Een verfijnde methode van weefselopruiming werd ontwikkeld en toegepast op het volwassen muizenhart. Deze methode is ontworpen om dicht, autofluorescent hartweefsel te zuiveren, met behoud van gelabelde fibroblastfluorescentie toegeschreven aan een genetische reporterstrategie.
Hart- en vaatziekten zijn wereldwijd de meest voorkomende oorzaak van mortaliteit en worden vaak gekenmerkt door verhoogde hartfibrose die kan leiden tot verhoogde ventriculaire stijfheid met veranderde hartfunctie. Deze toename van cardiale ventriculaire fibrose is te wijten aan activering van ingezeten fibroblasten, hoewel hoe deze cellen werken binnen het 3-dimensionale (3D) hart, bij aanvang of na activering, niet goed wordt begrepen. Om te onderzoeken hoe fibroblasten bijdragen aan hartaandoeningen en hun dynamiek in het 3D-hart, is een verfijnde op CLARITY gebaseerde weefselophelderings- en beeldvormingsmethode ontwikkeld die fluorescerend gelabelde hartfibroblasten in het hele muishart laat zien. Weefsel ingezeten fibroblasten werden genetisch gelabeld met behulp van Rosa26-loxP-eGFP florescent reporter muizen gekruist met de cardiale fibroblast uitdrukken Tcf21-MerCreMer knock-in lijn. Deze techniek werd gebruikt om fibroblastlokalisatiedynamiek in de hele volwassen linkerventrikel te observeren bij gezonde muizen en bij fibrotische muismodellen van hartaandoeningen. Interessant is dat in één verwondingsmodel unieke patronen van hartfibroblasten werden waargenomen in het gewonde muizenhart dat banden van verpakte vezels in de contractiele richting volgde. In ischemische letselmodellen trad fibroblastdood op, gevolgd door herbevolking uit het grensgebied van het infarct. Gezamenlijk maakt deze verfijnde hartweefselverhelderingstechniek en gedigitaliseerd beeldvormingssysteem 3D-visualisatie van hartfibrose in het hart mogelijk zonder de beperkingen van antilichaampenetratiefalen of eerdere problemen rond verloren fluorescentie als gevolg van weefselverwerking.
Hoewel cardiomyocyten de grootste volumefractie in het hart vormen, zijn hartfibroblasten overvloediger en zijn ze kritisch betrokken bij het reguleren van de structurele en herstellende basiskenmerken van dit orgaan. Cardiale fibroblasten zijn zeer mobiel, mechanisch responsief en fenotypisch variërend afhankelijk van de mate van activering. Cardiale fibroblasten zijn nodig om normale niveaus van extracellulaire matrix (ECM) te handhaven, en te weinig of te veel ECM-productie door deze cellen kan leiden tot ziekte1,2,3. Gezien hun belang bij ziekten zijn hartfibroses een steeds belangrijker onderwerp van onderzoek geworden om nieuwe behandelingsstrategieën te identificeren, vooral in een poging om overmatige fibrose te beperken4,5,6,7. Bij letsel activeren en differentiëren fibroblasten in een meer synthetisch celtype dat bekend staat als een myofibroblast, die proliferatie kan zijn en overvloedig ECM kan afscheiden, evenals contractiele activiteit heeft die helpt bij het remodelleren van de ventrikels.
Terwijl hartfibroblasten uitgebreid zijn geëvalueerd op hun eigenschappen in 2D-culturen6,8,9,10, wordt veel minder begrepen van hun eigenschappen en dynamiek in het 3D-levende hart, hetzij bij aanvang of met ziektestimulatie. Hier is een verfijnde methode beschreven om het volwassen muishart te zuiveren met behoud van de fluorescentie van fibroblasten gelabeld met een Rosa26-loxP-eGFP x Tcf21-MerCreMer genetisch verslaggeversysteem. In het hart is Tcf21 een relatief specifieke marker van rustgevende fibroblasten4. Nadat tamoxifen is gegeven om het induceerbare MerCreMer-eiwit te activeren, zullen in wezen alle rustgevende fibroblasten permanent versterkt groen fluorescerend eiwit (eGFP) uitdrukken van de Rosa26-locus, waardoor ze in vivo kunnen worden gevolgd.
Er bestaan talrijke gevestigde weefselophelderingsprotocollen, waarvan sommige zijn toegepast op het hart11,12,13,14,15,16,17. Veel van de reagentia die in verschillende weefselontruimingsprotocollen worden gebruikt, blijken echter endogene fluorescentiesignalen te onderdrukken18. Bovendien is het volwassen hart moeilijk te wissen vanwege overvloedige heemgroephoudende eiwitten die autofluorescentie genereren19. Daarom was het doel van dit protocol om fibroblastmarkerfluorescentie te behouden met de gelijktijdige remming van heemautofluorescentie in het gewonde volwassen hart voor optimale 3D-visualisatie in vivo12,13,14,16,17,20.
Eerdere studies die probeerden de cardiale fibroblast in vivo te onderzoeken, gebruikten geperfuseerde antilichamen om deze cellen te labelen, hoewel dergelijke studies beperkt waren door antilichaampenetratie en cardiale vasculaire structuur14,16,17,20. Hoewel Salamon et al. weefselontruiming hebben aangetoond met behoud van topische neuronale fluorescentie in het neonatale hart, en Nehrhoff et al. hebben aangetoond dat fluorescentie myeloïde cellen markeert, is het behoud van endogene fluorescentie door de gehele ventriculaire wand nog niet aangetoond, inclusief de visualisatie van volwassen cardiale fibroblasten bij aanvang of na letsel13,20. Dit weefselontruimingsprotocol verfijnt een mengsel van eerdere protocollen op basis van de CLARITY-methode (clear lipid-exchanged acrylamide-hybridized rigid Imaging/immunostaining/in situ-hybridization-compatible tissue hydrogel) en PEGASOS (polyethyleenglycol (PEG)-geassocieerd oplosmiddelsysteem). Dit verfijnde protocol maakte een robuuster onderzoek van hartfibrose in het muizenhart bij aanvang mogelijk en van hoe ze reageren op verschillende soorten letsel. Het protocol is eenvoudig en reproduceerbaar en zal helpen het gedrag van hartfibrose in vivo te karakteriseren.
Dit artikel presenteert een verfijnde methode voor weefselopruiming die visualisatie van hartfibroblasten in vivo mogelijk maakt, zowel bij aanvang als na letsel, om fibroblasten in het muizenhart te karakteriseren en beter te begrijpen. Dit verbeterde protocol pakt beperkingen aan in bestaande weefselontruimingsprotocollen die hebben geprobeerd specifieke celtypen in het volwassen of neonatale hart te identificeren12,13,14…
