Here, we present a comprehensive capillary zone electrophoresis protocol for the assessment of intrinsic physicochemical heterogeneity of monoclonal antibodies as a quality attribute.
Biotherapeutic proteins, such as monoclonal antibodies (mAbs), are feasible alternatives for the treatment of chronic-degenerative diseases. The biological activity of these proteins depends on their physicochemical properties. The use of high-performance techniques like chromatography and capillary electrophoresis has been described for the analysis of physicochemical heterogeneity of mAbs. Nowadays, capillary zone electrophoresis (CZE) technique constitutes one of the most resolutive and sensitive assays for the analysis of biomolecules. Besides, the electro-driven separation in CZE is governed by extensive properties of matter and offers the advantage of analyzing proteins close to their native state. However, the successful implementation of this technique for routine analysis depends on the skills of the analyst at the critical steps during sample and system preparation. The purpose of this tutorial is to detail the steps to succeed in the CZE analysis of mAbs. Further, this protocol can be used for the development and improvement of skills of the personnel involved in protein analytical chemistry laboratories.
Monoklonale antistoffer (mAb'er) er bioterapeutiske proteiner med stigende interesse på grund af deres evne til at handle mod flere kroniske og degenerative sygdomme 1. Ligesom andre biomolekyler, mAb'er er tilbøjelige til at undergå flere fysisk-kemiske modifikationer på alle stadier af deres livscyklus (dvs. fra biosyntese til det endelige produkt). Sådanne modifikationer indbefatter, men er ikke begrænset til: deamidering, glycosylering, oxidation, cyklisering, isomerisering, aggregering og proteolytisk spaltning 2. Derfor er analyseteknikker stand til at løse iboende isoformer nødvendige for at overvåge mAb'er heterogenitet og stabilitet for at etablere kvalitetsspecifikationer.
Kapillarelektroforese (CE) er en højtydende separationsteknologi bæres outinside af en smal kvartsglas rør (um) fyldt med en baggrund elektrolyt (BGE). Ved påføring af et elektrisk felt (op til 30.000 V), ladet molekyles migrere til elektroden med modsat ladning (dvs. elektro-drevet separation). Anvendelsen af høje spændinger i CE tillader hurtige analyser og øget effektivitet, som er overlegen i forhold til klassiske gelelektroforese. Kapillær zone elektroforese (CZE) er et CE-baserede teknik rutinemæssigt anvendes i den biofarmaceutiske industri til vurdering produktkvalitet 3-9. I modsætning til andre former for CE (f.eks kapillær gelelektroforese, kapillær isoelektrisk fokusering) eller kromatografi-baserede metoder, kan CZE udføres uden anvendelse af denatureringsmidler eller fast-fase-grænseflader, hvilket muliggør analysen af den iboende heterogenitet mAb'er tæt på deres native tilstand 10 . CZE adskillelse af mAb-isoformer opstår inde i en kvartsglas kapillar dækket med en hydrofil polymer (neutral kapillær) og er baseret på deres forskellige elektroforetiske mobilitet, som er styret af ladning, masse, størrelse og form (eller hydrodynamisk volumen) 11. detekteres mAb-dele, nårde mobiliseres og passere gennem påvisning vinduet, som registreres af en ultraviolet (UV) absorbans detektor ved 214 nm 4.
Den vellykkede gennemførelse af denne analyseteknik vil afhænge af behørig opmærksomhed på detaljer før og under forsøget. Handler ellers vil øge omkostningerne og tid til at udføre analysen, i sidste ende fører til konstant fiasko og frustration.
Her præsenterer vi en trin-for-trin guide til at gennemføre en vellykket analyse af mAb heterogenitet ved CZE gennem detaljeret forklaring af fremstilling af opløsninger og prøver, udarbejdelse af CE-systemet, instrumentets metoder, der er op, datafangst, og forarbejdningen. Med henblik på denne gennemgang, et rekombinant fuldt humant anti-tumornekrosefaktor-alfa (anti-TNFa) mAb anvendes som protein model; dog kan denne protokol kan let tilpasses til analyse af andre proteiner overvejer korte modifikationer. ENdditionally, flere anbefalinger afbøde foreslås potentielle problemer. Læseren opfordres til nøje at følge den foreslåede protokol, som sandsynligheden for at lykkes, vil stige.
