Cette approche a été largement utilisée pour les tests de mutagénicité des cancérogènes dans les cellules de mammifères.
Un certain nombre de modèles animaux transgéniques et les systèmes de détection de mutation ont été développé pour les tests de mutagénicité des cancérogènes dans les cellules de mammifères. Parmi ceux-ci, les souris transgéniques et le Lambda (λ) Select cII système de détection de Mutation ont été employés pour des expériences de mutagénicité par de nombreux groupes de recherche dans le monde entier. Ici, les auteurs décrivent un protocole détaillé pour le test de mutation cII sélectionnez Lambda, qui peut être appliqué à des cellules de souris/rats transgéniques ou les animaux correspondants traitement avec un agent chimique/physique d’intérêt. Le protocole se compose des étapes suivantes : (1) isolement de l’ADN génomique des cellules ou des organes/tissus d’animaux transgéniques traitent in vitro ou in vivo, respectivement, avec un composé ; (2) récupération du vecteur navette lambda portant un gène de journaliste mutationnelle (c.-à-d., cII transgène) de l’ADN génomique ; (3) emballage des vecteurs sauvés dans les bactériophages infectieuses ; (4) infectant une bactérie hôte et mise en culture dans des conditions sélectives pour permettre la propagation des mutations induites cII ; et (5) marquant la cII-mutants et ADN de séquence analyse afin de déterminer la fréquence des mutants cII et spectre de mutation, respectivement.
Un large éventail de modèles animaux transgéniques et les systèmes de détection de mutation ont été développés pour les tests de mutagénicité des cancérogènes dans les cellules de mammifères. Parmi ceux-ci, des souris transgéniques de Big Blue (dénommé ci-après BB) et le λ Select cII système de détection de Mutation ont été employées pour des expériences de mutagénicité par ce groupe et de nombreuses autres recherches groupes dans le monde1,2, 3,4,5,6,7,8,9. Pour les 16 dernières années, nous avons étudié les effets mutagènes des divers agents physiques ou chimiques, à l’aide de ces animaux transgéniques ou traitement avec un composé leurs cultures de cellules de fibroblastes embryonnaires correspondant et par la suite analysé la phénotype et génotype du transgène cII par le λ Select cII dosage et le séquençage de l’ADN, respectivement10,11,12,13,14 , 15 , 16 , 17 , 18 , 19 , 20 , 21 , 22 , 23 , 24. le génome de ces animaux transgéniques contient un vecteur bactériophage λ navette (λLIZ) intégré sur le chromosome 4 une multi-copie tête-à-queue concatemer1,2,25. Le vecteur navette λLIZ porte deux gènes mutationnelle, à savoir la lacI et cII transgènes1,2,25,26,27, 28,29,30,31,32,33,34,35,36, 37,38,39,40,41,42,43,44,45 , 46 , 47. le λ Select cII est basé sur la récupération des vecteurs navettes λLIZ partir de l’ADN génomique des cellules provenant d’organes/tissus d’animaux transgéniques1,2,25 . Les vecteurs navettes λLIZ récupérés sont ensuite emballés dans têtes de phage λ capables d’infecter un hôte indicateur Escherichia coli. Par la suite, les bactéries infectées sont cultivés en sélectif permettant de notation et de l’analyse des mutations dans le cII transgène1,3.
Ici, les auteurs décrivent un protocole détaillé pour le dosage de Select cII λ, qui se compose de l’isolement de l’ADN génomique des cellules et des organes d’animaux transgéniques traités in vitro/in vivo avec un test de récupération composé, de le λLIZ navette vecteurs de l’ADN génomique, emballage des vecteurs dans des phages λ infectieuse, une infection de l’hôte Escherichia coli avec les bactériophages, identification de la cII-mutants dans des conditions sélectives afin de déterminer la fréquence des mutants cII , et Analyse de séquence d’ADN pour établir le spectre de mutation cII . Le protocole peut être appliqué aux souris/rat transgénique cell cultures traitées in vitro avec un agent chimique/physique des intérêts, ou tissus/organes des animaux correspondants traités in vivo avec le test agent1, 2,4,48,49,50,51,52. Le test de sélection cII λ est utilisé pour la détection des mutations dans le transgène cII récupéré de l’ADN génomique de cellules provenant d’organes/tissus de BB rongeurs3.
