Migração de células coletivas em desenvolvimento, cicatrização de feridas e metástase oncológica é muitas vezes guiada pelos gradientes de fatores de crescimento ou moléculas de sinalização. Descrito aqui é um sistema experimental que combina microscopia de tração com um sistema microfluídico e uma demonstração de como quantificar a mecânica da migração coletiva gradiente bioquímico.
As células alteram os padrões de migração em resposta a estímulos químicos, incluindo os gradientes dos estímulos. A migração celular na direção de um gradiente químico, conhecida como quimiotaxia, desempenha um papel importante no desenvolvimento, na resposta imune, na cicatrização de feridas e na metástase do câncer. Enquanto a quimiotaxia modula a migração de células simples, bem como coleções de células in vivo, in vitro pesquisa centra-se em quimiotaxis de uma única célula, em parte devido à falta de ferramentas experimentais adequadas. Para preencher essa lacuna, descrita aqui é um sistema experimental único que combina microfluídica e micropadronização para demonstrar os efeitos de gradientes químicos na migração de células coletivas. Além disso, a microscopia de tração e a microscopia de estresse monocamada são incorporadas ao sistema para caracterizar mudanças na força celular no substrato, bem como entre as células vizinhas. Como prova de conceito, a migração de ilhas circulares micromodeladas de células de rim canino Madin-Darby (MDCK) é testada um gradiente de fator de crescimento de hepatócitos (HGF), um fator de espalhamento conhecido. Encontra-se que as pilhas situadas perto da concentração mais elevada de HGF migram mais rapidamente do que aquelas no lado oposto dentro de uma ilha da pilha. Dentro da mesma ilha, a tração celular é semelhante em ambos os lados, mas o estresse intercelular é muito menor no lado da maior concentração de HGF. Este novo sistema experimental pode proporcionar novas oportunidades para estudar a mecânica da migração quimotáxica por coletivas celulares.
A migração celular em sistemas biológicos é um fenômeno fundamental envolvido na formação de tecidos, na resposta imune e nacicatrização de feridas1,2,3. A migração celular também é um processo importante em algumas doenças como o câncer4. As células geralmente migram como um grupo em vez de individualmente, o que é conhecido como migração de célula coletiva4,5. Para que as células se movam coletivamente, a detecção do microambiente é essencial6. Por exemplo, as células percebem estímulos físico-químicos e respondem alterando a motilidade, interações entre células e células, resultando em migração direcional aolongo de umgradiente químico7,8, 9,10. Baseado nesta conexão, os avanços rápidos foram feitos nas tecnologias Lab-on-a-chip que podem criar microambientes químicos bem controlados tais como o inclinação de um chemoattractant11,12,13 . Quando estes microfluídica Lab-em-um-chip-baseados tiverem sido usados previamente para estudar quimiotaxia do Ensemble celular ou dos esferoides celulares14,15,16,17, têm sido utilizados principalmente no contexto da migração de uma única célula18,19,20,21. Os mecanismos subjacentes a uma resposta coletiva celular a um gradiente químico ainda não são bem compreendidos14,22,23,24,25,26 . Assim, o desenvolvimento de uma plataforma que possibilite o controle espaciotemporal dos fatores solúveis, bem como a observação in situ das células biofísicas ajudarão a desvendar os mecanismos por trás da migração de células coletivas.
Desenvolvido e descrito aqui é um sistema microfluídico multicanalizado que possibilita a geração de um gradiente de concentração de fatores solúveis que modulam a migração de aglomerados de células padronizadas. Neste estudo, o fator de crescimento de hepatócitos (HGF) é escolhido para regular o comportamento migratório das células do rim canino de Madin-Darby (MDCK). HGF é conhecido por atenuar a integridade celular e aumentar a motilidade das células27,28. No sistema microfluídico, a microscopia de tração da transformada de Fourier e a microscopia de tensão monocamada também são incorporadas, o que permite a análise da motilidade, da força contrátil e da tensão intercelular induzida pelas células constituintes em resposta a um HGF Gradiente. Dentro da mesma ilha, as células localizadas perto da maior concentração de HGF migram mais rápido e apresentam níveis de estresse intercelulares inferiores aos do lado com menor concentração de HGF. Os resultados sugerem que este novo sistema experimental é adequado para explorar outras questões em áreas envolvendo migração celular coletiva gradientes químicos de vários fatores solúveis.
