Anastasis tekniskt utmanande för att upptäcka i vivo eftersom cellerna som återförts cell dödsprocessen kan vara morfologiskt oskiljbara från normala friska celler. Här beskriver vi protokoll för att upptäcka och spåra celler som genomgår anastasis i levande djur genom att använda vår biosensor nyutvecklade i vivo CaspaseTracker system.
Anastasis (grekiska för ”rising till liv”) är ett nyligen upptäckt cell återhämtning fenomen där döende celler kan återföra sent stadium cell death processer som generellt antas vara oupplösligt irreversibel. Att främja anastasis kunde i princip undsättnings- eller bevara skadade celler som är svåra att ersätta exempelvis hjärtmuskelceller eller nervceller, vilket underlättar vävnad återhämtning. Omvänt, undertrycka anastasis i cancerceller, som genomgår apoptos efter anti-cancer terapier, kan säkerställa cancer celldöd och minska risken för återfall. Dessa studier har dock hindrats av bristen på verktyg för att spåra celler som genomgår anastasis i levande djur öde. Utmaningen är att identifiera de celler som har återförts cell dödsprocessen trots deras morfologiskt normala utseende efter återhämtning. För att övervinna denna svårighet, har vi utvecklat Drosophila och däggdjur CaspaseTracker biosensor system som kan identifiera och spåra permanent anastatic celler in vitro eller in-vivo. Här presenterar vi i vivo protokoll för generering och användning av CaspaseTracker dubbla biosensor systemet att identifiera och spåra anastasis i Drosophila melanogaster efter övergående exponering för cell death stimuli. Medan konventionella biosensorer och protokoll kan märka celler aktivt genomgår apoptotisk celldöd, den CaspaseTracker biosensor permanent kan märka celler som har återhämtat sig efter aktivering av kaspas – ett kännetecken för sent stadium apoptos, och samtidigt identifiera aktiva apoptotiska processer. Detta biosensor kan också spåra återvinning av de celler som försökt andra former av celldöd som direkt eller indirekt inblandade kaspas aktivitet. Därför detta protokoll gör det möjligt för oss att kontinuerligt följa dessa celler och deras avkomma, underlätta framtida studier av biologiska funktioner, molekylära mekanismer, fysiologiska och patologiska konsekvenserna och terapeutiska konsekvenser av öde Anastasis. Vi diskuterar också lämpliga kontroller för att skilja celler som genomgår anastasis från dem som visar icke-apoptotiska kaspas aktivitet in vivo.
Programmerad celldöd, såsom apoptos, spelar en viktig roll i embryonal utveckling och normala homeostas genom att eliminera oönskade, skadade eller farliga celler i flercelliga organismer1,2,3. Förlust av balans mellan celldöd och överlevnad kan leda till fatala konsekvenser såsom cancer, hjärtsvikt, autoimmunitet och degeneration4,5,6,7,8. Aktivering av bödeln kaspaserna har traditionellt betraktats som den ”point of no return” apoptos9,10,11, eftersom det utlöser snabba och massiva cellulära rivning12, 13,14,15,16. Utmanande denna allmänna dogm, visat vi att odlade döende primära celler och cancerceller kan återställa inte bara efter kaspas aktivering, men också följande viktiga cell death kännetecken inklusive plasmamembranet blebbing, cell krympning, mitokondriell fragmentering, frisläppandet av mitokondriell cytokrom c till cytosolen, kärnkraft och kromatin kondens, DNA-skador, nukleära fragmentering, cell surface exponering av Fosfatidylserin (PS) och bildandet av apoptotiska kroppar 17 , 18 , 19 , 20 , 21. vi föreslår att anastasis är ett inneboende cell återhämtning fenomen, som döende celler kan återhämta sig efter borttagning av cell death stimuli17,18,19,20, 21. vi myntade termen ”Anastasis” (Αναστάσης)18, vilket betyder ”rising till liv” på grekiska, till beskriva fenomenet oväntad cell återhämtning. Vår observation av anastasis stöds ytterligare av senaste oberoende studier som också avslöjar återvinning av celler efter fosfatidylserin externalisering22,23,24, begränsat mitokondriell yttre membran permeabilisering25, aktivering av blandade härstamning kreatinkinas-liknande (MLKL) och cell krympning26.
