Anastasis è tecnicamente difficile da rilevare in vivo perché le cellule che hanno invertito il processo di morte cellulare possono essere morfologicamente indistinguibili dalle cellule sane normali. Qui descriviamo i protocolli per il rilevamento e il monitoraggio delle cellule che subiscono anastasis animali vivi utilizzando il nostro sistema di biosensore del CaspaseTracker recente sviluppato in vivo .
Anastasis (dal greco “risurrezione alla vita”) è un fenomeno di recupero cellulare recentemente scoperto per cui morire cellule può invertire i processi di morte delle cellule della fase tardiva che sono presupposti generalmente per essere intrinsecamente irreversibili. Promozione anastasis potrebbe in principio salvataggio o preservare ferito le cellule che sono difficili da sostituire come cardiomiociti o neuroni, facilitando così il recupero dei tessuti. Al contrario, sopprimendo anastasis in cellule tumorali, che subiscono gli apoptosi dopo terapie anti-cancro, può garantire la morte della cellula tumorale e ridurre le probabilità di recidiva. Tuttavia, questi studi sono stati ostacolati dalla mancanza di strumenti per il monitoraggio il destino delle cellule che subiscono anastasis in animali vivi. La sfida consiste nell’identificare le cellule che hanno invertito il processo di morte cellulare nonostante il loro aspetto morfologicamente normale dopo il recupero. Per superare questa difficoltà, abbiamo sviluppato in Drosophila e sistemi di biosensore CaspaseTracker mammiferi che possono identificare e tracciare definitivamente anastatica cellule in vitro o in vivo. Qui, presentiamo in vivo protocolli per la generazione e l’utilizzo del sistema dual biosensore CaspaseTracker per rilevare e tenere traccia di anastasis in Drosophila melanogaster dopo esposizione transitoria agli stimoli di morte delle cellule. Mentre convenzionale biosensori e protocolli possono marcare le cellule attivamente che subisce la morte apoptotica delle cellule, il biosensore CaspaseTracker permanentemente può marcare le cellule che hanno recuperato dopo l’attivazione di caspase – un marchio di garanzia dell’apoptosi in fase avanzata, e contemporaneamente identificare processi apoptotici attivo. Questo biosensore può anche monitorare il recupero delle cellule che hanno tentato di altre forme di morte delle cellule che direttamente o indirettamente interessate attività di caspase. Di conseguenza, questo protocollo permette di monitorare continuamente il destino di queste cellule e la loro progenie, facilitando gli studi futuri delle funzioni biologiche, meccanismi molecolari, le conseguenze fisiologiche e patologiche e implicazioni terapeutiche di Anastasis. Inoltre discutiamo i controlli appropriati per distinguere le cellule che subiscono anastasis da quelle che visualizzano non apoptotiche caspase attività in vivo.
Morte programmata delle cellule, come apoptosi, svolge un ruolo essenziale nello sviluppo embrionale e normale omeostasi eliminando le cellule indesiderate, ferite o pericolose in organismi multicellulari1,2,3. La perdita di equilibrio tra sopravvivenza e morte cellulare può portare a conseguenze fatali come cancro, insufficienza cardiaca, dell’autoimmunità e degenerazione4,5,6,7,8. Attivazione del boia caspases tradizionalmente è stato considerato come il “punto di non ritorno” in apoptosi9,10,11, come si innesca rapida e massiccia demolizione cellulare12, 13,14,15,16. Impegnativo questo dogma generale, abbiamo dimostrato che cellule primarie morente coltivate e le cellule tumorali possono recuperare non solo dopo l’attivazione delle caspasi, ma anche seguente cellula importante tra cui membrana plasmatica blebbing, restringimento delle cellule, tratti distintivi di morte frammentazione mitocondriale, rilascio di mitocondriale del citocromo c nel cytosol, nucleare e condensazione della cromatina, danno del DNA, frammentazione nucleare, l’esposizione di superficie delle cellule della fosfatidilserina (PS) e la formazione di corpi apoptotici 17 , 18 , 19 , 20 , 21. vi proponiamo quella anastasis è un fenomeno di recupero intrinseco delle cellule, come le cellule morte possono recuperare dopo la rimozione delle cellule morte stimoli17,18,19,20, 21. abbiamo coniato il termine “Anastasis” (Αναστάσης)18, che significa “rising alla vita” in greco, a descrivere questo fenomeno di recupero cellulare inaspettato. La nostra osservazione di anastasis è ulteriormente supportata da recenti studi indipendenti che rivelano anche il recupero delle cellule dopo la fosfatidilserina esternalizzazione22,23,24, limitata mitocondriale esterna di permeabilizzazione della membrana25, attivazione di stirpe misto chinasi-come (MLKL) e cella restringimento26.
