Hier presenteren we een protocol te ontwikkelen en karakteriseren van tolerogenic dendritische cellen (TolDCs) en evalueren van hun immunotherapeutische nut.
Het immuunsysteem werkt door een strakke evenwicht tussen coördinerende reacties tegen buitenlandse antigenen en handhaven van een niet-reagerende staat tegen zelf-antigenen en antigenen afkomstig van commensale organismen. De verstoring van deze immuun homeostase kan leiden tot chronische ontsteking en voor de ontwikkeling van auto-immuniteit. Dendritische cellen (DC’s) zijn de professionele presentatie van antigeen cellen van het aangeboren immuunsysteem betrokken bij het activeren van de naïeve T cellen om te starten van de immuunrespons tegen vreemde antigenen. DCs kunnen echter ook worden onderscheiden in TolDCs die handelen te handhaven en T cel tolerantie te bevorderen en te onderdrukken effector cellen bij te dragen tot de ontwikkeling van beide voorwaarden van auto-immuunziekten of chronische ontsteking. De recente vooruitgang in ons begrip van TolDCs suggereert dat DC tolerantie kan worden bereikt door het moduleren van de voorwaarden van hun differentiatie. Dit verschijnsel heeft geleid tot een enorme groei in TolDC therapieën voor talrijke immuun aandoeningen veroorzaakt door te breken in immuun tolerantie te ontwikkelen. Succesvolle studies in preklinische autoimmuniteit lymfkliertest modellen hebben verder de immunotherapeutische nut van TolDCs in de behandeling van auto-immune aandoeningen gevalideerd. TolDCs hebben tegenwoordig een veelbelovende immunotherapeutische hulpmiddel in de kliniek voor herstel van immuun tolerantie in verschillende immuun aandoeningen door pathogene auto-immune reacties zich te richten terwijl protectieve immuniteit intact blijven. Hoewel een array van strategieën heeft voorgesteld door meerdere labs voor het opwekken van TolDCs, is er geen consistentie in het karakteriseren van de cellulaire en functionele fenotype van deze cellen. Dit protocol biedt een stapsgewijze handleiding voor de ontwikkeling van beenmerg-afgeleide DCs in grote aantallen, een unieke methode die wordt gebruikt om hen te onderscheiden in TolDCs met een synthetische triterpenoid 2-cyano-3,12-dioxooleana-1,9-dien-28-oic zuur-difluoro-propyl-amide (CDDO-DFPA), en de technieken gebruikt om te bevestigen hun fenotype, met inbegrip van analyses van essentiële moleculaire signaturen van TolDCs. Tot slot laten we zien een methode om te beoordelen van de TolDC functie door het testen van hun immunosuppressieve reactie in vitro en in vivo in een preklinische model van multiple sclerose.
Dendritische cellen (DC’s) werden zijn een integraal onderdeel van het aangeboren immuunsysteem en eerst ontdekt en gekenmerkt door Ralph Steinman en Zanvil Cohn in 1973 als primaire professionele antigeen presentatie1 cellen. DCs is aangetoond dat een belangrijke rol spelen in de immuunactivatie door de presentatie van verwerkte antigeen aan T-cellen en B cellen via grote comptabiliteit complexen (MHC) in secundaire lymfoïde organen te koppelen van de aangeboren en adaptieve immuunsysteem2. In de zoogdieren immuunsysteem zijn er ten minste twee categorieën van DCs die worden omschreven als myeloïde DCs en plasmacytoid DCs (PDC’s)3. Myeloïde DCs, ook bekend als conventionele DCs (cDCs), worden gekenmerkt door de uitdrukking van CD11c en kan worden gedifferentieerd als onvolwassen DCs (iDCs) in vitro van voorlopercellen van het beenmerg of perifere bloed monocyten gebruiken granulocyt-macrofaag-kolonie-stimulerende factor (GM-CSF) en IL-4 bij lymfkliertest of menselijke diersoorten, respectievelijk4.
Activeren ‘gevaar’ signalen, zoals pathogen-geassocieerde moleculaire patronen (PAMP) of schade-geassocieerde moleculaire patronen (VOCHTIGE), rijdt iDCs rijping richting immunogene DCs als volwassen DCs (mDCs) via verschillende patroon erkenning receptoren bindt de DC-oppervlakte5. Immunogene DCs verder prime naïef T-celproliferatie en differentiatie via opregulatie van MHCII2, costimulatory liganden (CD80, CD86 en interactie CD40)6, cytokines en andere oplosbare bemiddelaars7. Een cascade van pro-inflammatoire mediator productie van immunogene DCs is essentieel voor de cytokine gemedieerde T celdifferentiatie. Bijvoorbeeld, zowel IFN-γ en IL-12 zijn nodig voor de Th1 differentiatie8 en IL-1, IL-6, en IL-23 zijn kritisch voor de naïeve T cel polarisatie naar Th17 cellen9. Hoewel volwassen DCs op vreemde antigenen reageren, kan ongecontroleerde DC activering door zelf-antigenen leiden tot tolerantie ablatie en bevordering van de ontwikkeling van auto-immune ziekten door het genereren van autoreactieve T cellen waarvan Activering tot weefselvernietiging10 leidt .
