Mostramos que uma implantação de microeletrodos no córtex motor de ratos provoca déficits motor imediatos e duradouros. Os métodos propuseram neste documento contorno uma cirurgia de implante de microeletrodos e três roedores tarefas comportamentais para elucidar possíveis alterações na função motor fina ou bruta devido aos danos causados a implantação para o córtex motor.
Dispositivos médicos implantados no cérebro têm enorme potencial. Como parte de um sistema de Interface de máquina cerebral (IMC), intracortical microeletrodos demonstram a capacidade de gravar os potenciais de ação de individuais ou pequenos grupos de neurônios. Tais sinais gravados com êxito tem sido usados para permitir pacientes para interface com ou controlar seus próprios membros, membros robóticos e computadores. No entanto, estudos anteriores com animais mostraram que uma implantação de microeletrodos no cérebro não só danifica o tecido circundante, mas também pode resultar em déficits funcionais. Aqui, vamos discutir uma série de testes comportamentais para quantificar potenciais deficiências motor seguindo a implantação de microeletrodos intracortical no córtex motor de um rato. Os métodos para a grade de campo aberto, cruzamento de escada e testes de resistência de aderência fornecem informações valiosas sobre as potenciais complicações resultantes de uma implantação de microeletrodos. Os resultados dos testes comportamentais estão correlacionados com histologia de ponto de extremidade, fornecendo informações adicionais sobre os resultados patológicos e os impactos deste procedimento no tecido adjacente.
Microeletrodos intracortical foram originalmente usados para mapear os circuitos do cérebro e desenvolveram-se em uma valiosa ferramenta para habilitar a detecção de intenções motor que pode ser usado para produzir saídas funcional1. Saídas funcionais detectadas podem oferecer a indivíduos que sofrem de lesões na medula espinhal, paralisia cerebral, esclerose lateral amiotrófica (ela) ou outras condições de limitação de movimento do controle de um cursor de computador2,3 ou robótica 4,5,6do braço, ou restaurar a função própria de membro com deficiência7. Portanto, tecnologia de microeletrodos intracortical tem emergido como um promissor e rapidamente crescente campo8.
Devido o sucesso visto no campo, estudos clínicos estão em andamento para melhorar e entender melhor as possibilidades de BMI tecnologia5,9,10. Por perceber o potencial de comunicação com os neurônios no cérebro, as aplicações de reabilitação são percebidas como ilimitado8. Embora haja grande optimismo para o futuro da tecnologia de microeletrodos intracortical, também é bem conhecido que microeletrodos eventualmente falharem11, possivelmente devido a uma resposta aguda MPTP pós-implante. A implantação de um material estranho no cérebro resulta em danos imediatos para os tecidos circundantes e leva a mais danos causados por MPTP resposta que varia de acordo com propriedades do implante12. Além disso, um implante no cérebro pode causar um efeito de microlesion: uma redução no metabolismo da glicose, pensado para ser causado por edema agudo e hemorragia devido a inserção de dispositivo13. Além disso, a qualidade de sinal e o comprimento de tempo que os sinais úteis podem ser registrados são inconsistentes, independentemente do modelo animal11,14,15,16. Vários estudos têm demonstrado a conexão entre neuroinflammation e microeletrodos desempenho17,18,19. Portanto, o consenso da Comunidade é que a resposta inflamatória do tecido neural que rodeia os microeletrodos, pelo menos em parte, compromete a confiabilidade do eletrodo.
Muitos estudos têm examinado a inflamação local11,20,21,22 ou explorado métodos para reduzir os danos ao cérebro causado por inserção11,23, 24,25, com o objetivo de melhorar o desempenho de gravação ao longo do tempo14,26. Além disso, nós recentemente mostraram que uma lesão iatrogênica causada por uma inserção de microeletrodos no córtex motor de ratos provoca uma imediata e duradoura déficit motor bem27. Portanto, o objetivo dos protocolos aqui apresentados é dar pesquisadores um método quantitativo para avaliar possíveis déficits motor como resultado de trauma crânio-encefálico após a implantação e a presença persistente de dispositivos intracortical (microeletrodos na caso deste manuscrito). Os testes de comportamento descritos aqui foram projetados para destrinchar ambas as deficiências da motricidade grosseira e fina e podem ser usados em muitos modelos de lesão cerebral. Esses métodos são simples, reprodutível e podem ser facilmente implementados em um modelo de roedor. Além disso, os métodos aqui apresentados permitem uma correlação de comportamento motor para resultados histológicos, um benefício que, até recentemente, os autores não vi publicada no campo de IMC. Finalmente, como esses métodos foram projetados para testar a função motora fina28, a função motora bruta29e stress e ansiedade comportamento29,30, os métodos apresentados aqui também podem ser implementados em uma variedade de modelos de ferimento na cabeça, onde os investigadores querem governar para fora (ou em) eventuais déficits de função motora.
O protocolo descrito aqui tem sido usado para efetivamente e reproducibly medir déficit motor fino e bruto em um modelo de lesão cerebral de roedores.
