A method is described herein for the determination of inter-Kingdom association and competition (bacterial and fungal) for adherence to virus-infected HeLa cell monolayers. This protocol can be extended to multiple combinations of prokaryotes, eukaryotes, and viruses.
Studiet af polymikrobielle interaktioner på tværs af de taksonomiske riger der omfatter svampe, bakterier og virus er ikke tidligere blevet undersøgt med hensyn til hvordan virale medlemmer af microbiome påvirke de efterfølgende mikrobe interaktioner med disse virusinficerede værtsceller. Den sameksistens af virus med bakterier og svampe er hovedsagelig til stede på slimhindeoverfladerne i mundhulen og genitale. Slimhindeceller, især dem med vedvarende kroniske eller vedvarende latente virusinfektioner, kunne have en betydelig indvirkning på medlemmer af microbiome gennem virus ændring i antallet og typen af receptorer udtrykt. Den humane microbiome omfatter forskellige organismer fra flere taksonomiske riger der deler geografiske regioner i kroppen. I værter med naturlige tænder, er mundslimhinden deles af HSV-1 og C. albicans, medens de forreste nasale Nares er besat af S. aureus 28. Trods in vitro resultater, at S. aureus overholder mund epitelceller, 29,30 S. aureus er sjældent isoleres fra orale præparater, når normalt væv er til stede 29,30. Der vides kun lidt om kønsorganer co-kolonisering nicher ud over de kliniske fund, at S. aureus er forbundet med aerob vaginitis, kendetegnet ved genital inflammation, udledning og dyspareuni, mens C. albicans producerer slimhindelæsioner lignende, der blev observeret i mundhulen 31-35.