Recente rapporten hebben verstrekt duidelijk bewijs van DC plasticiteit, wordt geïllustreerd door hun vermogen om te communiceren met verschillende aanwijzingen binnen hun communicatie weefsel en te onderscheiden in verschillende effector/suppressorgenen DC subsets. De anti-inflammatoire mediatoren, zoals IL-10,11, TGF-β12en HO-113 hebben aangetoond dat een belangrijke rol spelen bij immuun onderdrukking door inducerende tolerogenic DCs (TolDCs). Deze TolDCs verwerven van de regelgevende taken en onderdrukken van T-cel proliferatie14. Bovendien, het gebrek aan co-stimulatie door DCs en de productie van anti-inflammatoire mediatoren van TolDCs zowel bijdragen tot de inductie van regulatoire T-cellen (Tregs) en ook effectief remmen zowel Th1 en Th17 differentiatie en uitbreiding15. In afgelopen twee decennia, is de therapeutische mogelijkheden van TolDCs gemeld door verschillende onderzoekers. In deze studies, het beheer van ex-vivo TolDCs niet alleen verbeterd pathologische symptomen gegenereerd in verschillende preklinische modellen van auto-immune ziekten16 , maar ook geleid tot de ontwikkeling van immune tolerantie bij patiënten17 ,18. Interessant is dat vandaag de TolDCs therapie heeft beschouwd als een alternatieve of adjuvante aanpak voor auto-immune ziekten in verschillende klinische proeven, met inbegrip van type 1 diabetes mellitus19, reumatoïde artritis20, 21, multiple sclerose (MS)22,23,24, en de ziekte van Crohn ziekte25.
Er zijn een verscheidenheid van protocollen die hebben gewerkt aan het ontwikkelen van TolDCs en verschillende laboratoria methoden hebben gemeld voor generatie en fenotypische karakterisering van TolDCs. Deze methoden kunnen worden gebruikt voor het genereren van reproducibly TolDCs in vitro van hematopoietische progenitorcellen en stabiel om ze te bewaren in een tolerogenic staat in vivo26,27,28,29. De iDCs kan worden omgezet in TolDCs door blootstelling aan verschillende immunomodulerende farmacologische agenten of anti-inflammatoire cytokines. Vitamine D3 is bijvoorbeeld een bekende farmacologische agent bekend te vergroten van de productie van de IL-10 en onderdrukken van IL-12 secretie van DCs en te stimuleren waardoor hun immunosuppressieve functie30. Bovendien, wanneer DCs worden blootgesteld aan krachtige inflammatoire stimuli, zoals lipopolysacchariden (LPS), verschillende farmacologische stoffen zoals dexamethason31, rapamycin32en33 van de corticosteroïden is gebleken voor het opwekken van de TolDC fenotype doordat DC oppervlakte expressie van CD40, CD80, CD86 en MHCII34. IL-10 en TGF-β zijn de meest bestudeerde anti-inflammatoire cytokines voor het opwekken van DC tolerantie35 en de daarmee gepaard gaande blootstelling aan beide van deze cytokines voor het opwekken van het fenotype van een tolerogenic in DCs36is gebleken.
Sinds de tolerogenic die DC wordt bepaald door functionele eigenschappen in plaats van door fenotypische markers, er is behoefte een grote een consistente methode voor cellulaire en functionele karakterisering van TolDCs te ontwikkelen. Bovendien een strenge en consequente protocol moet worden vastgesteld voor de consistente beoordeling en de karakterisering van het tolerogenic DC fenotype als wij gaan effectief en reproducibly vergelijken het vermogen van de nieuwe agenten voor het opwekken van het fenotype van de TolDC in de laboratorium. Hier voorzien wij een gedetailleerd protocol met stapsgewijze methoden beschreven om te isoleren iDCs van hematopoietische progenitorcellen van muizen en vervolgens analyseren van de werkzaamheid van nieuwe agenten onder evaluatie voor hun capaciteit iDCs omzetten in TolDCs, die bieden een robuuste functionele en fenotypische karakterisatie van TolDCs zowel in vitro als in vivo. Deze beschrijving bevat een uitgebreide methode karakteriseren de TolDCs door hun oppervlakte liganden, cytokine profiel en immunosuppressieve functies in vitro. We bieden ook een voorbeeld van een methode om te verkennen van de mogelijke therapeutische toepassing van deze TolDCs in een preklinische model van MS, experimentele autoimmune encefalomyelitis (EAE). Deze gangbaar protocol helpt onderzoekers om de capaciteit van de nieuwe agenten ter bevordering van de inductie van TolDCs en vergemakkelijkt de inspanning uit te breiden het bereik van TolDC therapeutische ontwikkeling.
Dit witboek beschrijft een efficiënt protocol dat kan worden gebruikt voor reproducibly voor het genereren van iDCs en vervolgens om hen te onderscheiden in TolDCs, en wij stellen voor dat dit kan worden toegepast om te evalueren van de capaciteit van de nieuwe moleculaire doel agenten voor het opwekken van de TolDC fenotype.
