieren CaspaseTracker biosensor systemen die kunnen identificeren en de anastatic cellen in vitro of in vivopermanent kan bijhouden. Hier presenteren we in vivo protocollen voor de generatie en het gebruik van het systeem van de dubbele biosensor CaspaseTracker op te sporen en het bijhouden van anastasis bij Drosophila melanogaster na voorbijgaande blootstelling aan cel dood prikkels. Terwijl conventionele biosensoren en protocollen cellen actief ondergaat apoptotic celdood etiketteren kunnen, de CaspaseTracker biosensor kan permanent het label van cellen die zijn hersteld na activering van caspase – een waarmerk van laat-stadium apoptosis, en om actieve apoptotic processen gelijktijdig te identificeren. Deze biosensor kan ook het herstel van de cellen die geprobeerd andere vormen van dood van de cel die direct of indirect caspase-activiteiten betrokken bijhouden. Daarom dit protocol stelt ons in staat om continu bijhouden van het lot van deze cellen en hun nakomelingen, vergemakkelijking van toekomstige studies van de biologische functies, moleculaire mechanismen, fysiologische en pathologische gevolgen en therapeutische implicaties van Anastasis. We bespreken ook de nodige controles om te onderscheiden van de cellen die u ondergaan van anastasis uit die het weergeven van niet-apoptotic caspase activiteit in vivo.
Geprogrammeerde celdood, zoals apoptosis, speelt een essentiële rol in de embryonale ontwikkeling en normale homeostase door het elimineren van ongewenste, gewonde of gevaarlijke cellen in meercellige organismen1,2,3. Het verlies van evenwicht tussen celdood en overleving kan leiden tot fatale gevolgen hebben, zoals kanker, hartfalen, auto-immuniteit en degeneratie4,5,6,7,8. Activering van de beul caspases heeft traditioneel beschouwd als de “point of no return” in apoptosis9,10,11, zoals het snelle en massale cellulaire sloop12, triggers 13,14,15,16. Uitdagend deze algemene dogma, we laten zien dat kunnen gekweekte stervende primaire cellen en kankercellen dood kenmerken waaronder plasmamembraan blebbing, cel krimp, herstellen aan niet alleen na activering van caspase, maar ook volgende belangrijke cel mitochondriale fragmentatie, introductie van mitochondriale cytochroom c in het cytosol, nucleaire en condensatie van de chromatine, DNA-beschadiging, nucleaire fragmentatie, mobiele oppervlakte blootstelling van fosfatidylserine (PS), en de vorming van apoptotic organen 17 , 18 , 19 , 20 , 21. Wij stellen voor dat anastasis is een intrinsieke cel herstel fenomeen, zoals stervende cellen na verwijdering van cel dood stimuli17,18,19,20, herstellen kunnen 21. wij bedacht de term “Anastasis” (Αναστάσης)18, wat betekent “rising tot leven” in het Grieks te beschrijven dit onverwachte cel herstel verschijnsel. Onze waarneming van de anastasis wordt verder ondersteund door recente onafhankelijke studies die ook herstel van cellen onthullen na fosfatidylserine externalization22,23,24, mitochondriale beperkt buitenste membraan permeabilization25, activering van gemengde afkomst kinase-achtige (MLKL) en cel krimp26.
Karakterisering van de mechanismen tot regeling van de anastasis, worden de paradigma-shifting fysiologische, pathologische en therapeutische gevolgen zal hebben. Anastasis kon vertegenwoordigen een voorheen onbekende cytoprotective mechanisme te redden of te bewaren van belangrijke postmitotic cellen en weefsels die moeilijk te vervangen, en eventueel rekening voor hartfalen omkering door ventriculaire lossen met links ventriculaire apparaten (LVADs)27,28, herstel van fotoreceptor cellen na voorbijgaande blootstelling van buitensporige lichte29,30,31, of reparatie van neuronen helpen na hersenen letsel32. Als dus het bevorderen van anastasis cel- en weefseltransplantaties herstel versterken kan. Daarentegen zou anastasis een tactiek van de onverwachte ontsnappen door kankercellen gebruikt om te overleven van cel-dood-inducerende therapie, veroorzaakt kanker herhaling17,18. Dus, het onderdrukken van anastasis in het sterven van kankercellen tijdens en na de kankerbehandeling zou een nieuwe therapeutische strategie om te genezen van kanker door te voorkomen dat hun herval.
