Collectieve Celmigratie in ontwikkeling, wondgenezing en kanker metastasen wordt vaak geleid door de gradiënten van groeifactoren of signalering van moleculen. Hier beschreven is een experimenteel systeem dat tractie microscopie combineert met een microfluïdisch systeem en een demonstratie van het kwantificeren van de mechanica van collectieve migratie onder biochemische gradiënt.
Cellen veranderen migratiepatronen in reactie op chemische stimuli, met inbegrip van de hellingen van de stimuli. Cellulaire migratie in de richting van een chemische gradiënt, bekend als chemotaxis, speelt een belangrijke rol in de ontwikkeling, de immuunrespons, wondgenezing en kanker metastasen. Terwijl chemotaxis de migratie van enkelvoudige cellen en verzamelingen van cellen in vivo moduleert, is in vitro onderzoek gericht op eencellige chemotaxis, deels als gevolg van het ontbreken van de juiste experimentele gereedschappen. Om die kloof te dichten, hier beschreven, is een uniek experimenteel systeem dat microfluïdica en micropatterning combineert om de effecten van chemische gradiënten op collectieve Celmigratie aan te tonen. Bovendien worden tractie microscopie en monolaag stress microscopie in het systeem opgenomen om veranderingen in cellulaire kracht op het substraat en tussen naburige cellen te karakteriseren. Als proof-of-concept, de migratie van micro patroon circulaire eilanden van Madin-Darby Canine nier (MDCK) cellen wordt getest onder een gradiënt van hepatocyte groeifactor (HGF), een bekende verstrooiing factor. Het is gevonden dat cellen in de buurt van de hogere concentratie van HGF sneller migreren dan die aan de andere kant binnen een celeiland. Binnen hetzelfde eiland, cellulaire tractie is vergelijkbaar aan beide zijden, maar intercellulaire stress is veel lager aan de kant van hogere HGF concentratie. Dit nieuw experimentele systeem kan nieuwe mogelijkheden bieden om de mechanica van Chemotactische migratie door cellulaire collectieven te bestuderen.
Cellulaire migratie in biologische systemen is een fundamenteel fenomeen betrokken bij weefselvorming, de immuunrespons, en wondgenezing1,2,3. Cellulaire migratie is ook een belangrijk proces in sommige ziekten zoals kanker4. Cellen migreren vaak als een groep in plaats van afzonderlijk, die bekend staat als collectieve cel migratie4,5. Voor cellen om collectief te bewegen, is de detectie van de micro-omgeving essentieel6. Cellen waarnemen bijvoorbeeld fysisch-chemische stimuli en reageren door veranderende beweeglijkheid, celsubstraat interacties en celcelinteracties, resulterend in directionele migratie langs een chemische gradiënt7,8, 9,10. Op basis van deze verbinding zijn er snelle ontwikkelingen gemaakt in lab-on-a-chip-technologieën die goed gecontroleerde chemische micro omgevingen kunnen creëren, zoals de gradiënt van een chemoaantretant11,12,13 . Hoewel deze Lab-op-een-chip-gebaseerde microfluïdica eerder zijn gebruikt om chemotaxis van het cellulaire Ensemble of cellulaire sferoïden14,15,16,17te bestuderen, zijn meestal gebruikt in de context van eencellige migratie18,19,20,21. Mechanismen die aan een cellulaire collectieve respons op een chemische gradiënt ten grondslag liggen, zijn nog steeds niet goed begrepen14,22,23,24,25,26 . Zo zal de ontwikkeling van een platform dat de spatiotemporele controle van oplosbare factoren mogelijk maakt, evenals in situ observatie van cellen biofysische, helpen om de mechanismen achter collectieve Celmigratie te ontrafelen.
Ontwikkeld en beschreven hier is een multi-channeled microfluïdisch systeem waarmee het genereren van een concentratie gradiënt van oplosbare factoren die de migratie van patroon celclusters moduleert. In deze studie wordt hepatocyte growth factor (HGF) gekozen om het migratie gedrag van Madin-Darby Canine nier (MDCK) cellen te reguleren. Hgf staat erom bekend dat de celcelintegriteit wordt verzwakt en de beweeglijkheid van cellen27,28wordt verbeterd. In het microfluïdische systeem worden ook Fourier-transformatie tractie microscopie en monolaag stress microscopie verwerkt, waardoor de beweeglijkheid, de contracactielkracht en de intercellulaire spanning geïnduceerd door samenstellende cellen in reactie op een hgf Verloop. Binnen hetzelfde eiland migreren cellen in de buurt van de hogere concentratie van HGF sneller en vertonen lagere intercellulaire stress niveaus dan die op de zijkant met lagere HGF-concentratie. De resultaten suggereren dat dit nieuwe experimentele systeem geschikt is om andere vragen te onderzoeken op gebieden met betrekking tot collectieve cellulaire migratie onder chemische gradiënten van verschillende oplosbare factoren.
Collectieve migratie van samenstellende cellen is een belangrijk proces tijdens de ontwikkeling en regeneratie, en de migratie richting wordt vaak geleid door de chemische gradiënt van groeifactoren4,23.
