In deze paper bieden wij een gedetailleerd protocol voor het blootstellen van de soort van het geslacht Drosophila aan verontreinigende stoffen, met het doel van het bestuderen van de impact van blootstelling over een scala van fenotypische resultaten op verschillende ontwikkelingsstadia en voor meer dan één generatie.
Emergente eigenschappen en externe factoren (populatieniveau en ecosysteem-niveau interacties, in het bijzonder) een belangrijke rol spelen in het bemiddelen van ecologisch belangrijke eindpunten, hoewel ze zijn zelden beschouwd als in het toxicologisch onderzoek. D. melanogaster ontpopt zich als een model van de toxicologie voor de gedrags, neurologische en genetische effecten van toxische stoffen, om enkelen te noemen. Wat nog belangrijker is, kunnen soort van het geslacht Drosophila worden gebruikt als een modelsysteem voor een integratieve kaderbenadering ecologisch relevante vragen in de toxicologie onderzoek te nemen emergente eigenschappen. Het doel van dit document is bedoeld als een protocol voor het blootstellen van de soort van het geslacht Drosophila aan verontreinigende stoffen moet worden gebruikt als een modelsysteem voor een scala aan fenotypische uitgangen en ecologisch relevante vragen. Meer in het bijzonder, dit protocol kan worden gebruikt om 1) koppeling van meerdere biologische niveaus van organisatie en begrijpen van de invloed van toxische stoffen op zowel individueel – en bevolking-level fitness; 2) test de invloed van toxische stoffen in verschillende stadia van ontwikkelingsstoornissen blootstelling; 3) test multigeneratie en evolutionaire implicaties van verontreinigende stoffen; en 4) test meerdere contaminanten en stressoren gelijktijdig.
Elk jaar worden ongeveer 1.000 nieuwe chemicaliën ingevoerd door de chemische industrie1,2; echter, de milieueffecten van slechts een klein percentage van deze chemische stoffen zijn getest voordat de distributie2,3. Hoewel grootschalige rampen zijn ongewoon, subletale and chronische blootstelling aan een grote verscheidenheid van verontreinigende stoffen zijn wijdverspreid in zowel mensen als dieren in het wild4,5. De historische focus van ecotoxicologie en milieutoxicologie moest testen letaliteit, enkele blootstelling aan chemische stoffen, acute blootstelling en de fysiologische effecten van blootstelling, als een middel van het meten van de impact van verontreinigende stoffen op overleving6, 7 , 8 , 9 , 10. Hoewel er een verschuiving naar ethische en niet-invasieve benaderingen voor dierproeven bevorderen, huidige benaderingen zijn beperking vanwege de rol die ontwikkeling, emergente eigenschappen en externe factoren (zoals populatieniveau en ecosysteem-niveau interacties) spelen in het bemiddelen van ecologisch belangrijke eindpunten8. Daarom is er behoefte aan een methoden die een meer holistische benadering te nemen zonder in te boeten, wild en/of gewervelde dieren in het laboratorium.
Ongewervelde modelsystemen, zoals Drosophila melanogaster, zijn een aantrekkelijk alternatief inspelen op de behoefte aan een meer holistische benadering van toxiciteit. D. melanogaster, werd oorspronkelijk ontwikkeld als een ongewervelde modelsysteem voor mens-gerelateerde genetisch onderzoek ongeveer een eeuw geleden11. D. melanogaster is nu prominent gebruikt als een alternatief gewervelde model om verschillende redenen: 1) de instandhouding van genen en trajecten tussen D. melanogaster en mensen; 2) korte generatietijd in vergelijking met gewervelde modellen; 3) goedkope kosten van onderhoud; 4) gemak bij het genereren van grote steekproeven; en 5) overvloed van fenotypische en ecologisch-relevante eindpunten beschikbaar voor het testen van11,12,13,14,15,16,17 .
Verschillende laboratoria11,15,16,17,18,19,20,21,22, 23 , 24 , 25 nu gebruikt D. melanogaster als een alternatief gewervelde model voor toxiciteitstests te begrijpen van de effecten van verontreiniging op de mens. Plaatselijke wilde soorten van Drosophila kan worden gebruikt, zo goed, als toxiciteit modellen voor wilde dieren (en mensen) te beantwoorden ecologisch-, gedragsgestoorde-, en evolutionair relevante vragen op meerdere biologische niveaus van de organisatie. Met behulp van soorten binnen het geslacht Drosophila als een model, zijn verschillende meetbare eindpunten mogelijk11,15,16,18,19,20 ,21,22,23,24,25. In addition, met behulp van het model van de Drosophila , toxicologen kunnen: 1) ethisch koppelen effecten op meerdere biologische niveaus van de organisatie; 2) nemen de rol van de opkomende factoren en ontwikkeling; 3) studie ecologisch belangrijke eindpunten (naast medisch belangrijke eindpunten); 4) test meerdere stressoren tegelijkertijd; 5) en test op lange termijn multigeneratie (bijvoorbeeld evolutionaire en transgenerationele) gevolgen van stressoren. Daarom kunt gebruik van Drosophila als een modelsysteem een veelheid aan benaderingen, niet beperkt tot het bestuderen van mechanistische benaderingen met ingeteelde stammen van D. melanogaster in het laboratorium.
In deze paper presenteren we de methoden voor het fokken en het verzamelen van Drosophila om verschillende toxicologische vragen te beantwoorden. Meer in het bijzonder, beschrijven we de methodologie voor 1) kippen Drosophila in medium doorweven met een of meer verontreinigende stoffen; 2) verzamelen Drosophila in de gehele ontwikkeling (bijvoorbeeld zwerven derde-instar-larven, pop gevallen, nieuw-eclosed volwassenen en volwassen volwassenen); en 3) Drosophila fokken in de besmette middellange tot test tussen de generaties en transgenerationele overdracht, alsmede de evolutionaire gevolgen van langdurige blootstelling van de toxische. Met behulp van dit protocol, de vorige auteurs18,19,20,21,22,23,24hebben,25 gemeld verschillende fysiologische, genetische en gedragsmatige gevolgen van ontwikkelingsstoornissen leiden (Pb2 +) blootstelling. Dit protocol maakt toxicologen te gebruiken van een meer holistische benadering van toxicologische, die essentieel is om te begrijpen hoe verontreinigende stoffen zijn risicofactoren voor zowel mens en dier in een steeds meer en meer vervuilde omgeving.
Drosophila melanogaster is opgezet als een krachtig model voor een scala aan biologische processen als gevolg van de uitgebreide instandhouding van genen en trajecten tussen D. melanogaster en mensen13,14. Om dezelfde redenen dat er een krachtig model voor medische wetenschap, heeft Drosophila ontpopt als een geschikt modelsysteem te bestuderen van de gevolgen van antropogene vervuiling voor tal van toxicologische eindpunten.