Tijdens het proces van anastasis, hebben we gevonden dat sommige herstelde cellen verworven permanente genetische veranderingen en onderging oncogene transformatie, waarschijnlijk te wijten aan DNA-schade opgelopen tijdens apoptosis18,20,21 . Omkeren van het proces van de dood van DNA-beschadigde cellen zou een mechanisme van tumorvorming, potentieel ten grondslag liggen aan de observatie dat herhaald weefsel schade verhoogt het risico van kanker in een verscheidenheid van weefsels, zoals chronische thermische verwonding in de slokdarm geïnduceerde door de consumptie van zeer warme dranken33,34,35, leverschade als gevolg van alcoholisme36,37, tumor evolutie na genotoxische kanker therapie38, 39,40, en ontwikkeling van nieuwe kankergevallen van normale weefsels die zich tijdens de interval tussen de cycli van de anti-kanker therapie41,42,43,44 voordoen . Als de waarde true is, kon gericht op anastasis voorkomen of arresteren van de ontwikkeling van kanker en de progressie. We hebben gevonden dat stervende kiemcellen honger geïnduceerde anastasis in opnieuw gevoed Drosophila19ondergaan. Als anastasis in geslachtscellen met DNA-beschadiging optreedt, zou het account voor de observatie dat verlengd milieudruk bevordert de ontwikkeling van genetische ziekten. Bijvoorbeeld, bijdragen hongersnoden tot de ontwikkeling van transgenerationele overneembare ziekten zoals diabetes en hart-vaatziekten45. Daarom kan begrip anastasis leiden tot de strategieën voor de preventie van het ontwikkelen van overneembare ziekten veroorzaakt door dit potentiële mechanisme.
Om te profiteren van de ontdekking van anastasis en direct te ontwikkelen innovatieve therapieën, is het essentieel om te bestuderen van de oorzaak en het gevolg van anastasis in levende dieren. Het is echter technisch uitdagend om te identificeren en te volgen anastatic cellen in vivo, omdat de cellen die verhaald van cel dood proces morfologisch niet te onderscheiden van normale gezonde cellen weergegeven, en er geen biomerker voor anastasis is geïdentificeerd nog2118,17,. Om deze problemen aanpakken, ontwikkelden we onlangs een nieuwe in vivo caspase biosensor aangewezen “CaspaseTracker”19, identificeren en traceren van cellen die apoptosis na caspase activering19,46 overleven, de kenmerk van apoptosis10,14. Onderscheiden van de ‘real-time’ caspase-biosensoren zoals SCAT12,47, Apoliner48, CA-GFP49, ApoAlert18,50, C3AIs51 en iCasper52 dat sporen gaande caspase activiteit, de CaspaseTracker biosensor bovendien wezenstrek naar de vermogen voor permanent label cellen die uitdrukkelijke caspase-activiteit zelfs Transient. Daarom wordt het de CaspaseTracker biosensor lange termijn bijhouden ingeschakeld voor anastasis na omkering van caspase-gemedieerde cel dood proces in vivo.
Het systeem van de dubbele biosensor CaspaseTracker is een roman en uniek instrument dat maakt de ontdekking van recente of lopende caspase-activiteit, en het bijhouden van cellen die zijn omgekeerd celdood verwerken en overleven na het ervaren van caspase activiteit in vivo. Terwijl caspase activiteit traditioneel als een stempel van apoptosis aangenomen is, openbaren groeiende studies dat niet-apoptotic caspase activiteit potentiële rollen in uiteenlopende normale cel functies…
