Effekten av migrastatic hemmere på glioma kreftcelle migrasjon i tredimensjonale (3D) invasjon analyser ved hjelp av en Histone deacetylase (HDCA) inhibitor er preget av høyoppløselig konfokalmikroskopi mikroskopi.
Drug oppdagelse og utvikling i kreftforskning er i økende grad basert på narkotika skjermer i et 3D-format. Romanen hemmere rettet mot trekk og invasiv potensialet for kreftceller, og følgelig metastatisk spredning av sykdom, blir oppdaget og betraktes som komplementære behandlinger i svært invasiv kreftformer som gliomas. Dermed er det nødvendig å generere data som muliggjør detaljert analyse av celler i et 3D-miljø etter tilsetning av et medikament. Metodikken beskrevet her, kombinerer spheroid invasjon analyser med høyoppløselig bilde fangst og dataanalyse av konfokalmikroskopi laserskanning mikroskopi (CLSM), aktivert detaljert karakterisering av virkningene av den potensielle anti-trekk-hemmer MI-192 på glioma celler. Spheroids ble generert fra cellelinjer for invasjon analyser i lav tilhenger 96-brønn plater og deretter forberedt for CLSM analyse. Den beskrevne arbeidsflyten ble foretrukket fremfor andre brukte spheroid teknikker på grunn av både enkel og reproduserbarhet. Dette, kombinert med forbedret bildeoppløsning oppnådd ved konfokalmikroskopi mikroskopi sammenlignet med konvensjonelle bredt felt tilnærminger, tillot identifisering og analyse av distinkte morfologiske endringer i trekk celler i et 3D-miljø etter behandling med migrastatic stoffet MI-192.
Tredimensjonale spheroid teknologier for prekliniske narkotika oppdagelse og utvikling av potensielle kreft stoffer blir i økende grad favorisert over konvensjonelle narkotika skjermer; Dermed er det mer utvikling av migrastatic-migrasjon og invasjon forebygge-narkotika. Begrunnelsen bak denne utviklingen i kreft behandling er klar: 3D spheroid analyser representerer en mer realistisk tilnærming for screening potensielle anti-kreft narkotika som de etterligner 3D svulst arkitektur mer trofast enn celle monolag kulturer, recapitulate legemiddel-tumor interaksjoner (Kinetics) mer nøyaktig, og tillate karakterisering av narkotika aktivitet i en svulst-relaterte innstillingen. I tillegg, fremveksten av resistens mot chemotoxic narkotika i mange krefttyper og høy dødelighet blant kreftpasienter på grunn av metastasering forsterket av evnen til kreftceller til å migrere til fjerne tumor nettsteder støtter inkludering av kjemoterapeutiske agenter målretting av trekk potensialet av kreftceller som adjuvant behandling i fremtidig klinisk kreft forsøk1.
