Wir haben gezeigt, dass eine Mikroelektrode Implantation im motorischen Kortex von Ratten sofortige und dauerhafte motorische Defizite verursacht. Die Methoden vorgeschlagen hierin Umriss einer Mikroelektrode Implantation Operation und drei Nagetier Verhaltens Aufgaben, mögliche Änderungen in die feine oder grobe Motorik durch Implantation verursacht Schäden an den motorischen Kortex aufzuklären.
Medizinprodukte, die in das Gehirn implantiert halten enormes Potenzial. Als Teil eines Systems von Gehirn-Maschine-Schnittstelle (BMI) zeigen intracortical Mikroelektroden Aktionspotentiale von einzelnen oder kleinen Gruppen von Neuronen aufnehmen. Diese aufgezeichneten Signale sind erfolgreich benutzt worden, Patienten mit zulassen oder Computer, Roboter-Gliedmaßen und ihre eigenen Gliedmaßen zu kontrollieren. Jedoch haben frühere Tierstudien gezeigt, dass eine Mikroelektrode Implantation im Gehirn nicht nur das umliegende Gewebe schädigt, sondern auch funktionelle Defizite führen kann. Hier diskutieren wir eine Reihe von Verhaltensstörungen Tests, mögliche motorische Beeinträchtigungen nach der Implantation von intracortical Mikroelektroden in den motorischen Kortex einer Ratte zu quantifizieren. Die Methoden für die Freiland-Raster, Leiter Kreuzung und Griff Stärke testen liefern wertvolle Informationen über die möglichen Komplikationen infolge einer Mikroelektrode Implantation. Die Ergebnisse des Verhaltens Tests korrelieren mit Endpunkt Histologie, Bereitstellung zusätzlicher Informationen über die pathologische Ergebnisse und Wirkungen dieses Verfahrens auf das angrenzende Gewebe.
Intracortical Mikroelektroden dienten ursprünglich ordnen Sie die Schaltkreise des Gehirns, und entwickelten sich zu einem wertvollen Werkzeug ermöglicht die Erkennung von motor Absichten derer funktionellen Ausgänge1produzieren. Erkannten funktionale Ausgänge können Personen leiden Verletzungen des Rückenmarks, Zerebralparese, Amyotrophe Lateralsklerose (ALS) oder andere Bewegung einschränkenden Bedingungen zur Kontrolle von einem Computer Cursor2,3 oder Roboter anbieten. Arm4,5,6, oder Funktion auf ihre eigenen behinderten Extremität7wiederherstellen. Daher hat eine vielversprechende und schnell wachsenden Feld8intracortical Mikroelektrode Technologie entwickelt.
Aufgrund der Erfolge im Bereich gesehen sind klinische Studien im Gange, zu verbessern und besser zu verstehen, die Möglichkeiten des BMI Technologie5,9,10. Erkennen das volle Potenzial der Kommunikation mit den Neuronen im Gehirn, sind die Reha-Anwendungen als grenzenlose8wahrgenommen. Zwar gibt es viel Optimismus für die Zukunft der intracortical Mikroelektrode Technologie, ist es auch bekannt, dass Mikroelektroden schließlich11, möglicherweise durch eine akute schwere Reaktion nach der Implantation nicht. Die Implantation von Fremdmaterial im Gehirn führt unmittelbare Schädigung des umliegenden Gewebes und führt zu weiteren Schäden, die durch die schwere Reaktion, die variiert je nach Eigenschaften des Implantats12. Darüber hinaus kann ein Implantat im Gehirn verursachen einen Microlesion-Effekt: ein Rückgang der Glukose-Stoffwechsel vermutlich durch akute Ödeme und Blutungen durch das Gerät einstecken13verursacht werden. Darüber hinaus sind die Signalqualität und die Länge der Zeit, die nützliche Signale aufgezeichnet werden können inkonsistente, unabhängig von der Tier-Modell11,14,15,16. Mehrere Studien belegen den Zusammenhang zwischen Neuroinflammation und Mikroelektrode Leistung17,18,19. Daher ist der Konsens der Gemeinschaft, dass die entzündliche Reaktion des neuronalen Gewebes, die die Mikroelektroden umgibt zumindest teilweise, Elektrode Zuverlässigkeit Kompromisse.
Viele Studien haben untersucht lokale Entzündung11,20,21,22 oder erforscht Methoden zur Schadensbegrenzung im Gehirn verursacht durch Einfügung11,23, 24,25, mit dem Ziel der Verbesserung der Aufnahme über Zeit14,26. Darüber hinaus haben wir vor kurzem gezeigt, dass eine iatrogene Schädigung durch eine Mikroelektrode Einfügung im motorischen Kortex von Ratten führt zu einer sofortigen und dauerhaften feinen motorischen Defizit27. Daher der hier vorgestellten Protokolle soll Forschern eine quantitative Methode zur Bewertung möglich motorische Defizite durch Hirn-Trauma nach der Implantation und anhaltende Präsenz intracortical Geräte geben (Mikroelektroden in der im Falle dieser Handschrift). Die hier beschriebene Verhalten-Tests wurden entwickelt, um sowohl grob- und motorischen Beeinträchtigungen herauskitzeln und können in vielen Modellen der Hirnverletzung verwendet werden. Diese Methoden sind einfach, reproduzierbar und können leicht in einem Nagetier Modell umgesetzt werden. Des weiteren können hier vorgestellten Methoden für eine Korrelation von motorischen Verhalten zu histologischen Ergebnisse, ein Vorteil, den bis vor kurzem die Autoren nicht gesehen haben im BMI-Bereich veröffentlicht. Endlich, wie diese Methoden entwickelt wurden, um feine Motorik28, die Grobmotorik29und Stress und Angst Verhalten29,30, Testen der hier vorgestellten Methoden können auch umgesetzt werden in ein Modellauswahl Kopfverletzung wo wollen die Forscher (oder in) herrschen motorischen Defizite.
Die hier beschriebene Protokoll wurde zur fein- und Grobmotorik motorischen Defizit in einem Modell der Nagetier-Hirn-Trauma effektiv und reproduzierbar zu messen. Darüber hinaus ermöglicht es die Korrelation von feinen motorischen Verhalten zur histologischen Ergebnisse nach einer Mikroelektrode Implantation im motorischen Kortex.