Caratterizzare i meccanismi che regolano l’anastasis avrà implicazioni fisiologiche, patologiche e terapeutiche lo spostamento di paradigma. Anastasis potrebbe rappresentare un meccanismo di cytoprotective precedentemente sconosciuti per salvare o preservare importanti cellule postmitotic e tessuti che sono difficili da sostituire e possibilmente conto per l’inversione di insufficienza cardiaca ventricolare scaricando con ventricolare di sinistra assistere dispositivi (LVADs)27,28, recupero di cellule fotorecettrici dopo esposizione transitoria di luce eccessiva29,30,31, o la riparazione dei neuroni dopo la ferita di cervello32. Se è così promuovere anastasis potrebbe migliorare il recupero di cellule e tessuti. Al contrario, anastasis potrebbe essere una tattica di fuga inaspettata usata dalle cellule tumorali per sopravvivere terapia d’induzione di morte delle cellule, causando cancro ricorrenza17,18. Di conseguenza, sopprimendo anastasis morendo, le cellule tumorali durante e dopo il trattamento del cancro potrebbe essere una strategia terapeutica novella per curare tumori impedendo loro ricaduta.
Durante il processo di anastasis, abbiamo trovato che alcune cellule recuperate acquisito alterazioni genetiche permanente e ha subito la trasformazione oncogena, probabilmente a causa di danni al DNA sostenute durante l’apoptosi18,20,21 . Invertire il processo di morte delle cellule con DNA danneggiato potrebbe essere un meccanismo della tumorigenesi, potenzialmente sottostante l’osservazione che ripete la lesione del tessuto aumenta il rischio di cancro in una varietà di tessuti, come lesione cronica termica nell’esofago indotto tramite il consumo di bevande molto calde33,34,35, danni al fegato a causa di alcolismo36,37, evoluzione del tumore dopo genotossici cancro terapia38, 39,40e lo sviluppo di nuovi cancri dai tessuti normali che si verificano durante gli intervalli tra i cicli di terapia anti-cancro41,42,43,44 . Se true, targeting anastasis potrebbe prevenire o arrestare e progressione del tumore. Abbiamo trovato che indotta da inedia morente le cellule di germe subiscono anastasis in ri-nutriti Drosophila19. In caso di anastasis in cellule di germe con danno del DNA, potrebbe essere conto per l’osservazione che il prolungato stress ambientale promuove lo sviluppo di malattie genetiche. Ad esempio, carestie e contribuire allo sviluppo di transgenerazionale ereditabili malattie come il diabete e malattie cardiache coronariche45. Di conseguenza, anastasis comprensione potrebbe portare a strategie per la prevenzione di malattie ereditarie causate da questo meccanismo potenziale di sviluppo.
Per sfruttare la scoperta dell’anastasis e dirigerla per sviluppare terapie innovative, è essenziale per studiare la causa e conseguenza di anastasis in animali vivi. Tuttavia, è tecnicamente difficile da identificare e tracciare anastatica cellule in vivo, perché le cellule che ha recuperato dal processo di morte cellulare appaiono morfologicamente indistinguibili dalle cellule sane normali, e non c’è nessun biomarcatore dell’anastasis identificato ancora17,18,21. Per risolvere questi problemi, abbiamo recentemente sviluppato un nuova in vivo caspase biosensore indicato “CaspaseTracker”19, per identificare e tenere traccia di cellule che sopravvivono apoptosi dopo19,l’attivazione di caspase46, il segno distintivo di apoptosi10,14. Distinguendola dai biosensori di caspasi “tempo reale” come SCAT12,47, Apoliner48, CA-GFP49, ApoAlert18,50, C3AIs51 e iCasper52 che rileva l’attività della caspasi in corso, il biosensore CaspaseTracker inoltre offre la possibilità per marcare permanentemente le cellule che attività di caspase espressa anche transitoriamente. Pertanto, il biosensore CaspaseTracker consente la registrazione a lungo termine dell’anastasis dopo inversione di caspase-mediata delle cellule morte processo in vivo.
Il sistema di biosensore dual CaspaseTracker è un romanzo e l’unico strumento che permette la rilevazione di recente o attività di caspase in corso, rilevamento delle cellule che hanno invertito la morte delle cellule e di sopravvivere dopo aver sperimentato caspase attività in vivo.
