La migración celular colectiva en el desarrollo, la cicatrización de heridas y la metástasis del cáncer a menudo se guía por los gradientes de los factores de crecimiento o moléculas de señalización. Aquí se describe un sistema experimental que combina la microscopía de tracción con un sistema microfluídico y una demostración de cómo cuantificar la mecánica de la migración colectiva bajo gradiente bioquímico.
Las células cambian los patrones de migración en respuesta a los estímulos químicos, incluidos los gradientes de los estímulos. La migración celular en la dirección de un gradiente químico, conocido como quimiotaxis, desempeña un papel importante en el desarrollo, la respuesta inmune, la cicatrización de heridas y la metástasis del cáncer. Mientras que la quimiotaxis modula la migración de células individuales, así como las colecciones de células in vivo, la investigación in vitro se centra en la quimiotaxis de una sola célula, en parte debido a la falta de las herramientas experimentales adecuadas. Para llenar ese vacío, descrito aquí es un sistema experimental único que combina microfluídicos y micropatrones para demostrar los efectos de los gradientes químicos en la migración de células colectivas. Además, la microscopía de tracción y la microscopía de tensión monocapa se incorporan al sistema para caracterizar los cambios en la fuerza celular en el sustrato, así como entre las células vecinas. Como prueba de concepto, la migración de islas circulares micropatrónizadas de células renales caninas de Madin-Darby (MDCK) se prueba bajo un gradiente de factor de crecimiento de hepatocitos (HGF), un factor de dispersión conocido. Se encuentra que las células ubicadas cerca de la mayor concentración de HGF migran más rápido que las del lado opuesto dentro de una isla celular. Dentro de la misma isla, la tracción celular es similar en ambos lados, pero el estrés intercelular es mucho menor en el lado de una mayor concentración de HGF. Este novedoso sistema experimental puede proporcionar nuevas oportunidades para estudiar la mecánica de la migración quimiotáctica por colectivos celulares.
La migración celular en sistemas biológicos es un fenómeno fundamental implicado en la formación de tejidos, la respuesta inmune y la cicatrización de heridas1,2,3. La migración celular también es un proceso importante en algunas enfermedades como el cáncer4. Las celdas a menudo se migran como un grupo en lugar de individualmente, lo que se conoce como migración de celdas colectivas4,5. Para que las células se muevan colectivamente, la notorencia del microambiente es esencial6. Por ejemplo, las células perciben estímulos fisicoquímicos y responden cambiando la motilidad, las interacciones célula-sustrato e interacciones célula-célula, lo que resulta en una migración direccional a lo largo de un gradiente químico7,8, 9,10. Sobre la base de esta conexión, se han realizado rápidos avances en tecnologías de laboratorio en chip que pueden crear microambientes químicos bien controlados, como el gradiente de un quimioatractor11,12,13 . Mientras que estos microfluídicos basados en laboratorio en un chip se han utilizado previamente para estudiar quimiotaxis del conjunto celular o esferoides celulares14,15,16,17, se han utilizado principalmente en el contexto de la migración de una sola célula18,19,20,21. Mecanismos subyacentes a una respuesta colectiva celular a un gradiente químico todavía no se entiende bien14,22,23,24,25,26 . Así, el desarrollo de una plataforma que permita el control espaciotemporal de factores solubles, así como la observación in situ de la biofísica de las células, ayudará a desentrañar los mecanismos detrás de la migración celular colectiva.
Desarrollado y descrito aquí es un sistema microfluídico multicanal que permite la generación de un gradiente de concentración de factores solubles que modula la migración de clústeres de células con patrones. En este estudio, el factor de crecimiento de hepatocitos (HGF) se elige para regular el comportamiento migratorio de las células del riñón canino de Madin-Darby (MDCK). HGF es conocido por atenuar la integridad celular y mejorar la motilidad de las células27,28. En el sistema microfluídico, también se incorporan la microscopía de tracción de transformación de Fourier y la microscopía de tensión monocapa, que permite el análisis de la motilidad, la fuerza contráctil y la tensión intercelular inducida por las células constituyentes en respuesta a un HGF Gradiente. Dentro de la misma isla, las células ubicadas cerca de la mayor concentración de HGF migran más rápido y muestran niveles de estrés intercelular más bajos que aquellos en el lado con menor concentración de HGF. Los resultados sugieren que este nuevo sistema experimental es adecuado para explorar otras cuestiones en campos que implican la migración celular colectiva bajo gradientes químicos de diversos factores solubles.
