En este método, presentamos procedimientos bioquímicos para el aislamiento rápido y eficiente de proteínas de filamento intermedio (IF) de múltiples tejidos de ratón. Se pueden usar IFs aisladas para estudiar los cambios en las modificaciones postraduccionales por espectrometría de masas y otros ensayos bioquímicos.
Los filamentos intermedios (IF), junto con filamentos de actina y microtúbulos, forman el citoesqueleto, un elemento estructural crítico de cada célula. El funcionamiento normal de las IF proporciona a las células resistencia mecánica y al estrés, mientras que un citoesqueleto disfuncional de IF compromete la salud celular y se ha asociado con muchas enfermedades humanas. Las modificaciones postraduccionales (PTMs) regulan críticamente la dinámica de IF en respuesta a cambios fisiológicos y bajo condiciones de estrés. Por lo tanto, la capacidad de supervisar los cambios en la firma PTM de IFs puede contribuir a una mejor comprensión funcional y, en última instancia, condicionamiento del sistema IF como un respondedor de estrés durante la lesión celular. Sin embargo, el gran número de proteínas IF, que están codificadas por más de 70 genes individuales y se expresan de una manera dependiente de los tejidos, es un reto importante para clasificar la importancia relativa de diferentes PTMs. Con este fin, los métodos que permiten el monitoreo de PTMs sobre las proteínas IFA nivel de todo el organismo, y no para miembros aislados de la familia, puede acelerar el progreso de la investigación en esta área. Aquí presentamos métodos bioquímicos para el aislamiento de la fracción total, soluble en detergente y resistente al detergente de las proteínas IF de 9 tejidos de ratón diferentes (cerebro, corazón, pulmón, hígado, intestino delgado, intestino grueso, páncreas, riñón y bazo). Demostramos además un protocolo optimizado para el aislamiento rápido de las proteínas IF utilizando matriz de lisis y homogeneización automatizada de diferentes tejidos de ratón. El protocolo automatizado es útil para perfilar IFs en experimentos con alto volumen de muestra (tal como en modelos de enfermedad que implican múltiples animales y grupos experimentales). Las muestras resultantes pueden utilizarse para diversos análisis de aguas abajo, incluyendo el perfilado de PTM basado en espectrometría de masas. Utilizando estos métodos, proporcionamos nuevos datos para demostrar que las proteínas IF en diferentes tejidos de ratón (cerebro e hígado) experimentan cambios paralelos con respecto a su expresiónY los PTM durante el envejecimiento.
Las IF son una familia de proteínas que en los seres humanos están codificadas por 73 genes y categorizadas en seis tipos principales: los tipos I a IV son citoplásmicos ( por ejemplo, queratinas epiteliales y capilares (K), mi- cito desmina, neurofilamentos, proteína glicolar fibrilar (GFAP) y otros); Tipo V son las láminas nucleares; Y el tipo VI son IFs en la lente ocular 1 . En cuanto a su organización molecular, las proteínas IF tienen tres dominios comunes: un dominio "varado" de bobina en espiral altamente conservado y dominios globulares de "cabeza" y "cola". Los tetrámeros de proteínas IF se unen para formar precursores de filamentos cortos, que finalmente se incorporan en filamentos maduros que forman estructuras citoesqueléticas y nucleoesqueléticas dinámicas involucradas en la protección mecánica 2 , la detección de estrés 3 , 4 , la regulación de la transcripción 5 y el crecimiento y otras funciones celulares críticas"> 1 , 6 , 7 .
La importancia funcional del sistema de IF se pone de relieve por la existencia de muchas enfermedades humanas causadas por mutaciones missense en genes de IF, incluyendo neuropatías, miopatías, trastornos de la fragilidad de la piel, disfunciones metabólicas y síndromes de envejecimiento prematuro 8 . Algunas mutaciones del gen IF no causan, pero predisponen a sus portadores a la progresión de la enfermedad, como las queratinas epiteliales simples en la enfermedad hepática 9 . Este último se debe a las funciones críticas de protección del estrés de las IF en los epitelios. Las IF en general se encuentran entre las proteínas celulares más abundantes en condiciones basales, pero se induce además fuertemente durante varios tipos de estrés 10 . Por ejemplo, estudios recientes que evalúan los cambios en todo el proteoma en el nematodo C. elegans demostraron que múltiples IFs están altamente reguladas y propensas a agregatiDurante el envejecimiento del organismo 11 , 12 . Dado que el mantenimiento de una estructura de IF adecuada es esencial para la resistencia celular a diversas formas de estrés 10 , la agregación de IF también puede contribuir a la disminución funcional durante el envejecimiento. Sin embargo, los estudios a nivel de organismal que examinan múltiples proteínas de mamífero IF a través de diferentes tejidos sometidos a estrés son deficientes.
Las IF son estructuras altamente dinámicas que se adaptan para satisfacer las demandas celulares. Las queratinas, por ejemplo, experimentan un ciclo independiente de biosíntesis entre el pool de proteínas solubles (no filamentosas) e insolubles (filamentosas) 13 . En condiciones fisiológicas normales, aproximadamente el 5% del total de K8 / K18 se puede extraer en tampón exento de detergente, en comparación con aproximadamente 20% que se puede solubilizar en el detergente no iónico Nonidet P-40, que es bioquímicamente comparable al Triton- X100 14 , </sArriba> 15 . Durante la mitosis hay un notable aumento en la solubilidad de los epitelios de tipo simple K8 y K18 14 , que es menos evidente en las queratinas epidérmicas, pero más evidente en la vimentina y otras proteínas tipo III IF [ 15 , 16] . Las propiedades de solubilidad de las proteínas IF están estrechamente reguladas por la fosforilación, una modificación clave post-traduccional (PTM) para el reordenamiento de los filamentos y la solubilidad 17 , 18 , 19 , 20 . La mayoría de las IF se someten a regulación extensiva por una serie de PTM en sitios conservados, lo que resulta en cambios funcionales [ 17] .
El propósito de este método es introducir a los investigadores que son nuevos en el campo de IF a la extracción bioquímica y métodos analíticos para el estudio de las proteínas de IF a través de múltiples tejidos de ratón. EspecíficoNos centramos en el aislamiento de las proteínas IF utilizando un método de extracción de alta sal y la evaluación de los cambios en PTMs a través de espectrometría de masas y por anticuerpos dirigidos a PTM. Estos métodos se basan en los procedimientos publicados anteriormente 21, pero incluyen modificaciones para extraer diferentes tipos de proteínas IF para descubrir el mecanismo común de regulación en toda la familia IF. Por ejemplo, la acetilación de K8 en un residuo de lisina específico regula la organización del filamento, mientras que la hiperacetilación promueve la insolubilidad de K8 y la formación de agregados 22 . Recientes estudios de perfiles proteómicos globales han revelado adicionalmente que la mayoría de las proteínas específicas de tejido IF son también objetivos para la acetilación y que la mayoría de los sitios de acetilación IF están confinados al dominio de barras altamente conservadas. Esto pone de manifiesto la necesidad de métodos adecuados para el perfil global del sistema IF. También se introduce un método rápido de aislamiento IF proteínas de múltiples tejidos utilizando homogeneización automatizada en optiMatriz de lisis. Las preparaciones resultantes son adecuadas para análisis PTM aguas abajo a través de espectrometría de masas y otros métodos.
Los métodos que permiten la caracterización bioquímica de las proteínas IF pueden ser útiles para entender numerosos fenómenos fisiopatológicos en los sistemas de mamíferos, ya que las proteínas IF son a la vez marcadores y moduladores del estrés celular y tisular 29 .