Karakterisera de mekanismer som reglerar anastasis får paradigm-shifting fysiologiska, patologiska och terapeutiska konsekvenser. Anastasis kan representera en tidigare okänd cytoprotektiva mekanism att rädda eller bevara viktiga postmitotic celler och vävnader som är svåra att ersätta, och möjligen konto för hjärtsvikt återföring av ventrikulära lossning med vänsterkammardysfunktion hjälpa enheter (LVADs)27,28, återvinning av ljusmätare celler efter övergående exponering av överdriven ljus29,30,31, eller reparation av nervceller efter hjärnan skada32. Om så att främja anastasis kunde förbättra cell- och återhämtning. Anastasis kan däremot vara en oväntad fly taktik används av cancerceller för att överleva cell-död-inducerande terapi, orsakar cancer återfall17,18. Därför undertrycka anastasis i döende cancerceller under och efter behandling av cancer kan vara en roman terapeutisk strategi att bota cancer genom att förhindra deras återfall.
Under processen för anastasis, har vi funnit att vissa återvunna celler förvärvat permanent genetiska förändringar och genomgick Onkogen transformation, sannolikt på grund av DNA-skador som uppstått under apoptos18,20,21 . Backning dödsprocessen DNA-skadade celler kan vara en mekanism för uppkomst, potentiellt bakomliggande iakttagelsen att upprepade vävnadsskada ökar risken för cancer i en mängd olika vävnader, såsom kronisk termisk skada i matstrupen inducerad av konsumtion av mycket varma drycker33,34,35, leverskador på grund av alkoholism36,37, tumör evolution efter genotoxiska cancer behandling38, 39,40, och utvecklingen av de nya cancerfallen från normal vävnad som uppstår under pauserna mellan cykler av cancerbehandling41,42,43,44 . Om sant, kunde inriktning anastasis hindra eller arrestera cancerutveckling och progression. Vi har funnit att svält-inducerad döende könsceller genomgår anastasis i åter fed Drosophila19. Om anastasis sker i könsceller med DNA-skador, det konto för observationen att långvarig miljöbelastning främjar utvecklingen av genetiska sjukdomar. Exempelvis bidrar svältar till utvecklingen av transgenerationell ärftliga sjukdomar som diabetes och koronara hjärtsjukdomar45. Förståelse anastasis kan därför leda till strategier för att förebygga utveckling av ärftliga sjukdomar som orsakas av denna potentiella mekanism.
För att utnyttja upptäckten av anastasis och direkt kunna utveckla innovativa behandlingar, är det viktigt att studera orsak och konsekvens av anastasis i levande djur. Dock det tekniskt svårt att identifiera och spåra anastatic celler i vivo, eftersom cellerna som återhämtat sig från cell death process verkar morfologiskt oskiljbara från normala friska celler, och det finns ingen biomarkör för anastasis identifierade ännu17,18,21. För att lösa dessa problem, vi nyligen utvecklat ett nytt i vivo kaspas biosensor designerat ”CaspaseTracker”19, att identifiera och spåra celler som överlever apoptos efter kaspas aktiveringen19,46, den kännetecknande för apoptos10,14. Skilja det från ”realtid” kaspas biosensorer som SCAT12,47, Apoliner48, CA-GFP49, ApoAlert18,50, C3AIs51 och iCasper52 att upptäcka pågående kaspas aktivitet, den CaspaseTracker biosensor har dessutom förmågan att permanent etikett celler express kaspas aktiviteten även övergående. Den CaspaseTracker biosensor kan därför långsiktigt spårning av anastasis efter återföring av kaspas-medierad cell death process i vivo.
Systemets CaspaseTracker dubbla biosensor är en ny och unikt verktyg som möjliggör detektering av senaste eller pågående kaspas aktivitet, och spårning av celler som har återförts celldöd och överleva efter att ha upplevt kaspas aktivitet i vivo.
